Ultrastructure of rat renal tubular basement membrane--meshwork structure demonstration by negative staining.

The tubular basement membrane (TBM) (i.e. tubular basal lamina) of rat kidney was shown to be a fine meshwork by electron microscopy after negative staining. Strands of the meshwork formed a regular three dimensional lattice work. The pores of the meshwork were polygonal. There were two main pore sizes: one approximately 30 A in diameter, the other 42--60 A. In view of our previous observation that glomerular and alveolar basement membranes were made up fine meshwork, it is quite possible that the basement membranes of other organs are also made up such fine meshwork.

Study on the specificity of Shore's method for the determination of histamine in various tissues.

Specificity of Shore's method for the histamine determination was investigated by examining the fluorescence spectrum of the extract from rat gastric mucosa, biopsy specimen of human gastric mucosa, human gastric juice and human whole blood. An almost identical spectrum to that of pure histamine was obtained with the extract from rat gastric mucosa and human whole blood, whereas the spectrum with the extract of human gastric biopsy specimen and human gastric juice was different from that of histamine. By means of high-performance liquid chromatography, an approximately three times more concentrated extract from human gastric mucosa gave a distinct peak of histamine while a hundred times concentrated gastric juice gave no measurable peak of histamine. Fluorescence spectrum of the substances which might be extracted with histamine was examined. The ratio of the fluorescence of histidine to that of equimolar concentration of histamine at the wave length for histamine determination was 4.62%. From the above results, it has been concluded that Shore's method is useful for the determination of histamine contents in the gastric mucosa and whole blood if approximately 5% contamination with histidine is taken into consideration.

A double-blind comparative study of the inhibitory effect of intraduodenally administered hyoscine-N-butylbromide on human duodenal motility.

In a double-blind crossover study hyoscine-N-butylbromide (HBB) was compared with placebo in its inhibitory effect on duodenal motility stimulated by metoclopramide. Measurements were made both by the balloon technique and the open-tipped tube method with pressure transducer. 20 mg or 50 mg HBB or placebo were instilled in random sequence into the intestinal tract of 12 healthy volunteers. The frequency of Type II waves was determined in each experiment after administration of the test substance following prior metoclopramide stimulation. This frequency was used as the inhibitory index for each drug. The test substance was administered 15 minutes before a second stimulation. A four-way analysis of variance (mixed model) was applied to the calculated number of waves per period. Subjects were required to respond equally to stimulation with increased motility on both trial days as evidenced by a minimal difference in the averages following the first stimulation on each trial day and before application of the test substance. The frequency of Type II waves after 20 mg and 50 mg HBB was reduced in a statistically significant manner in each case when compared with placebo. The probabilities of error were p less than 0-05 and p less than 0-025.

Electron microscopic demonstration of meshwork structure in human and bovine glomerular basement membranes.

With the electron microscopic and the negative staining method, the glomerular basement membrane of human and bovine kidneys was shown to have a definite fine meshwork structure. The pores of the meshwork of bovine glomerular basement membrane appeared to be pentagonal or hexagonal in shape. Strands of the meshwork branched three-dimensionally and made up the whole basement membrane. The portion of the sgrand between two neighboring branches was presumed to be a structural unit of the basement membrane. Glomerular basement membrane in man showed a structure similar to that seen in cattle, although the pore size of the meshwork was smaller in man than in cattle.
