Calcium-calmodulin-dependent phosphorylations in the control of muscular contraction?

Muscular contraction is triggered by the increase in free calcium concentration and modulated by cyclic nucleotide-dependent phosphorylation. Beside a direct trigger of sarcomeric muscle contraction through binding of troponin C, calcium ions trigger or modulate contractility through calcium-calmodulin-dependent myosin light chain kinases, and increase the rate of relaxation through the calmodulin-dependent phosphorylation of phospholamban, the activator of the cardiac sarcoplasmic reticulum calcium pump. In both cases, a concerted regulation by calcium and cyclic nucleotides is observed. Hyperactivation of the calcium pump is brought about by additional phosphorylation of phospholamban by cAMP-dependent protein kinase. Similarly myofibrillar myosin light chain kinases from smooth and skeletal muscles are substrates of the cAMP-dependent protein kinase. The calmodulin-dependent protein kinases are probably organized into supramolecular regulatory complexes.

Estimation of bovine colostral immunoglobulins by refractometry.

The total protein of colostral whey from dairy cows as determined by a refractometer were compared with the immune globulin concentration obtained by cellulose acetate membrane electrophoresis and the immunoglobulin (IgA + IgG + IgM) contents determined by radial immunodiffusion. The coefficient of correlation between the results obtained by refractometry and electrophoresis was 0.98 (P < 0.001). The correlation between refractometry and radial immunodiffusion was 0.89 (P < 0.001), while that of electrophoresis and radial immunodiffusion was 0.87 (P < 0.001). Refractometry can be used as a simple, fast and inexpensive indirect method of assessing immunoglobulins in colostral whey.

Critical assessment of small bowel biopsy in children.

176 biopsies of small bowel mucosa taken during a 5-year period from 162 children were reviewed, and 92 of them were subjected to quantitative measurement using surface/volume ratio and intraepithelial lymphocyte counting. There was good general agreement between stereomicroscopical appearance of fresh tissue, routine pathology reporting, and our independent histological assessment. Most of those in which there was divergence of opinion (11--15%) had intermediate mucosal changes. There was a significant difference between the surface/volume ratio for each of the three grades of histology (P less than 0.001, less than 0.001, less than 0.01), indicating a relationship between qualitative and quantitative methods of assessment. The range of measurements in intermediate biopsies overlapped those of the other two grades. In the 92 biopsies there was a correlation between surface/volume ratio and intraepithelial lymphocyte count (r = -0.56; P less than 0.001) which may reflect our clinical experience, in that 88% of the biopsies were graded as either normal or flat (probable coeliac disease). Very short children had shorter villi than those with nonspecific diarrhoea and a more normal height.

Immunoglobulin levels in calves fed colostrum by stomach tube.

Three female and three male unsuckled newborn Holstein calves were tube-fed pooled colostrum and fresh colostrum respectively at a mean dose of 81.1 ml per kg birth weight at about six hours of age. The pre-colostral mean total serum immunoglobulin gradually increased from 0.07 mg per ml to 31.73 mg per ml by one day of age. IgG1, IgG2, IgG and IgM gradually increased to maximum concentrations by one day of age. IgA was higher in the 14 hour than in the 24 hour sample due to catabolic loss. The pre-colostral packed cell volume (PCV) of 41.83 per cent significantly (P less than 0.05) decreased to 37.17 per cent 18 hours after intubation. The pre-ruminant rumen of the newborn calf can empty itself efficiently and adequate intestinal absorption of colostral proteins can take place before closure sets in. Normogammaglobulinaemia can be established in almost all normal calves and neonatal morbidity and mortality reduced by force-feeding newborn calves with good quality colostrum immediately after birth.

The antigenic determinant of fibrin(ogen) measured with agglutination inhibition immunoassays.

Agglutination inhibition immunoassays using antifibrinogen antiserum measure an antigenic determinant of fibrinogen which is located on the fragment D parts of this molecule. This antigenic determinant of fragment D, in contrast to others, does not form an immune precipitate with antifibrinogen antiserum. Upon very extensive hydrolysis by plasmin, this antigenic determinant has a reduced affinity for its antibody.
