Control of cytosolic calcium activity during low sodium exposure in cultured chick heart cells.

We investigated the roles of sodium-calcium exchange, sarcoplasmic reticulum, and mitochondria in Cai homeostasis in cultured chick ventricular cells. Specifically, the influence of low sodium medium on contractile state, calcium fluxes, and cytosolic free [Ca] [( Ca]i) was examined. [Ca]i was measured using fura-2. Mean [Ca]i in control medium was 126 +/- 14 nM. Exposure of cells to sodium-free or sodium- and calcium-free medium (choline-substituted) resulted in contracture development, which returned toward the baseline level over 2-3 minutes. The Nao-free contracture was associated with a tenfold increase in [Ca]i (1,280 +/- 110 nM) followed by a gradual decrease to a level fourfold above control [Ca]i (460 +/- 58 nM). Nao- and Cao-free contracture was associated with a fivefold increase in [Ca]i (540 +/- 52 nM) followed by a rapid decrease to below 80 nM. Sodium-free medium failed to produce an increase in [Ca]i or contracture in cells preexposed to calcium-free medium, although caffeine, when subsequently added to sodium- and calcium-free medium, was able to elicit a transient increase in [Ca]i and contracture. Brief, 5-second preperfusion of cells with La3+ (1 mM) or EGTA (1 mM) abolished the Nao-free contracture and the increase in [Ca]i. In the presence of 20 mM caffeine, removal of Nao resulted in minimal changes in the resting position of the cell although 45Ca uptake and [Ca]i were increased in response to sodium-free medium; the subsequent decrease in [Ca]i was greatly slowed. Addition of caffeine during the relaxation phase of the sodium-free contracture produced an additional transient contracture and transient increase in [Ca]i. Ryanodine (1 microM) abolished this effect of caffeine. Caffeine or ryanodine abolished Nao- and Ca-free contracture. CCCP (2 microM), a potent oxidative phosphorylation inhibitor, did not significantly affect calcium efflux rate. In the presence of 2 microM CCCP, removal of sodium resulted in an augmented contracture signal and a rise in [Ca]i, followed by a slow decrease. We conclude that removal of extracellular sodium enhances transsarcolemmal entry of calcium via sodium-calcium exchange, but this effect alone does not lead to the development of sodium-free contracture. Calcium displaceable by lanthanum or EGTA appears to contribute to Nao-free or Nao- and Cao-free contracture. Studies using caffeine and ryanodine suggest that removal of Nao leads to release of calcium from the sarcoplasmic reticulum (presumably via calcium-induced calcium release).(ABSTRACT TRUNCATED AT 400 WORDS)

Usefulness of blood parameters, especially viscosity, for the diagnosis and elucidation of pathogenic mechanisms of the hand-arm vibration syndrome.

In the present study it was found that, in vibrating-tool operators with Raynaud's phenomenon, whole blood viscosity was significantly higher than in operators without Raynaud's phenomenon at shear rates from 230 to 11.5 s-1. In addition rats were experimentally exposed to local vibration (60 Hz, 5 g) on their hind limbs for 4 h/d for 30 or 90 d. In the case of 30-d exposure, the small arteries in the exposed site did not change. However, after exposure for 90 d, disruption of the internal elastic lamina was observed in the small arteries. The disruption was followed by focal cell proliferation with regenerative formation of collagen and elastic fibers. The fibro-cellular thickening of the intima was further augmented, and, in addition, a complete stenosis of the small lumen of the small artery was observed. Medial thickness did not show significant differences between the control and exposed groups for either exposure duration. The whole blood viscosity was significantly increased by the 90-d but not by the 30-d exposure. These results suggest that there are some relationships between the increase in whole blood viscosity and the intimal thickening of some small arteries in the exposed site.

Surgical treatment of congenital dilatation of bile duct (choledochal cyst) with technical considerations.

Since the concept of an anomalous junction of the pancreaticobiliary ductal system was introduced, there has been rapid progress in the study of the pathophysiology and the surgical treatment of choledochal cyst, or congenital dilatation of the bile duct. Because of its various disadvantages, cystoenterostomy as a routine procedure for treatment of this condition has been superseded by hepaticoenterostomy after removal of the devastated bile ducts with blocking of communication with the pancreatic ducts. This article describes the technical details of the surgical correction of choledochal cyst.

Nuclear-magnetic-resonance studies on the conformation of membrane-bound alpha-mating factor. Transferred nuclear Overhauser effect analysis.

