Cryopreservation of human fetal adrenal medullary cells.

Cryopreservation of human fetal adrenomedullary cells was performed, after dissociation of the gland and purification of chromaffin aggregates, using 10% dymethyl sulfoxide as cryoprotectant. More than 90% of thawed cultured clusters appear morphologically intact, showing extensive neurite outgrowth and absence of appreciable cellular loss. Immunoreactivity for tyrosine hydroxylase and response to nerve growth factor were demonstrated. These findings indicate that human medullary cells maintain their distinctive functional characteristics after cryopreservation.

Primary cultures of human caudate nucleus.

It is possible to grow functional primary dissociated cultures and explants from stereotactic biopsies of human parkinsonian caudate nuclei. Two major classes of cells were identified on morphological grounds. The culture cells appear to be stimulated by an unidentified soluble factor(s) obtained from human fetal neuronal cells in vitro. Culture of primary neuronal and glial cells from human adult cerebral nuclei seems to be a useful tool for several research purposes and in particular for studying both trophic factor action and target effects on afferent neurons for prospective human brain grafting.

[Conduction disorders and sites of calcification of the mitral ring: 2-dimensional echocardiographic study].

The association between a calcified mitral annulus and impairment of cardiac impulse transmission is well known. However informations on the relations between the site of mitral calcification and conduction disturbance are still scanty. Twenty-nine patients (22 women and 7 men, mean age 64.4 +/- 13.1 years) with M-mode and two-dimensional echocardiographic evidence of mitral annulus calcifications were studied. The "annulus" was subdivided in 4 segments: antero-medial, antero-lateral, postero-medial and postero-lateral. Conduction disturbances were present in 15 patients (51.7%). Atrio-ventricular or intra-ventricular conduction defects were found in 12 out of 15 patients with calcifications of the antero-medial segment and only in 3 of 14 subjects without antero-medial calcifications (P less than .01). The presence of conduction defects therefore seems to be significantly increased when calcium deposition is located near the conduction system.

[Prostaglandins and peptic ulcer disease].

The action of prostaglandins with regard to ulcers is made clear by their inhibition of gastric secretion, on the one hand, and their so-called cytoprotective effect, on the other. Experimental data are used in a summary of the explanations offered of the mechanisms underlying these two effects. In addition, attentions is directed to the possible rôle of the prostaglandins in gastric physiology, and hence in the physiopathology of ulcers. The therapeutic results obtained with prostaglandins in the management of gastroduodenal ulcer offer firm grounds for supposing that further developments will take place in the clinical field.

Maintained cellular function of adrenal medullary cells in parkinsonian dysautonomia.

Adrenal gland involvement in Parkinson's disease was reported by different authors. Further studies became relevant after adrenal was proposed as dopaminergic donor for neurotransplantation. Chromaffin cells were grown in culture and the effects of nerve growth factor (NGF) tested: no differences were observed between parkinsonian and control cells. The expression of the beta-NGF mRNA in the parkinsonian adrenal was analyzed: a specific cDNA was synthesized and a 168 bp portion amplified using PCR. The products were identified and the identity of the fragment was confirmed by sequencing. Quantitative PCR demonstrated a beta-NGF mRNA concentration exceeding 5 fg/micrograms of total adrenal RNA. These findings demonstrate the retained functional capacity of the parkinsonian adrenal to respond to NGF and express the beta-NGF mRNA.

[Effect of prostaglandins on the cardiovascular system].

Ever since the effect of prostaglandins on vasal tone and cardiac function was discovered, an impressive series of studies has been conducted on their relations with the cardiovascular system. These studies have been extended to the prostaglandin system compounds discovered in more recent times. Of these, particular importance is being attached to prostacycline, though to be implicated in the physiological control of platelet clumping and also in that of renin secretion. Further investigation of the part played by prostacycline and other prostaglandins in cardiovascular disease has given unusually promising interim results.

[Evaluation of a registration card for logging electrocardiographic records into standard personal computers].

BACKGROUND: The MS200 Cardioscope, from MRT Micro as., Norway, is a 12 channel ECG card to be directly inserted into a standard personal computer (PC). The standard ISA Bus compatible half length card comes with a set of 10 cables with electrodes and the software for recording, displaying and saving ECG signals. The system is supplied with DOS or Windows software. The goal of the present work was to evaluate the affordability and usability of the MS200 in a clinical setting. MATERIALS AND METHODS: We tested the 1.5 DOS version of the software. In 30 patients with various cardiac diseases the ECG signal has been recorded with MS200 and with standard Hellige CardioSmart equipment. The saved ECGs were recalled and printed using an Epson Stylus 800 ink-jet printer. Two cardiologists reviewed the recordings for a looking at output quality, amplitude and speed precision, artifacts, etc. RESULTS: 1) Installation: the card has proven to be totally compatible with the hardware; no changes in default settings had to be made. 2) Usage: the screens are clear; the commands and menus are intuitive and easy to use. Due to the boot-strap and software loading procedures and, most important, off-line printing, the time needed to obtain a complete ECG printout has been longer than that of the reference machine. 3) Archiving and retrieval of ECG: the ECG curves can be saved in original or compressed form: selecting the latter, the noise and non-ECG information is filtered away and the space consumption on disk is reduced: on average, 20 Kb are needed for 10 seconds of signal. The MS200 can be run on a Local Area Network and is prepared for integrating with an existing informative system: we are currently testing the system in this scenery. 4) MS200 includes options for on-line diagnosis, a technology we have not tested in the present work. 5) The only setting allowed for printing full pages is letter size (A4): the quality of printouts is good, with a resolution of 180 DPI. CONCLUSIONS: In conclusion, the MS200 system seems reliable and safe. In the configuration we tested, it cannot substitute a dedicated ECG equipment: from this point of view, a smaller PCMCIA-type card with a battery-operated notebook PC will be more suitable for clinical uses. Nevertheless, the possibility to log and track ECG records, integrated into the department informative system, may provide a valuable tool for improving access to medical information.
