Expression of ABO blood-group antigenic determinants identified by monoclonal antibodies B7H4 and D1E8 in normal human tissues and organs.

Human blood group antigens are genetically determined structures of undefined nature found not only in red blood cells but also in various tissues. Monoclonal antibodies against A and B human blood group antigens (B7H4 and D1E8) were obtained. Their tissue compatibility was tested in 11 different normal human organs taken from people with A1 and B blood group phenotypes. The antigen-antibody reaction was visualized using the indirect immunoperoxidase technique. The results of the study showed that the antibodies B7H4 and D1E8 react specifically with the tissue blood group antigens in healthy subjects. The findings obtained in most of the tested organs were in accord with those described by other researchers. Differences were found only in the stomach, urothelium, the lungs and the thyroid gland. A possible explanation of the results is the difference in the epitopes recognized by the monoclonal antibodies used by us and the blood group antibodies applied by other researchers. It is suggested that the monoclonal antibodies against the human blood group antigens A and B obtained by us could be used to establish suspected modification of ABH-antigens in the process of oncogenesis.

[Immunohistochemical evidence of vimentin in fibrocystic breast disease and mammary carcinoma].

Immunohistochemical investigations were conducted of cryostat sections taken from 25 cases of fibrocystic breast disease and 100 cases of mammary carcinoma (among them 15 cases with lymph node metastases) to obtain information on expression of vimentin. Immunoreactive epithelial cells were recordable from 12 cases of fibrocystic disease (48%) and 32 tumors (32%). In the majority of all cases with vimentin-expressing carcinoma, positive staining was exhibited by less than 10% of the neoplastic cells. No correlation was found to exist between histological tumor grade, lymph node status and vimentin expression.

Breast Neoplasms

Carcinosarcoma of the breast. Immunohistochemical and ultrastructural studies.

A case of a carcinosarcoma in the breast of a 51-year-old woman is described. The tumor consisted of two diversely differentiated components with cells possessing epithelial and mesenchymal characteristics. Plenty of cytokeratin and CEA were immunohistochemically recordable from the carcinomatous component. Some neoplastic cells also stained positively for actin, S-100 protein and smooth muscle antigen. The sarcomatous component was positive only for vimentin. Electron microscopic analysis revealed presence of the following cell types: epithelial cells, myoepithelial cells and polymorphous mesenchymal cells.

Morphological characteristics of the endocrine pancreas in alloxan diabetes after cyclosporin A administration.

Histological, immunocytochemical, morphometric and electron microscope studies were carried out on the pancreas of alloxan diabetic rats pretreated with cyclosporin A. High mortality, severe destruction of pancreatic B-cells and presence of sporadic mononuclear infiltrations in islets and around excretory ducts were observed. The results obtained show that cyclosporin A potentiates the toxic effect of alloxan on the pancreatic B-cells.

[Lymphoid cell infiltration into the pancreas of C57BL/6J mice].

Peri-insular lymphoid infiltrations were observed in nondiabetic C57BL/6J mice. The type of infiltrating cells was determined by direct immunoperoxidase technique. A larger part of lymphoid cells did not show reaction with a goat antimouse IgG Rh conjugate. Intensive hyperplastic reaction and the presence of many immunoglobulin producing cells were observed in the lymph nodes adjoining the pancreas. Probably the observed infiltrations in the pancreas of nondiabetic C57BL + 6J mice were composed mainly of T lymphocytes, belonging to T helper subpopulation.

[Effect of laser irradiation on the serological and morphological indices of experimental immune reactivity (a preliminary report)].

The authors examined guinea pigs, irradiated for a period of six days with helium-neon lazer according to a scheme, proposed by them. They determined the changes in the complement serum activity, blood level of heparine and histamine as well as histologic changes in adrenals, liver and spleen. When they compared the control animals (nonirradiated) with the experimental animals, they established a statisticaly significant elevation of the complement activity in serum, a tendency to lowering of histamine with an increase of heparine and hyperplastic reaction in zona glomerulosa of the adrenals. The obtained results suggested stimulation of immune mechanisms with antiallergic and antinflammatory effect.

[Effects of reduced diet and physical stress on liver lysosomes. Morphological and biochemical studies].

Groups of 15 rats each (I--non-reduced diet; II--non-reduced diet and physical stress; III--diet reduced by 40%; IV--reduced diet and physical stress) were examined after 20 and 140 days, respectively. The electron microscopic examination revealed that the most important changes had occurred in group III; there exists an excess of autophagolysosomes. The biochemical examinations show that the reduced diet (group III) also produces a considerable increase in activity of most of the lysosome enzymes in the liver. The occurrence of autophagy evidences that this is either physiologically induced or the manifestation of a beginning pathological process.

