Chromosome delineation induced by a combined potassium permanganate-sodium bisulfite treatment.

A cytological procedure for in vitro chromosome delineation has been studied using human and mouse (Mus musculus) chromosomes. This method, consisting of slide incubation in KMnO4 at 0--5 degrees C for 24 h followed by a short exposure to NaHSO3 (1--3 min) and Giemsa staining, induces extraction of chromatin from human and mouse interphase nuclei and chromosomes. Autoradiography after 3H-ThD incorporation in vitro and cytophotometry confirmed that DNA is removed. Well contour-delineated and non-distorted chromosomes are observed in both species allowing the identification of all human chromosome groups. Contour chromosome delineation and its relationship to chromosome organization is briefly discussed.

Acetate intolerance during hemodialysis.

Acetate is frequently substituted for bicarbonate in hemodialysis solutions. Plasma acetate and bicarbonate concentrations were measured in nine patients with chronic renal failure undergoing hemodialysis with dialyzate containing acetate. In three patients (2 children and 1 adult) plasma acetate concentrations exceeded 15 mEq/liter during the dialysis. The mechanisms leading to acetate intolerance are probably multiple. It cannot be assumed that dialysis with acetate containing solutions will restore the buffer anion deficit characteristic of chronic renal failure.

Use of fiberoptic bronchoscopy in asthmatic children with lung collapse.

Segmental atelectasis during asthmatic attacks as a consequence of bronchospasm and hypersecretion are very frequent in children. The collapse of a lung instead is a very rare occurrence. We report two patients with acute exacerbation of asthma, showing an in complete response to bronchodilatory therapy. The clinical findings and chest X-ray were suggestive for collapse of a lung. An urgent fiberoptic bronchoscopy revealed obstruction of a stem main bronchus caused by sticky mucopurulent secretions. The aspiration improved dramatically the respiratory distress and the radiological picture. In these cases we recommended to perform an immediate bronchial aspiration by means of a flexible instrument without waiting a further deterioration of respiratory function before taking this decision.
