Semi-micro quantitative gel precipitin reaction.

Quantitative gel precipitin reactions were made on a semi-micro-scale. This required overall reduction of the dimensions of the necessary apparatus. Resulting from this was the ability to use agarose gel at low concentration not possible with the conventional macro quantitative gel precipitin technique. Further advantages were the reduction of time for the completion of the experiments and also that of the volume of the reactants. Diffusion coefficients of antigens determined by the semi-micro technique agreed with those obtained with the more accurate optical methods.

Isolation of a cytoplasmic polyhedrosis virus by physical and immunological techniques.

Procedures are described for the separation and purification of cytoplasmic polyhedrosis viruses obtained from multiply infected Heliothis armigera larvae. Separation was achieved by differential centrifugation and density gradient zone electrophoresis followed by complexing with nuclear polyhedrosis virus specific antibody. The yield of cytoplasmic polyhedrosis virus was increased by passage in larvae reared on synthetic media.

Purification of filamentous plant viruses by thin layer centrifugation.

The construction of a modified thin layer ultracentrifuge rotor is described. This rotor was used in the purification of five filamentous plant viruses, viz. TMV, SCMV, PVX, SGV and YMC. The purification and concentration of these viruses in their monomeric forms is hazardous when conventional "tube" rotors are used since they invariably result in dissociation and aggregation of the virus particles. Using the thin layer rotor these infective agents may be concentrated in volumes of fluid equal to approximately 1% of the starting suspension and not as pellets obtained after ultracentrifugation is conventional "tube" rotors. Electron microscopy revealed that the virus particles concentrated by thin layer centrifugation were not aggregated and that only few fragments of the virus filaments were present in the final preparations.

Polyethylene glycol purification of influenza virus with respect to aggregation and antigenicity.

Influenza virus may be precipitated and aggregated by polyethylene glycol into clusters comprising ten to many hundred virions per aggregate. These aggregates are sparingly soluble and may be freed of contaminating polymer by washing in the appropriate buffer at room temperature or by precipitation in 30% (v/v) ethanol at subzero temperatures.Immunogenic studies in guinea-pigs of the virus in different states of dispersion revealed that the aggregated virus is the superior antigen to the virus in the monomeric form or in the dissociated state following treatment with ether.
