Br/Rb ratio obtained by XRF analysis of kidneys of normal and tumor-bearing mice treated with cis-DDP.

The Br/Rb ratio in kidneys of normal and tumor-bearing mice was studied and a significant difference was found between the two groups. The mice were treated with cis-DDP with and without selenite, with selenite alone, and compared with controls. The animals were sacrificed 7d, 14d, and 21-28d after treatment. The kidneys were removed and subjected to Br and Rb determination by a novel X-ray fluorescence (XRF) method. The results are presented in terms of estimated net count rates under the BrK alpha and RbK alpha peaks. No significant differences were found in Br and Rb levels or in the Br/Rb ratio in kidneys of treated and untreated mice belonging to the same group-normals or lung carcinoma inoculated. However, there was a significant difference in the Br/Rb ratio between the two tested groups (normals and inoculated). Preliminary results on the Br/Rb ratio obtained from normal and malignant tissues of colorectal cancer (CRC) patients and from mice inoculated with melanoma, show the same trend as those from the group of mice with lung carcinoma. In all three groups tested the Br/Rb ratio was 2-4 times higher in normal than in malignant tissue.

Percutaneous exposure to ethyl parathion in a feral Griffon vulture (Gyps fulvus).

A feral Griffon vulture (Gyps fulvus) was found with tremors, weakness, digit and wing flexion, and an inability to fly. A zero blood cholinesterase activity and a favorable response to treatment with pralidoxime hydrochloride indicated exposure to an anticholinergic pesticide. The bird died after 7 d, and traces of the organophosphate insecticide ethyl parathion were found in the liver and from a blue discolored skin area of the neck. Continuous exposure to ethyl parathion through dermal absorption was presumed the cause of death of the vulture.

Myocarditis following envenoming with Vipera palaestinae in two horses.

Cardiac disease developed several days to weeks after resolution of local inflammation caused by envenoming by Vipera palaestinae in two horses. In one horse (case A), referred to the hospital for recurrent abdominal pain 8 days after envenoming, a tachyarrhythmia was detected; ventricular premature depolarizations were diagnosed by ECG. A second horse (case B) was found dead without premonitory signs 60 days after envenoming. In both horses, there was extensive necrosis of cardiac ventricular tissues.

Effects of high-fiber diets on pathological changes in DMH-induced rat colon cancer.

The protective role of dietary fibers on tumorigenic effects of 1,2-dimethylhydrazine (DMH) on the rat colon was studied using histochemical, immunohistochemical, and biochemical analyses. Rats were injected with DMH (20 mg/kg sc) for five weeks, once a week, and were fed laboratory chow (Control I), semisynthetic fiber-free diet (Control II), or 18% or 25% assorted fiber (cellulose, beans, corn) diet. The rats were sacrificed 12, 16, and 24 weeks after the carcinogenic injections. Adenomatous tumors developed in 90-100% of the animals fed chow or fiber-free diets. A diet with high concentrations (25%) of corn dietary fiber significantly decreased the tumor incidence (40% and 42%) and tumor yield (p < 0.01). Diets with lower concentrations of fibers (18%) did not protect against tumorigenic effects of DMH. No differences were found in pathological parameters of tumors obtained from different dietary groups: the damage index (percentage of damaged cells per section) was very similar in all groups studied. Diets enriched with dietary fibers (25%) had a significant protective effect in DMH-induced rat colon cancer.

Perianal squamous cell carcinoma in goats.

Two cases of squamous cell carcinoma arising from the perianal skin of aged female Sannen crossbred goats are described. The occurrence of a tumour in this specific location is very rare, and the presence of the identical tumour in two animals of the same herd is unusual.

Intratubular spermatic seminoma in a Fischer-344 rat.

A case of a spontaneous intratubular spermatic seminoma is described in a 98-wk-old Fischer-344 (F-344) rat. The differential diagnosis of spermatic seminoma from the other morphological forms of the tumor was based on the recognition of 3 cell types. The tumor cells were positive for S-100 antigen and cytokeratin. To the best of our knowledge, this is the second case of a seminoma reported in an F-344 rat and the first dealing with electron microscopic and immunohistochemical features of this tumor.

Polarization microscopy of picrosirius red-stained collagen from oxodipine-induced hyperplastic gingiva of beagle dogs.

Using the model of oxodipine-induced gingival hyperplasia in beagle dogs, we investigated the collagen fibres of the hyperplastic gingiva by measuring the polarization colours from Picrosirius-red stained sections. The predominant polarization colours of the thick collagen fibres (1.6-2.4 microns diameter) of male oxodipine-treated dogs were greenish-yellow to yellow, while in controls and in female oxodipine-treated dogs, the same size fibres mostly appeared orange and red. These differences may be indicative of gender variation in the genesis of hyperplastic gingiva in calcium channel blocker treated dogs.

Possible pitfalls in rat extended dermal toxicity testing: an hepatic-ocular syndrome.

During a sub-chronic dermal toxicity study, hepatic and ocular toxicity were detected unrelated to the compound being tested. The changes in the liver were characterized by hepatic centrilobular degeneration and fibrosis due to chronic passive congestion. Clinical chemistry results confirmed the active hepatic damage and cholestasis. The ocular changes were limited to the retina where diffuse retinal atrophy was seen originating adjacent to the optic nerve and progressing centrifugally. It is suggested that as a consequence of the bandaging technique, a state of passive congestion and cholestasis resulted. The decrease in flow of bile acids to the intestine caused disturbances in vitamin E absorption. The deficiency of the antioxidant vitamin was responsible for the retinal changes seen.