Sterol composition of Bulgarian soya and corn oils.

The free sterols, the sterol esters and the sterol glycosides of the raw soya and corn oils as well as those of the technical lecithin and the deodorizer distillated of the latter oils were isolated by preparative TLC. The composition of each of the isolated sterol derivatives was determined by GLC and MS. Sitosterol, campesterol, stigmasterol and an unknown sterol with a molecular weight of 428 are contained in almost all of the examined fractions of the soya oil and its refinement byproducts. Dehydrocampesterol is present in the free sterols of the raw soya oil and the soya lecithin. Stigmasterol is contained in the soya deodorizer distillate in high amounts. It was established that cholesterol was present in the sterol esters of the raw soya oil high amounts. Delta7-stigmastenol occurs only in the sterol esters of the latter oil. Sitosterol, campesterol and stimgasterol are the main components of all sterol fractions of the corn oil and its refinement products. Dehydrocampesterol and unknown sterols with molecular weights of 428 are present in the free sterols of the raw corn oil. Some sterol glycosides of the soya and corn lecithin are esterified with the same major fatty acid components of the glycerides, palmitic acid being the main one. The fatty acid compositon of sterol esters of the raw soya and corn oil roughly corresponds to the fatty acid composition of oils.

[Morphology of the spleen and immune response in experimental nutritional deficiency in rats].

The author examined 60 white rats of the strain "Wistar" with initial age of four months and body weight of 180--220 gm, subjecter to nutritive deficit regimens with various protein content. He described the morphological changes in the spleen, made a qualitative analysis of IgG and IgM in serum and examined also some links of the immune response after antigen stimulation with human gamma-globulin. Very limited nutrition (5,74 of protein) for 16 weeks caused immune deficit with definite morphological substrate-atrophy and fibrosis of the lymphoid tissue. Moderate limited feeding (11, 48% of protein) did not induce substantial deviations from the functions the immune system.

[Structuro-functional parallels in several liver lesions in rats resulting from dietary protein deficiency].

The authors examined 60 white rats of the Wistar strain with initial age of four monts and weight of 200 -- 15 gm, subjected to deficient food regime with varous protein content (5.74%, 11.48%, but the control was 17.50% of protein) for a period of 16 weeks. After this period half of the animals were stimulated by an antigen -- 10% of human gamma globulin. They determined total protein and protein fractions in serum, histological changes in liver and some morphometric indices. It was established that there was a correlation between the level of serum proteins and the observed changes in liver parenchyma and mesenchyma, in which the factor "protein deficit" had significantly greater effect than the factor "heterologous serum protein" as an antigenic stimulus.

[Morphological changes in the organs of rats subjected to limited feeding and physical loading].

Changes in the liver, stomach, small intestine, kidneys, lungs, myocardium, aorta, spleen and mesenterial lymph nodes are studied histologically in 120 male white rats from the Wistar line, subjected to a deficient alimentation regime, with and without physical exertion. The observed slight degree of adiposogenital dystrophy of the liver during free feeding regime indicates that the latter is equivalent to overfeeding. Variations from the normal histological picture in some organs (liver, myocardium, stomach and small intestine) are detected in the groups undergoing rather prolonged, moderate undernourishment--a sign of the sensitivity of these organs to the limited supply of nutritive ingredients. Physical loading, combined with free alimentary regimen, exert a beneficial effect on the organism which is suggested by the morphological data disclosing a slightly enhanced function of the myocardium and kidneys in these animals.

[Lysosomes, cellular digestion and immune processes].

The article presents a survey of the available literature concerned with the problems of relationships between the functions of the lysosomal apparatus of cells of the reticuloendothelial system and immune processes. A connection between the primary lysosoma and endocytic vacuole, desintegration of absorbed antigens by micro- and macrophages, the significance of antigenic information transmitted by macrophage to an immune lymphocyte, and realization of the immune response are traced carefully and in succession. Participation of lysosomas in antigen-antibody reactions (anaphylaxis, Arthus' phenomenon, supersensitivity of a delayed type) and their role in induction of immune tolerance and in some autoimmune processes are also considered.

[Free and bound sterol content of sunflower, soy bean and maize oils].

The amounts of free sterols, steryl esters and steryl glycosides in sunflower, soya and maize oils as well as in the phosphatides of the same oils were determined. The refinement byproducts were enriched with sterols. The highest contents of total sterols and steryl glycosides (2,68 and 1,9%, respectively) were established in soya phosphatides followed by those in sunflower oil phosphatides (2,03 and 1,68%, respectively) and maize oil phosphatides (0,81 and 0,54%, respectively).
