Carcinogenic risk assessment may be based on epidemiological as well as experimental data; the aim of the present study is to compare these two procedures and to verify the efficiency of some animal bioassays for the prediction of tumor incidence and relative risk in man. The predictability obtained from animal models has been found to be fairly consistent with the data obtained from epidemiological studies.
A quantitative EEG analysis was performed on the posterior lead of 18 patients with their eyes closed 3-10 days after minor head injury. There was a significant increase of the mean power of slow alpha (8-10 c/sec), a reduction of fast alpha (10.5-13.5 c/sec), with a shift of mean alpha frequency towards lower values, and a reduction of fast beta (20.5-36 c/sec) in the patients with head injuries compared with the age- and sex-matched patients in the control group. Therefore, power spectral EEG analysis performed with the above-mentioned criteria may be suggested as a sensitive tool to be added to others during the neurophysiological follow-up studies of people with head injuries.

Single oral doses of aluminium hydroxide (50 to 200 mg/kg) were found to induce in mice a dose-dependent diminution of the power of the 7.5 to 12 Hz frequency band, with a parallel dose-dependent increase of aluminium content in the brain, as early as 45 min after administration, and indicated that aluminium hydroxide is readily absorbed through an empty stomach or duodenum and is able to induce alterations of background EEG rhythms at doses equivalent to the ones used in human therapy. These data suggest that the EEG disturbances of the background type, (which are observed during the early stage of dialysis encephalopathy in man), may be partly due to a pharmacological and therefore reversible effect induced by an increase in aluminium level in the brain.

Clinical EEG and biochemical data were recorded in 10 children with classical PKU and 5 with variant forms of hyperphenylalaninaemias during the first year of life. A semiquantitative evaluation of the EEG showed a high correlation between epileptiform abnormalities and phenylalanine blood levels in the first 90 days of life and therefore with the delay before dietary therapy in PKU children. Although performed on a limited population, such an approach may indicate an additional non-computerized EEG tool for the clinical management of hyperphenylalaninaemias, and suggest some criteria for neurophysiological risk evaluation during the first year of life.

The effects of flicker photic stimulation (6, 8, 9, 10, 11, 12, 14, 16 flashes/sec) and of white noise (80 dB SPL) on the EEG of man were investigated with the aid of power spectral analysis. In order to evaluate both the alpha attenuation response (AAR) and the 'driving' reaction induced by flicker as well as the effect of noise, the analysis was carried out on broad (delta, theta, alpha, beta) and on narrow (bandwidth: 1 Hz) spectral bands. A significant correlation between the amount of AAR induced by noise and a measure of the tendency to exhibit photic driving was found, though there was no clear-cut interaction between the photic stimuli and noise when delivered stimultaneously.
The effects of some ergot derivatives on the electroencephalogram and behavior of laboratory animals (mice, rats, rabbits) have been surveyed. The interference of bromocriptine on the dopaminergic central systems has been studied using behavioral (interference with the effects induced by L-dopa, 5HT, TRF or neuroleptic drugs) and electrophysiological (EEG) methods. While no clear effects of bromocriptine could be evidenced in mice, EEG recordings from the dorsal hippocampus of the rabbit showed a slight disruption of epsilon-waves after administration of 2 mg/kg i.v. of the drug or more.
The behavioral effects (tremors, tail erection, increased motility) of intracerebrally injected TRF are enhanced by pretreatment with chlorpromazine, reserpine and sulpiride; haloperidol does not exert appreciable effects. L-Dopa attenuates or abolishes the potentiation.
The effects of delta6-THC on the frequency spectrum of the hippocampal EEG were examined with the assistance of an electronic integrator. Six rabbits bearing chronically implanted cortical and hippocampal electrodes were dosed intravenously with delta6-THC (2.5 percent solution in polyethylene glycol 300; doses of 10, 50, 100, 250 and 1000 mug/kg) once a week. A comparison of epochs of arousal in the pre-drug control and post-drug periods demonstrated that a dose as low as 50 mug/kg produced a consistent, statistically significant diminution in "power" of the 7,8,9 Hz frequencies of the hippocampal EEG, with 8 Hz the most sensitive. Doses above 250 mug/kg produced an overall decrease in the "power" spectrum from 7 to 22 Hz. Spectral analysis of the EEG offers a method which is sufficiently sensitive to reveal a central effect of detla6-THC in rabbits at doses equivalent to the human threshold for psychic effects.

Brain-stem auditory evoked potentials (BAEPs) and flash visual evoked potentials (F-VEPs) were gathered from 8 early treated phenylketonuric (PKU) children in a prospective longitudinal investigation during the 1st to the 12th months after birth. No consistent differences were found in the wave morphology of evoked potentials in PKU children from that of age-matched controls. Studying the latency of some components showed that in BAEPs, wave I latency was similar to control values for the whole year, but that the I-V interpeak mean latency (I-V IPL) was always significantly longer than in controls. In F-VEPs wave N1 latency was significantly longer than in controls only at 1-2 months of age, but returned to control values at 3-4 months (when all children were on dietary therapy) and remained in this range up to the 12th month. The mean latency of the P2 wave of flash VEPs was always significantly longer in PKU children than in controls. These results show that relevant alterations in evoked potentials may be found in PKU children several months after starting dietary therapy. This suggests that information processing in the brain may be impaired for a long time, due to abnormal metabolic conditions between birth and the onset of dietary therapy.
