In mice repeated administration of electroconvulsive shock or desmethylimipramine produces rapid alterations in 5-HT2-mediated head-twitch responses and cortical 5-HT2 receptor number.

Mice treated with a single electroconvulsive shock (ECS) daily for up to 3 days showed progressively enhanced head-twitch to 5-methoxy-N,N'-dimethyltryptamine 24 h after 1,2 and 3 shocks. Injection of desmethylimipramine (DMI, 5 mg/kg) twice daily for up to 3.5 days progressively decreased head-twitch responses on days 2, 3 and 4. Cortical [3H]ketanserin binding was increased by ECS X 3 and decreased by 3.5 days DMI treatment. These data demonstrate that in mice antidepressant treatments produce rapid changes 5-HT2 function.

The effects of the GABA-mimetic drugs, progabide and baclofen, on the biochemistry and function of 5-hydroxytryptamine and noradrenaline.

Administration to mice of a single dose of (+/-)-baclofen (5 mg/kg) or progabide (100 mg/kg) significantly inhibited the head-twitch response mediated by 5-hydroxytryptamine (5-HT2) receptors 30 min (but not 3 hr) later, when the response was produced by injection of carbidopa (25 mg/kg) plus 5-hydroxytryptophan (5-HTP; 100 mg/kg). No change was seen in the head-twitch response when induced at this time by 5-methoxy-N,N-dimethyltryptamine (5-MeODMT; 5 mg/kg). Inhibition of the head-twitch response after injection of 5-HTP was produced by pretreatment with (-)-baclofen, but not (+)-baclofen; injection of (+)-baclofen with the (-)-baclofen did not alter the attenuation of the behaviour produced by the active isomer. Twenty-four hours after the last injection of progabide, given repeatedly (100 mg/kg injected 5 times over 10 days) specific binding of [3H]ketanserin in the frontal cortex was enhanced and the head-twitch response to both 5-HTP and 5-MeODMT was markedly increased. The sedation response mediated by alpha 2-adrenoceptors, which followed the injection of clonidine (0.25 mg/kg) was attenuated. Repeated administration of baclofen (10 mg/kg per day in drinking water) also increased the number of 5-HT2 receptors in the frontal cortex (16%) and enhanced the head-twitch behaviour after injection 5-HTP or 5-MeODMT. Clonidine-induced sedation, number of beta-adrenoceptors in the cortex and apomorphine-induced locomotor activity were all unchanged.(ABSTRACT TRUNCATED AT 250 WORDS)

The administration of baclofen to mice increases 5-HT2-mediated head-twitch behaviour and 5-HT2 receptor number in frontal cortex.

Mice were injected with baclofen (10 mg/kg) and then given baclofen in drinking water (10 mg/kg/day). After 1 day of administration of baclofen the head-twitch response to the precursor 5-hydroxytryptophan (5-HTP) was reduced but the response to the agonist 5-methoxy-N,N-dimethyltryptamine (5-MeODMT) was unaltered. However, after 14 days the head-twitch response to both 5-HTP and 5-MeODMT was enhanced and this enhancement was present for 14 days after drug withdrawal. After 14 days of administration of baclofen the number of 5-HT2 receptor binding sites in frontal cortex (labelled by [3H]-ketanserin) was also elevated. It is suggested that the enhanced 5-HT2 function, following longer-term administration of baclofen is the consequence of the drug inhibiting 5-HT release in vivo, as indicated by the observations after acute administration.

Intracerebroventricular administration of 5,7-dihydroxytryptamine to mice increases both head-twitch response and the number of cortical 5-HT2 receptors.

5-Hydroxytryptamine-containing (5-HT) neurones in brain of the mouse were selectively destroyed by intracerebroventricular injection of 5,7-dihydroxytryptamine (5,7-DHT, 50 micrograms). Sham-lesioned controls received vehicle (2 microliters, i.c.v.). Two weeks later the head-twitch response induced by 5-methoxy-N,N-dimethyltryptamine (2.0 mg/kg) and mediated by 5-HT2 receptors was markedly enhanced in the lesioned mice. Furthermore, lesioning also increased 5-HT2 binding sites in the cortex, assessed by the binding of [3H]ketanserin in these animals, and decreased levels of 5-HT in the cortex (70%) and mid/hindbrain (64%) regions. A second group of mice, lesioned with less 5,7-DHT (5-20 micrograms, i.c.v.) showed unaltered head-twitch responses to 5-methoxy-N, N-dimethyltryptamine (2.0 mg/kg) and did not have increased 5-HT2 receptor binding in the cortex. Depletions of 5-HT were 32 and 40% in the cortex and mid/hindbrain, respectively. Comparison of the extent of depletion of 5-HT in the mid/hindbrain after lesioning, with the increase in head-twitch response, suggested that depletions of less than 40% did not affect this behaviour. However, depletions greater than 40% produced marked increases in this response and there was a good correlation between these two variables (r=0.701, P less than 0.01). In conclusion, these data suggest that enhanced head-twitch responses occurring after lesioning with 5,7-DHT may result from increased 5-HT2 receptors in brain. Moreover, the magnitude of the observed behavioural enhancement was dependent upon the extent of depletion of 5-HT produced by the lesioning.

