An experimental model of human leukemic meningitis in the nude rat.

An experimental animal model of meningeal leukemia was developed in the nude rat, rnu/rnu, using the human-derived acute lymphoblastic leukemia cell line HPB-ALL. Anesthetized rats were placed in a modified stereotaxic frame and then injected intrathecally, at the level of the cisterna magna, with human leukemic cells. Cerebrospinal fluid and tissue samples from brain, spinal cord, heart, liver, kidney, spleen, bone marrow, and cervical lymph nodes were subjected to histopathologic examination and molecular genetic screening by clonotype primer-directed polymerase chain reaction (CPD-PCR). Ninety-three percent of animals (n = 14) developed signs of meningeal irritation leading to death 30 to 63 days postinjection (median, 36.0 days, mean, 38.7); death occurred between 30 and 39 days in 77% of all animals. Leukemic cells progressively infiltrated the pericerebellar and pericerebral subarachnoid space and infiltrated the Virchow-Robin (perivascular) space. The infiltrating meningeal leukemia closely resembled the pathologic presentation in the human condition. By CPD-PCR, leukemic cells were first detected in cerebrospinal fluid (CSF) on day 4 postinjection, were variably present over the ensuing 17 days, and were consistently detected after day 21. At terminal stages, CPD-PCR tissue surveys showed leukemic DNA in all brains and spinal cords and rarely in cervical lymph nodes, but leukemic DNA was not detected in any other tissue screened. Leukemic meningitis was reliably produced with a predictable survival time. Intrathecal administration of leukemic cells was an efficient means of transmitting leukemic meningitis and it compartmentalized the disease to the central nervous system (CNS), eliminating potential complications of systemic illness. The use of human-derived cell lines may render this model more relevant to the development of future therapeutic strategies to treat leukemia and lymphoma that invade the CNS.

Parental representation of children during the first year of life: a longitudinal comparison of mothers' and fathers' responses on Semantic Differential Scales.

The aim of this paper was to investigate parental representation over time using a specially devised Semantic Differential Scale, developed in the context of longitudinal research, carried out during children's first two years of life. 42 parent-couples, during their first experience of parenthood were asked separately to rate the concept of "my child" in specific periods of their child's first year of life. The analysis highlighted the complexity of parental representation of babies.

Pleiotropic expression of heterologous cytokine/receptor genes in HTLV-1 associated diseases: candidate TRS for chimeric gene therapy.

DNA motifs that encode for specific transcriptional regulatory sequences (TRS) when engineered adjacent to the structural protein coding domain of a suicide enzyme can provide cell-lineage specific protein expression. The disparate up-regulation of several genes in adult T-cell leukemia (ATL) versus HTLV-1 associated myelopathy/tropical spastic paraparesis (HAM/TSP), seropositive carriers (SPC) and uninfected normals may reflect events at the molecular level related to leukemogenesis or to processes maintaining the heme-oncologic phenotype. Further, the genetic transduction of cytokine and receptor genes uniquely associated with ATL may provide targets for the development of leukemia-specific gene therapies aimed at exploiting differences in the production of certain growth factors and growth factor receptors. Comparisons of the transcriptional and translational levels of interleukin-2 receptor alpha chain (IL-2R alpha), transforming growth factor-beta 1 (TGF-beta 1) and intracellular adhesion molecule-1 (ICAM-1) in ATL, HAM/TSP, and SPC and in several control populations revealed selectively up-regulated expression in ATL. We evaluated the feasibility of using lymphoid-specific TRS to activate herpes simplex virus thymidine kinase (HSVtk) to achieve selective cytotoxicity in leukemias expressing terminal deoxynucleotidyl transferase (TdT). Selective and efficient leukemic cell killing was produced and suggests that similar chimeric gene constructs containing TRS elements for IL-2R alpha, TGF-beta 1, or ICAM-1 may prove useful in designing gene therapies to treat ATL.

[The effect of cryoapplication time using liquid nitrogen for effectiveness of basocellular epithelioma treatment].