The C-H proton resonances of alpha-mating factor, yeast pheromone, in 2H2O solution were assigned. The phase transition temperature of perdeuterated dipalmitoylglycerophosphocholine (suspension) was found to be 35.5 degrees C. In the presence of vesicles of this phospholipid, the exchange broadening and transferred nuclear Overhauser effect (TRNOE) of peptide proton resonances (at 50 degrees C) were analyzed. The mode of binding of this peptide with the phospholipid bilayer was elucidated. The N-terminal nine residues (Trp1-Gly9) are tightly bound to the bilayer, while the C-terminal four residues (Gln10-Tyr13) are left free in aqueous phase. This is consistent with the previous observation that the C-terminal three residues (Pro11-Tyr13) are not essential for the activity of this pheromone [Masui, Y. et al. (1977) Biochem. Biophys. Res. Commun. 78, 534-538]. Furthermore, from the TRNOE analyses, the conformation of the membrane-bound N-terminal part of alpha-mating factor was elucidated; the residues Trp1-Gln5 form a compact helical structure while the residues Lys7-Gly9 form an extended structure. A similar TRNOE was also observed for an active decapeptide analog Trp1-Gln10. This confirms the previous conclusion that the physiological activities of this pheromone and analog peptides are correlated with the conformations of membrane-bound peptide molecules [Higashijima, T. et al. (1983) FEBS Lett. 159, 229-232].

Effects of dietary histidine and methionine loading in rats with a portacaval shunt.

To elucidate disturbances of brain amine metabolism in hepatic coma, the effects of dietary histidine and methionine loading on the brain levels of amino acids and transmitter amines and on behavior were examined in rats with a portacaval shunt (PCS rats). Surgical construction of a portacaval shunt exaggerated the increase in brain histamine caused by dietary histidine loading 4 weeks after operation. Although the marked increase in the brain level of methionine itself did not affect on the brain levels of catecholamines, serotonin, and histamine, brain level of 5-HIAA was decreased in PCS rats on methionine-enriched diet. Diminished spontaneous activity was observed in PCS rats on either diet, which could not be related to the disturbance of brain amine metabolism caused by excess histidine and methionine. These results indicated that histamine was involved in the derangement of brain amine metabolism in PCS rats, and that direct effect of dietary methionine loading on the brain levels of transmitter amines was not observed in PCS rats. The etiological relation of these substances to hepatic coma should be further investigated.

[Changes in the forearm tcPO2 following the cold water immersion test].

The purpose of this study is to apply changes in forearm transcutaneous PO2 (tcPO2) during the cold water immersion test (5 degrees C, for 10 min) to the diagnosis of vibration syndrome. Forearm tcPO2 in healthy controls increased gradually up to 3 min after the start of the cold water immersion and decreased thereafter. It returned to almost the same level before immersion 5 min after the start. In the workers using vibrating tools manifesting Raynaud's phenomenon, forearm tcPO2 also increased up to 3 min after the start as in healthy controls, but no change was observed thereafter. The difference between the level of forearm tcPO2 at 3 min and that at 10 min after the start of the immersion (Forearm tcPO2 recovery index, newly devised by authors) in workers using vibrating tools was significantly lower than that in healthy controls. Therefore, investigation of changes in forearm tcPO2 following the cold water immersion test is considered to be a useful objective item which can contribute to the diagnosis of vibration syndrome.

Serum levels of coenzyme Q10 and lipids in patients during total parenteral nutrition.

Serum levels of coenzyme Q10 (CoQ10) as well as lipids were determined in patients during total parenteral nutrition (TPN). The mean CoQ10 levels (M +/- SD) were 0.77 +/- 0.30 microgram/ml for 108 normal subjects and 0.59 +/- 0.35 microgram/ml for 95 patients before TPN. The mean CoQ10 level of the patients decreased significantly to 0.35 +/- 0.23 microgram/ml one week after the start of TPN, and then remained almost unchanged during TPN for up to 6 weeks. When the patients receiving TPN (TPN patients) were grouped according to their clinical diagnoses, the mean CoQ10 level of patients with cancer was significantly lower than that of the other patients without cancer in 4 week therapy, but there was no difference in the levels between the patients with and without diseases of the gastrointestinal tract. Serum levels of total cholesterol (T-Chol) and esterified cholesterol in TPN patients also declined below their respective normal ranges, but not to the same extent in comparison to CoQ10. The levels of triglycerides (TG), phospholipids (PL), non-esterified fatty acids, low density lipoproteins, very low density lipoproteins, chylomicrons, and cholesterol in the high density lipoprotein fraction in serum of TPN patients were within their normal ranges. The levels of CoQ10 in TPN patients were correlative to those of T-Chol, TG, and PL, and decreased rapidly prior to the latter levels.