Changes in platelet alpha 2-adrenoceptor binding post partum: possible relation to maternity blues.

Platelet alpha 2-adrenoceptor binding characteristics were investigated in 28 healthy women during the peripartum period. The number of platelet alpha 2-adrenoceptors fell after childbirth, corresponding to a fall in the circulating levels of oestrogen and progesterone. Two-thirds of the women had an episode of maternity blues. At 7-10 days post partum the platelet alpha 2-adrenoceptor capacity in these subjects was significantly higher than in those women who did not show signs of maternity blues and significantly higher than a control group of normally menstruating women. We conclude that platelet alpha 2-adrenoceptors may be affected by the circulating levels of endogenous oestrogen and progesterone and that a delayed or diminished fall in platelet alpha 2-adrenoceptor capacity after childbirth may be associated with the development of maternity blues.

[Age dependence of beta-lipoproteins in comparison with cholesterol and triglycerides in the serum of SPF rats].

Age-dependency of beta-Lipoproteins as Compared to Cholesterol and Triglycerides in the Serum of SPF Rats Total cholesterol and triglyceride values increase with age in rats (male greater than female). Correlated to these lipid increases, an increase of serum turbidity and of total beta-lipoproteins (pre-beta- and beta) can also be observed. The results of gel-electrophoretic separation show far greater pre-beta- and beta-lipoprotein increases than alpha-lipoprotein increases in aging animals. It can therefore be concluded that the relative decrease in alpha-lipoproteins is primarily caused by a more rapid increase of pre-beta-and beta-lipoproteins.

[Behavior of total serum cholesterol in aging rats in relation to food intake, weight development and mortality rate].

Rats (OFA - Sandoz) were kept under barrier conditions their whole lives in order to study the correlations between food intake, body weight, serum cholesterol and mortality rate. The cholesterol levels in males in the first life-half rose in correlation to food intake and body weight increase. In the second half, however, the cholesterol level increased again, whereas body weight and food intake decreased. The mortality rate rose, which was correlated to the second increase of the cholesterol level and the body weight loss (probit analysis). These findings were not made in females, which had lower food intake, body weight and cholesterol levels.

Influence of 4'-6'-diamidino-2-phenylindole on the secondary structure and template activities of DNA and polydeoxynucleotides.

The interaction between 4'-6-Diamidino-2-Phenylindole-hydrochloride (DAPI) and a variety of DNAs and synthetic polydeoxynucleotides was investigated in order to delineate the nucleic acid structural features necessary for binding. The spectra of DAPI-DNA complexes, measured at various DAPI:DNA molar ratios (r), are hypochromic relative to DNA in the region of its maximum absorption. All the curves pass through an isosbestic point at 268 nm. A new maxima appears in the region of 380-392 nm for DAPI-DNA COMplexes. The magnitude of the peaks in the region are directly proportional to the amount of drug present in the complex. Studies with various DNA types and synthetic polydeoxynucleotides indicate that the drug preferentially binds to dAT-rich regions of DNA. This was also confirmed by enzymatic studies. The inhibition of template action by DAPI in a purified DNA-polymerase reaction was dependent on the dAT-content of the template. The implication of these data to explain a selective binding of DAPI to mitochondrial DNA have been discussed.

The effects of the beta-adrenoceptor blocking agent LL 21-945 on renal hypertensive (Grollman) rats.

Prophylactic administration of LL 21-945 [4-(3-tert. butyl-amino-2-pivaloyloxypropoxy)-9-fluorenone] during 15 weeks to Grollman rats depressed the development of hypertension, tachycardia and myocardial pathogical changes. The tachycardia was eliminated and the degree of the myocardial pathological changes and coronary sclerosis was less severe than in the untreated Grollman rats. The survival rate was slightly improved with LL 21-945. With respect to the biochemical parameters studied, only a tendency of LL 21-945 to moderate the rises in blood cholesterol, total glycerides and urea was observed towards the end of the experiment.

[A comparative study of total protein and albumin in man, monkey, dog and rat employing different analytical methods (author's transl)].

In the present study, serum albumin was determined colorimetrically in three animal species and in man with the help of four currently used dye-reagents (protein binding reagents). The results were compared with those obtained from corresponding electrophoretic and biuret determinations, using Versatol as a standard-control throughout. Using 2-(4-hydroxyazobenzene)-benzoic acid and bromocresol purple as reagent for the albumin determinations, species-specific differences from the electrophoretic results were found. No such differences occurred with bromo-cresol green, except in rats. There was no significant difference in albumin concentrations between man, monkeys and rats. Total protein concentration was only similar in man and monkeys.