Cryosurgery is well suited to treat skin tumors. A cure rate of high percentage is achieved paying attention to freeze time. The course of treatment of 217 basaliomas was analysed. The duration of follow-up after treatment was from 6 to 14 months. 147 lesions were treated with one-cycle cryosurgery. In 64 (45.5%) cases cure was achieved after the first cycle of treatment and in 83 (56.5%) cases the second cycle brought complete healing. 70 basaliomas treated with double freeze-thaw cycle were cured in 100% of cases.

In vitro assessment of neurotrophic activity from the striatum of aging rats.

Neurotrophic factors are produced in the striatum following trauma and have a demonstrable effect on in vitro bioassays and on in vivo graft survival. We have previously measured the in vitro effect of these factors following trauma to the striatum of young rats. However, the effect of age on this neurotrophic response has not been evaluated. In this study we report on the in vitro effects of extracts (obtained from gelfoam) removed from striatal cavities 7 days following trauma. Gelfoam extract from aged rats (18-24 months) had a reduced neurite-promoting response in dorsal root ganglia (DRG) and SH-SY5Y (a dopamine-producing neuroblastoma cell line) assays, compared to gelfoam from young rats (2-3 months). In contrast, extracts from both young and old rats showed significant neuroprotection of SH-SY5Y cells from the dopaminergic neurotoxins N-methy-4phenylpyridinium ion (MPP +) and 6-hydroxydopamine (6-OHDA). The results suggest that the striatum of aged individuals may have (1) a diminished capacity of neurite promotion and/ or (2) that neurite outgrowth and neuroprotection may be influenced by different factors or different levels of the same factors. The direct implication is that aged animals would be the most appropriate models to study experimental therapies for Parkinson's disease.

Traumatized rat striatum produces neurite-promoting and neurotrophic activities in vitro.

We have previously reported that ciliary neurotrophic factor (CNTF) mRNA is upregulated in the rat striatum following trauma and that its peak is coincident with a peak in the number of GFAP-positive astrocytes. CNTF, or other neurotrophic factors present in the traumatized striatum, may be involved in the dopaminergic fiber sprouting seen following cavitation or graft implantation in animal models of Parkinson's disease. This study was undertaken in order to further characterize the neurotrophic activity present following trauma through the use of bioassays. Adult rats underwent stereotaxic biopsy of the right striatum, and gelatin sponge [gelfoam (GF)] was placed in the resultant cavity. GF was collected from 1 to 30 days following trauma and homogenized. GF extracts (with equal protein concentrations) were assayed using dorsal root ganglion (DRG) explants, dissociated ciliary ganglia (CG), and human dopaminergic neuroblastoma cell (SH-SY5Y) cultures. The GF extracts had significant neurite-promoting activity (NPA) for DRG, CG, and SH-SY5Y cells, with the maximum effect seen 7 days after trauma. NPA was not blocked by anti-nerve growth factor (NGF) Ab, but anti-brain-derived neurotrophic factor (BDNF) Ab significantly blocked the activity for DRG. The GF extracts protected the SH-SY5Y cells from the neurotoxins 6-OHDA and MPP+, as did NGF and BDNF. This neuroprotective effect of GF was not blocked by anti-NGF Ab. This study suggests that the neurotrophic activity in GF extracts has CNTF-like and BDNF-like components as well as another, undefined component.

Decrease in milk and blood dioxin levels over two years in a mother nursing twins: estimates of decreased maternal and increased infant dioxin body burden from nursing.