A case of natural hydatidosis in a baboon.

Four large and several smaller cysts of Echinococcus granulosus were found in the liver of a baboon submitted for necropsy. Three of the large cysts had an atypical structure and were packed with a semi-solid mass of collapsed daughter cysts, small cysts containing fluid and involuted membranes. This atypical structure has been reported previously though only in baboons.

Use of dual-energy X-ray absorptiometry (DEXA) to follow mineral content changes in small ceramic implants in rats.

Dual energy X-ray absorptiometry (DEXA) imaging was used to quantify bone ingrowth into hydroxyapatite (HA) ceramic implants in rats. HA cylinders implanted in the proximal tibiae were followed for 13 wk. The increase in alkaline phosphatase (ALP) activity within the implants preceded the increase in mineral content as measured non-invasively by the DEXA technique. This was consistent with the timing of ALP activity in respect of mineralization as they occur during fracture healing. The results show that DEXA imaging is useful in measuring bone ingrowth into small ceramic HA implants in vivo, despite the high mineral content background of the implant scaffold.

Concurrent Hepatozoon canis and Toxoplasma gondii infections in a dog.

A female 1-year-old dog died suddenly and was submitted for necropsy. Numerous grey-tan-colored nodules were seen in the lungs, brain and lymph nodes. Microscopically, the predominant lesion was necrosis associated with numerous Toxoplasma gondii tachyzoites. The parasites reacted positively with anti-T. gondii serum in an immunohistochemical test. Schizonts of Hepatozoon canis were seen in sections of lymph nodes and the spleen.

The fate of altered hepatocytic foci as a result of treatment with oxodipine, a calcium channel blocker.

The dietary administration of the calcium channel blocker oxodipine to Fischer (F344) rats for 12 and 30 months resulted in increased incidence of altered hepatocytic foci (AHF). As the Environmental Protection Agency (EPA) regards AHF as potentially precancerous it is important to accumulate experimental evidence which may negate this theory. In the case of oxodipine we proved that with dosages close to maximum tolerated dose (MTD) for prolonged periods no hepatic neoplasms were produced. The possible nature of such AHF is discussed.

Morphological, histochemical and immunohistochemical differences between tumorous and adjacent tissues in chemically induced colon cancer in rats.

Methods of morphological, histochemical and immunohistochemical analyses were used to further characterize differences between tumourous and adjacent grossly normal tissues in chemically-induced colon cancer in rats. Colon tumors were induced by the treatment of rats with 1,2-dimethylhydrazine or with N-methyl-N'-nitro-N-nitrosoguanidine alone or with subsequent treatment with deoxycholic bile acid. Tissues were studied morphologically (for the presence of goblet cells in the colon crypts, and the extent of infiltration of lymphocytes into the crypts and between them), histochemically (for the presence of positive reaction to neutral and acid mucopolysaccharides) and immunohistochemically (for the presence of tissue polypeptide antigen). All data were evaluated quantitatively, and index of tissue damage was calculated for both tumorous and non-tumorous tissues. Significant morphological differences were found between tumorous and adjacent apparently normal tissue. Histochemically and immunohistochemically, both types of tissue reacted very similarly to exposure to the carcinogens. Index of damage was significantly different from normal untreated colon in both kinds of tissue. It was suggested that precancerous state in tissue adjacent-to-tumor could be detected using the combination of these methods.

Individual variability of pathological parameters in chemically induced rat colon tumors.

The development of tumorigenic conditions in the carcinogen-exposed rat colon was studied using selected morphological, histochemical, immunohistochemical and biochemical methods of analysis. Rats were treated with two carcinogens: 1,2-dimethylhydrazine and N-methyl-N'-nitro-N-nitrosoguanidine alone or with deoxycholic acid as a tumor promoter. It was found that 3 months after treatment of animals with the carcinogens the following changes were developed in colonic tissue: infiltration of lymphocytes in the mucous membrane, high increase in mitotic index among epithelial cells, negative reactions of colonic cells for neutral mucopolysaccharides and sulfomucins and positive reactions to carboxyl groups, nonsulfated acid mucosubstances and tissue polypeptide antigens. An increase in the activity of ornithine decarboxylase in colonic tissue was developed within the same time period and has been seen only in those tissues which were characterized by the development of precancerous conditions. Individual variations were observed in the manifestation of the studied parameters in rat neoplastic colonic tissues. It is suggested that these differences reflect an individual sensitivity of animals to carcinogens and the magnitude of the dysplastic processes induced in the colon.

Thickening of the adrenal zona glomerulosa in dogs induced by oxodipine, a calcium channel blocker.

Subchronic effects of oxodipine, a calcium channel blocker affecting the adrenal gland of the dog, are described. Thirteen wk of treatment at a high dose (24 mg/kg/day) of oxodipine resulted in drug-induced thickening of the zona glomerulosa and increased stimulation of its secretory activity. It is postulated that subchronic administration of oxodipine at this dosage resulted in a decrease in blood pressure, with uninterrupted stimulation of the adrenal zona glomerulosa to release aldosterone, causing an increase in the width of this portion of the gland involving cellular hyperplasia. Support for this indirect effect is found in the increased presence of renin granules in the juxtaglomerular apparatus.
