Factors influencing indomethacin toxicity in the rat.

The influence of various experimental conditions, such as nature of the diet, feeding schedule, sex and castration on indomethacin toxicity in the rat has been determined. In general, the following conclusions can be drawn: a) Independently of feeding schedule, sex or castration, indomethacin is significantly more toxic in animals on regular (RD) than in those on fat-free diet (FFD). b) Regardless of sex or castration, fasting significantly reduces indomethacin toxicity in RD fed animals. c) With the exception of RD unfasted animals, indomethacin is significantly more toxic in males than in females. d) Incidence and degree of intestinal ulceration are not sex dependent. e) Independently of the kind of diet and of the feeding schedule, castration reduces indomethacin toxicity in males but does not modify it in females. These findings are discussed.

Estrogen-progestogen interaction: factors influencing progesterone-induced endometrial changes of the uterus of the immature rabbit.

In orally estrogen-primed animals, four times as much progesterone is required to bring about secretory changes comparable to those obtained following subcutaneous priming. These findings might be interpreted as evidence that: (a) oral estrogen-induced proliferative changes are either different or inadequate, and (b) excess progesterone is conditio sine qua non for the induction of those proliferative changes susceptible of being transformed into secretory ones.

Influence of side chain configuration on anti-inflammatory analgesic and anti-pyretic properties of 4-biphenylyl alkanoic acids.

A study on the influence of the number of carbon atoms in the side chain as well as side chain configuration on some pharmacologic properties of 4-biphenylyl alkanoic acids is presented. Unlike the chemical structure dependent anti-inflammatory properties, mild analgesic and anti-pyretic properties were neither dependent upon number of carbon atoms nor side chain configuration.