This study addresses the issue of breast-feeding and its reduction of maternal dioxin body burden. Nursing is also a source of infant dioxin exposure. This study extends our previous efforts to investigate a nursing mother's milk and blood dioxin levels. We report polychlorinated dibenzo-p-dioxin (PCDD) and polychlorinated dibenzofuran (PCDF) dioxin toxic equivalents (TEQs) in milk (M) and blood (B) both before and also after two years of nursing twins to be 16.9 ppt (M), 14.9 ppt (B), and 3.1 ppt (M) and 4.9 ppt (B), respectively. The ratios of measured congeners comparing milk to whole blood from a nursing mother taken initially and after two years of nursing vary from 0.36 to 8.40 in 1992 and 0.17 to 1.0 in 1994. The mother's body burden was initially calculated to be 329 ng TEQ from milk levels and 291 ng TEQ from blood levels using samples taken in February 1992 and decreased to 60.1 ng TEQ from milk and 96 ng TEQ from measured blood using samples collected in December 1994. We calculate that the excretion of dioxin TEQ by the mother through breast-feeding is 269 ng TEQ, which is similar to the 303 ng TEQ estimated total dioxin intake by the twins over two years. The average daily dioxin intake from nursing is 66 pg TEQ/kg-BW/day for each twin over the two years.

Concentrations of polychlorinated dibenzo-p-dioxins and dibenzofurans in human placental and fetal tissues from the U.S. and in placentas from Yu-Cheng exposed mothers.

We report the concentrations of polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) found in human placentas and fetal tissue from the U.S. and in placentas from Yu-Cheng mothers. In placental material from sources in the U.S. we found concentrations, expressed as dioxin toxic equivalence (TEQ) values and on a lipid basis, between 8.4 and 17.6 ng/kg. In a pooled sample of fetal tissue (8-14 week) the concentration, 5.3 ng/kg, was somewhat smaller. In placental material from Taiwanese Yu-Cheng subjects much higher concentrations were found, in the range 610 to 9010 TEQ ng/kg, again on a lipid basis.

[Etiology of pleural exudates from personal material].

Etiology of pleural exudates was established in 87 (79%) out of 110 men, in age 18-82 years, mean-51 years. In this group of patients tuberculosis was recognized in 19.1%, neoplastic disorders in 25.4%, infectious-nontuberculous disease in 34.5%. Etiology of pleural exudates in 20.9% of patients was unknown. However, 80% of them improved in the course of antituberculous treatment.

Dioxin concentrations in the blood of workers at municipal waste incinerators.

OBJECTIVES: Increased concentrations of polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzo-furans (PCDFs) in pooled blood samples from workers at municipal waste incinerators have been reported. This study was undertaken to confirm these results in individual blood samples from potentially exposed and unexposed workers at municipal waste incinerators compared with matched unexposed controls and compared with concentrations in the slag and fly ash from the municipal waste incinerators. METHODS: Concentrations of PCDDs and PCDFs were determined in the blood of 10 workers from an old municipal waste incinerator without adequate pollution controls, 11 workers from a newer incinerator with modern pollution controls, and 25 controls from the general population group matched for age (+/- 10 years), sex, and race, and in the slag and fly ash from the older incinerator. RESULTS: Significant increases of certain PCDDs and PCDFs were found in the blood of the workers from the older incinerator compared with the controls as follows: octaCDD (1051 (438) v 637 (344), P < 0.001), hexaCDF (52.3 (28.7) v 30.2 (18.2), P < 0.01), heptaCDF (43.9 (30.4) v 22.7 (12.4), P < 0.001), total PCDDs (1262 (484) v 825 (454), P < 0.001), total PCDFs (133.0 (68.1) v 93.7 (36.7), P < 0.05), and total PCDD/Fs (1395 (537) v 918 (437), P < 0.001). The workers from the older incinerator with the greatest exposure were found to have the most significant increases of the blood PCDDs and PCDFs, and the pattern of increased PCDD and PCDF congeners in the blood corresponded to the pattern in the incinerator slag and ash. No significant differences were found between the blood concentrations of the workers at the newer incinerator and the controls. CONCLUSION: Occupational exposure to slag and fly ash from municipal waste incinerators may increase the blood concentrations of PCDDs and PCDFs. Modern pollution control technology in new incinerators may be able to minimise potential exposure to slag and fly ash and thus the absorption of PCDDs and PCDFs from this source.

