[Open heart surgery for removal of secondary cardiac tumors].

Recent remarkable development of the non-invasive diagnostic apparatus such as two dimensional echocardiography, computed tomography and RI angiography improves early diagnosis for secondary cardiac tumors and promotes surgical treatments for them. Five cases of secondary intracardiac tumors underwent open heart surgery using cardiopulmonary bypass with successful removal. Although a case of direct invasion of a malignant thymoma into the pericardium, RA and SVC was lost at ICU after surgery because of massive hemorrhage followed by LOS, another 4 cases, i.e., a 27 year-old female with implantation of LA myxoma into the RA and RV, a case with intracavitary extension of leiomyosarcoma of the IVC, a case of intravenous extension of the uterine leiomyomatosis through the IVC to the pulmonary artery and a case of intravenous extension of chondrosarcoma of the left fibula to the heart are alive actively. Open heart surgery has become acceptable treatment for cases with secondary intracavitary cardiac tumors in order to rescue emergency cases with circulatory obstruction by the tumor mass, to make sure a pathological diagnosis of unknown origin and to promote further chemotherapy and/or radiation therapy after surgery. Our surgical results may support these active treatments for secondary cardiac tumors.

Photolysis of pyridinoline, a cross-linking amino acid of collagen, by ultraviolet light.

Pyridinoline, a cross-linking amino acid of collagen, was degraded by irradiation of ultraviolet light. The decomposition rate varied with pH of the solution and wavelength of irradiation light. The maximum of the degradation rate at individual pH coincides with the ultraviolet absorption maximum. Namely, it was maximally degraded by irradiation at 295 nm in acidic solution and at 325 nm in neutral and alkaline solution. At the optimum wavelength, the photolysis occurred more rapidly in neutral and alkaline solution than in acidic solution. The quantum yield in neutral solution was approximately 0.11 and independent of wavelength. One of the photolysis products was identified as hydroxylysine on an amino acid analyser, indicating that the cleavage of the pyridinium ring occurred.

Left ventricular morphometric estimation of the coarctation complex in the first year of life.

Coarctation complex is a serious congenital cardiovascular malformation in infants. It is associated with severe congestive cardiac overload and high mortality even after surgical treatment. To investigate left ventricular performance, quantitative morphometry and histometry of the left ventricle were carried out in 19 autopsied patients who died without surgical intervention. The following results were obtained: 1. Coarctation of the aorta was found to selectively affect the left ventricle, showing a volume overload response without effectively adapting to the pressure overload; the right ventricle showed a significant reaction to the pressure overload. 2. The coronary arterioles responded to the pressure load in the left ventricle. In order to promptly recover left ventricular function, simple coarctectomy without VSD closure or other procedures is advisable for treatment of the coarctation complex in the early months of life.

Expression in Escherichia coli of chemically synthesized gene for a human immune interferon.

A 450 base pair fragment of double stranded DNA consisting of a gene for a human immune interferon (146 amino acid residues), initiation and termination signals plus appropriate restriction endonuclease sites was totally synthesized. The synthesis involved preparation of 62 oligodeoxyribonucleotides by rapid, solid phase procedures, and enzymatic ligation of the oligonucleotides. This synthetic gene was expressed in E. coli under the control of the lac UV5 promoter. Molecular weight of IFN-gamma produced by E. coli was estimated to be about 32,000 and 17,000 dalton by gel filtration and SDS-polyacrylamide gel electrophoresis respectively, suggesting that a dimer-form molecule was expressed in E. coli.

Effect of L-glutamic acid on [14C]GABA release from isolated rat retina.

The effect of various putative neurotransmitters on the release of [14C]GABA taken up by isolated rat retina was investigated by a perfusion technique. L-Glutamic acid initially enhanced the GABA release and subsequently reduced it to a level below the baseline rate. This enhancing effect of L-glutamic acid was Ca2+-dependent and tetrodotoxin-sensitive. L-Aspartic acid only reduced the GABA release, whereas kainic acid exhibited only a marked enhancement of its release. Acetylcholine, norepinephrine, dopamine, glycine and taurine did not have any significant effects on GABA release.

Regulation of vitamin B12 and bacteriochlorophyll biosynthesis in a facultative methylotroph, Protaminobacter ruber.

The regulation of vitamin B12 and bacteriochlorophyll formation was studied in a facultative methylotroph, Protaminobacter ruber, classified as a non-photosynthetic bacterium. Vitamin B12 was formed at an almost constant level under various cultivation conditions, while bacteriochlorophyll synthesis was drastically influenced by the growth conditions. Addition of B12 and hemin to the medium and the change of culture conditions from light to darkness in the early growth phases stimulated the pigment synthesis, but no pigment was formed under the continuous illumination and the addition of chloramphenicol inhibited the pigment formation. delta-ALA synthase and delta-ALA dehydratase activities were demonstrated in P. ruber. delta-ALA synthase formation was induced when cultures were transferred from light to dark conditions, which promoted the bacteriochlorophyll formation. Bacteriochlorophyll-protein complex had an absorption maximum at 870 nm and a very small peak at 800 nm.

The effect of antivitamin B6 administration on gamma-aminobutyric acid metabolism in retina and electroretinogram.

The effect of several antivitamin B6 on gamma-aminobutyric acid (GABA) metabolism was studied in the rat retina. The rat electroretinogram (ERG) was also recorded after administration of these drugs. Aminooxyacetic acid (AOAA) and hydrazine administration increased the GABA content and inhibited the GABA degrading enzyme, GABA transaminase in retina. In addition, there drugs elongated the peak latency of the oscillatory potential in the rat ERG. In contrast, 4-deoxypyridoxine (DOP) or isonicotinic acid hydrazide (INAH) administration decreased the GABA content and inhibited the GABA synthesizing enzyme, glutamic acid decarboxylase in retina, and administration of these drugs together with AOAA lessened the degrees of elevation of GABA content and of the elongation of the peak latency produced as compared with AOAA alone, though neither of the former drugs had a significant effect on ERG. The retinal GABA seems to play an important role in relation to the oscillatory potential of ERG.
