Rapid purification of human erythrocyte 6-phosphogluconate dehydrogenase by means of affinity chromatography.

A rapid two-step procedure is reported by which homogeneous 6-Phosphogluconate dehydrogenase can be isolated from human erythrocytes. This method is based upon direct affinity chromatography of the hemolysates on adenosine 2',5'-bisphosphate-agarose (yielding a 4,500-fold purification), followed by anion exchange chromatography on a micro-column of DEAE-Sephadex. The present method represents a considerable simplification over previously available procedures for the purification of this enzyme protein from human erythrocytes.

[Changes in the level of erythrocyte phosphorylated compounds in patients with hepatic encephalopathy].

Automated ion exchange chromatography was used to compare red cell phosphorylates in clinically healthy subjects and 6 patients with hepatic encephalopathy. Significant changes in the distribution of these compounds were noted, including a marked increase in total acid-soluble content (particularly 2.3 DPG, R5P, 3MPG and G16DP) and a sharp fall in ATP. Increased 2.3 DPG explained the rightward shift of the haemoglobin dissociation curve seen in cirrhosis of the liver, but does not seem to fit in with enhanced blood ammonia. Deep-seated changes in red cell energy metabolism may have the same pathogenesis as the CNS metabolic change observed in hepatic encephalopathy.

[Inverted papilloma of the bladder (Report of a clinical case)].

Inverted papilloma of the urinary bladder is a rare variant of the common transitional cell papilloma. This tumor, although histologically distinctive and clinically benign, has often been pathologically mistaken for a low grade transitional cell carcinoma. For this reason it seems appropriate to document one case seen in recent years at Maria Vittoria Hospital - Turin.

Whipple's disease: a continuous challenge.

Whipple's disease is a rare disease with protean clinical manifestations, often mimicking those of other pathological conditions. We describe two new cases, one admitted to hospital only after Giardia lamblia infestation had drawn attention to gut symptoms, and the other who was treated for a long time with steroids for suspected Horton's arteritis. Once again, we stress the importance of bearing this polymorph disease in mind, especially in older people.

[Chronic therapy with ursodeoxycholic acid in a child with Alagille syndrome].

The effectiveness of oral bile acid therapy with ursodeoxycholic acid (10 mg/kg/day) was investigated in a 10 year old boy affected by Alagille's Syndrome, a chronic cholestatic disorder due to congenital hypoplasia of the intrahepatic biliary ducts. Cholestatic and hepatonecrotic indices were measured before and during ursodeoxycholic acid therapy and 1 month after stopping and 36 months after restarting the treatment. Ursodeoxycholic acid led to a marked improvement in the cholestatic and hepatonecrotic parameters which was maintained during all the treatment phase. Pruritus and steatorrhea disappeared during the treatment with ursodeoxycholic acid. Histological examination of the liver biopsy after the treatment revealed a disappearance of the biliary plugs but without increasing he intrahepatic bile ducts. The results suggest that ursodeoxycholic acid may improve the condition of the children affected by Alagille's Syndrome, specially when the liver transplantation is required, and indicate a need for long term studies in a larger number of patients.

Endoplasmic reticulum Ca(2+)-ATPase in microsomal vesicles isolated from bovine retinae.

Active Ca2+ transport was measured in microsomal vesicles prepared from bovine retinae and was compared with that in disk membranes of the photoreceptor cells of the same retina. The 45Ca uptake was dependent on the presence of Mg(2+)-ATP and was inhibited by vanadate or when GTP substituted for ATP. The dependence of calcium uptake on the external free Ca2+ concentration gave a KM = 13 microM or a KM = 0.1 microM for disks and microsomal vesicles, respectively. A phosphorylated intermediate (E-P) of Ca(2+)-ATPase of about 100 kDa was isolated in microsomal vesicles. The E-P formation was strongly inhibited by thapsigargin and partially by 2,5-di-(-butyl)benzohydroquinone. Digestion of disks or microsomes with calpain had no effect on the phosphorylated intermediate, while digestion with trypsin produced two fragments of approximately 55 kDa and 35 kDa. These results suggest that bovine retinal microsomes contain a calcium pump belonging to the SERCA family.
