Morphometric study of nucleolar organizer regions in human oral normal mucosa, papilloma and squamous cell carcinoma.

A morphometric study of nucleolar organizer regions (NOR) was performed to analyze their distribution, volume, number and shape in the different strata of human normal oral mucosa epithelium and papilloma and in squamous cell carcinoma employing microphotographs of silver-stained paraffin sections. The different NOR-related parameters evidenced significant differences between normal mucosa, papilloma and squamous cell carcinoma. The functional polarity of normal mucosa epithelium and of papilloma is also evidenced in terms of NOR-related parameters. The discriminative value of certain NOR parameters was demonstrated.

Inclusion bodies containing adenovirus-like particles in the intestine of a psittacine bird affected by inclusion body hepatitis.

This paper reports a case of inclusion body hepatitis with intranuclear inclusion bodies in the liver and the intestine of a Yellow-naped Amazon parrot (Amazona ochrocephala). Structurally, basophilic intranuclear inclusion bodies were found in hepatic cells and enterocytes. Ultrastructurally, icosahedral adenovirus-like particles, 60-75 nm in diameter, were found in the same cells.

Acyclovir in the treatment of laryngeal papillomatosis.

We report the results obtained in a study of 3 young patients with laryngeal papillomatosis, who were treated with acyclovir after tumor excision using forceps was performed under microlaryngoscopy. No recurrence of the papillomatosis occurred between 18 and 42 months post-therapy. Laryngeal papillomatosis is the most common proliferative laryngeal lesion in children. Different non-surgical therapies have been used to prevent its recurrence but with poor results. Acyclovir is a drug with proved efficacy in DNA viral diseases. Since papillomata of the larynx is most likely a viral disease which is imputed to have a DNA virus etiology, acyclovir may be effective to control it.

Probucol enhances cholesterol efflux from cultured human skin fibroblasts.

To explore whether the reported xanthoma-reducing effects of probucol are related to tissue-specific mechanisms of the drug, the influence of probucol on cholesterol efflux from cultured human skin fibroblasts was investigated. Incubation of cells with probucol led to a 2-fold enhancement of high-density lipoprotein-mediated cholesterol efflux. This result raises the possibility that probucol may reduce cholesterol accumulation in tissues through a cholesterol-mobilizing action.

Lipid and lipoprotein levels in undialyzed patients with chronic renal failure.

Lipid and lipoprotein levels were measured in undialyzed uremic men and compared with levels of healthy control subjects. The patients had lower high-density lipoprotein (HDL) cholesterol (mean +/- SD, 46.3 +/- 14.8 vs 54.1 +/- 18.2 mg/dl) and increased serum triglycerides (197 +/- 151 vs 142 +/- 87.7 mg/dl). When compared with patients maintained on dialysis, the undialyzed patients had higher HDL cholesterol (46.3 +/- 14.8 vs 30.3 +/- 13.3 mg/dl), serum cholesterol (226 +/- 81 vs 151 +/- 42 mg/dl), and triglycerides (197 +/- 151 vs 146 +/- 105 mg/dl). The low-density lipoprotein (LDL): high-density lipoprotein cholesterol ratios were 3.78, 3.24, and 2.84 in the dialyzed, undialyzed, and control groups, respectively. Lecithin: cholesterol acyl transferase activity of undialyzed patients was similar to controls when measured by an endogenous substrate assay (148 +/- 112 vs 112 +/- 28 nmol/ml/hr), but were significantly lower when assayed using exogenous substrate (73.3 +/- 25.6 vs 112.8 +/- 7 nmol/ml/hr). The rate of cholesterol transfer (RCT) from HDL to very low density lipoprotein and LDL was in the following order: dialyzed patients (1.85 +/- 1.29 mg/dl/hr) less than undialyzed patients (2.41 +/- 1.57 mg/dl/hr) less than control subjects (4.83 +/- 3.13 mg/dl/hr). Within the undialyzed patient group, black patients had lower serum cholesterol and triglycerides and higher HDL and HDL3 cholesterol than white patients. Apoprotein A-I levels were higher and apoprotein B levels were lower in the black patients. There was a strong correlation (r = 0.9501) between apoprotein A-I and HDL levels in the black patients, but not in the white patients (r = -0.148).(ABSTRACT TRUNCATED AT 250 WORDS)

Rat liver pyruvate carboxylase. Purification, detection and quantification of apo and holo forms by immuno-blotting and by an enzyme-linked immunosorbent assay.

A simple scheme for the purification of pyruvate carboxylase from rat liver mitochondria is described. It is rapid and provides high-purity pyruvate carboxylase with excellent yield and reproducibility. The final enzyme preparations appear to be homogeneous by the following criteria: elution behaviour on molecular-sizing matrix, SDS/polyacrylamide-gel electrophoresis, Ouchterlony double-diffusion analysis and Western blotting. Detection and quantification of nanogram amounts of pyruvate carboxylase (apo and holo forms) in total tissue homogenates by immuno-blotting and by enzyme-linked immunosorbent assay are described. The data provided suggest that under normal physiological conditions (both in vivo and in vitro) essentially all the pyruvate carboxylase molecules are biotinylated.

Verbal behaviors and patterns exhibited by physicians during genetic counseling sessions.

Research in interaction and interviewing behaviors usually has focused on the verbal behaviors exhibited by the interactants. These behaviors have been looked upon and analyzed in isolation, that is, as a single event. There is little or no information on physician-patient interactions that analyzes verbal behaviors as being a series of events related to each other. These behaviors, when seen as a chain of events, provide a different and revealing picture of those who exhibit them. A study was undertaken to gather information on physicians' verbal behaviors and verbal patterns during interactions involving genetic counseling. Physicians who were rated high in communication skills spent more time in the affective dimension of the interaction and exhibited more reflection of feelings, emotional probing, reassurance/support, and silence during the interaction with patients. These differences were found in both the sequential verbal pattern data and the individual verbal behavior data.

Enzyme replacement therapy in porphyrias--IV. First successful human clinical trial of delta-aminolevulinate dehydratase-loaded erythrocyte ghosts.

A patient with chronic lead intoxication was treated with only one course of highly purified human blood aminolaevulinate dehydratase entrapped in autologous erythrocyte ghosts given intravenously. No untoward effects were observed during or after infusion. An immediate increase in the patient's erythrocyte dehydratase activity was detected 1 hr after enzyme administration, reaching its maximum and nearly normal level 2 days later, values remained unchanged for a week, to slowly diminish after 2 weeks of initiated the treatment, and finally recovered activity was kept practically leveled off for weeks. This novel therapeutic trial produced complete improvement both clinical and biochemical, showing that enzyme infusion has been beneficial and can be safely and successfully used in the treatment of human lead intoxication.