Congener-specific levels of dioxins and dibenzofurans in U.S. food and estimated daily dioxin toxic equivalent intake.

Food, especially meat, milk, and fish, is the immediate source of almost all polychlorinated dibenzo-p-dioxins (PCDDs), dibenzofurans (PCDFs), and dioxinlike compounds in the general population. To estimate intake of these highly toxic compounds, we performed congener-specific dioxin analyses for the first time on U.S. food for 18 dairy meat, and fish samples from a supermarket in upstate New York. 2,3,7,8 Tetrachlorodibenzo-p-dioxin (TCDD, "dioxin") toxic equivalents (TEqs) on a wet weight basis for the dairy products ranged for 0.04 to 0.7 ppt, meat TEqs ranged from 0.03 to 1.5 ppt, and fish TEqs ranged from 0.02 to 0.13 ppt. Previous human breast milk and infant formula analyses were used with the current preliminary food data to estimate a range of dioxin intake for Americans. Average daily food intake of TEqs for an adult weighing 65 kg was estimated to be between 0.3 and 3.0 pg/kg body weight, for a total of 18-192 pg TEq, using 1986 American consumption rates. Due to the relatively high level of PCDDs and PCDFs commonly found in human breast milk from American women and from women in other industrial countries, a nursing infant may consume an average of 35-53 pg TEq/kg body weight/day in its first year of life. This may be compared with the current U.S. EPA virtually safe dose of 0.006 pg TCDD/kg body weight per day over a 70-year lifetime based on an upper limit cancer risk of 10(-6), or the 10 pg/kg/day used by some European government agencies.

Study of Longeot's test of formal operational thinking in a group of Italian adolescents.

This paper describes a study of development of formal logical thought in 230 Italian adolescents by applying Longeot's Echelle de la Pensée Logique (Scale of Logical Thought). For age groups 11 to 13 and 14 to 18 years, change from preformal to formal thought and acquisition of such thought could be influenced by experience, sex, social-cultural level. The Echelle de la Pensée Logique should produce some special aspects to be compared with other classical intelligence tests.

Unilateral spatial neglect and recovery from hemiplegia: a follow-up study.

A follow-up study was undertaken in order to investigate the outcome of recovery from right and left hemiplegia on simple motor function and activities of daily living. The role of concomitant neurophysiological deficits was also investigated. The main results indicate that after six months from onset, left hemiplegics show a lesser degree of improvement in independence and social adjustment coupled with a tendency to a poorer recovery of motor function than the corresponding group of right hemiplegics. Unilateral spatial neglect, which is more frequent and severe in the group of left hemiplegics, seems to be crucial in hampering their performance.

Dioxins in U.S. food and estimated daily intake.

Congener-specific analyses for polychlorinated dioxins and dibenzofurans were performed on eighteen dairy, meat, and fish samples obtained from a supermarket in upstate New York. Dioxin toxic equivalents (TEqs) on a whole or wet weight basis for these food samples ranged from 0.02 to 1.5 parts per trillion (ppt). Data on American food consumption rates were used to estimate a U.S. range of dioxin intake from food. Based on these first congener-specific dioxin analyses of U.S. food, U.S. average daily intake of "International" dioxin toxic equivalents for an adult weighing 65 kilograms (kg) was estimated to be between 18 to 192 picograms TEq or 0.3 to 3.0 picograms per kilogram of body weight. Our previous results for cow's milk, human breast milk, and soy-based infant formula were used to calculate intake for infants. The relatively high level of dioxins commonly reported in human breast milk from America contributes from 35 to 53 picograms of dioxin toxic equivalents per kilogram of body weight to the nursing infant per day in its first year of life. This value greatly exceeds the U.S. Environmental Protection Agency estimated dose which would lead to one excess cancer per one million people of 0.006 pg of TCDD per kg of body weight per day over a seventy year lifetime or similar reference doses used by some other countries of up to 10/pg/kg/day over a 70 year lifetime.
