Experimental simulation of the environment of the delta opioid receptor. A 500 MHz study of enkephalins in CDCl3.

Complexes of [Met5] and [Leu5]enkephalin amides with 18-crown-6-ether have been studied in CDCl3 solution by means of 500 MHz NMR spectroscopy, in order to simulate two of the features of the opioid receptor: the apolar environment and the binding of the charged N atom. Contrary to all previous studies in polar solvents the NH resonances are spread in a huge range (ca. 4 ppm) as in the spectra of rigid cyclic peptides. The two observed intramolecular hydrogen bonds are consistent with the existence of a single, folded, conformation, i.e. a C10 beta-turn in which the Phe4 NH is linked to the Tyr1 CO group.

Auditory brain-stem response and hearing threshold in cerebellopontine angle tumours.

The auditory brain-stem responses (ABR) in 19 patients with confirmed cerebellopontine angle tumours are evaluated in relation to the hearing threshold as obtained by pure-tone audiometry. ABR pathological alterations, in terms of either absence or presence of components, appear to relate to the suprathreshold stimulus intensity and to match the changes which can be observed in subjects with normal hearing, when stimulated at the equivalent suprathreshold intensity. Of the diagnostic indexes considered (I-V interval, IT5, delta V), IT5 and delta V could be evaluated in the largest patient population. They may be used in combination to enhance their sensitivity in retrocochlear lesion detection.

Circulating immune complexes in infected ventriculoatrial and ventriculoperitoneal shunts.

Distinguishing infected from noninfected ventriculoatrial (VA) or ventriculoperitoneal (VP) shunts is prognostically and therapeutically important. Eighty-seven serum samples from twenty-seven patients with VA or VP shunts were studied for the presence and quantification of circulating immune complexes. Eighty-three percent of the samples from infected shunts presented circulating immune complexes. Mean values of immune complexes in patients with infected shunts were significantly higher than in those without infection. In febrile, septicemic patients with few clinical symptoms, immune complexes were present, and their measurement in serial serum samples was a significant diagnostic aid. If circulating immune complex levels are not detectable, a shunt infection is less likely to be present.

Monocyte Fc-IgG receptors expression and soluble suppressor factor in skin squamous cell carcinoma.

The expression of Fc-IgG receptors on mononuclear phagocytes (monocytes/macrophages), as well as the activity of soluble immune suppressor supernatant of T-cell proliferation (SISS-T) factor have been investigated in 23 patients with squamous cell carcinoma (SCC) of the lower lip, including 8 patients with metastases in the regional lymph glands. The results demonstrated a significant increase of the expression of Fc-IgG receptors only on circulating monocytes of patients with metastatic SCC. The suppressor function evaluated by means of the SISS-T factor proved normal in patients with nonmetastatic skin SCC and reduced in 8 patients with metastatic SCC.

Characterization of the circulating immune complexes in acute and chronic liver diseases.

The composition of immune complexes (IC) found in 28 patients with HBsAg positive and negative acute and chronic hepatitis was analysed. Components were defined in PEG-precipitated material isolated by preparative ultracentrifugation on linear sucrose density gradients and analysed by the Ouchterlony plate technique and by the radioimmunoassay of hepatitis B virus markers. Sedimentation rate ranged from 8 to 19s in the serum of patients with chronic hepatitis, whereas heavier IC (greater than 19s) were present in the active phase of the disease. The participation of hepatitis B virus in acute hepatitis was shown by the presence of its antigens. In contrast, a low incidence of vital components was seen in IC of chronic active hepatitis and liver cirrhosis. Thus, other causes must contribute to the formation of IC in chronic liver disease.

[Significance of immunocomplexes and complement in renal transplantation. Preliminary data].

The authors have measured circulating immune complexes (ic) and some components (C1q, C4, C3 and properdin factor B) in 87 serum samples obtained during the follow-up of five renal transplantation. Results showed high serum levels of ic with normal renal function of the graft especially in one patient, and an increase of the values in some rejection crises. The behaviour of the complement system was variable for the different components, while frequently low properdin factor B serum levels were noticed in some renal transplantations during the normal function of the graft and during the rejection crisis. In conclusion, the Authors report that a) high levels of ic can be seen also in grafts with normal function and their presence may be correlated with viral infections; b) an increase of ic during the rejection crisis is due to humoral immune response; c) the reduction of C3 and C1q associated to an increase of ic has been frequently seen, and finally; d) the low levels of properdin factor B show a probable intervention of the alternative complement pathway in the rejection crisis.
