The mean transit time (MTT), coefficient of variation (CoV), and index of skewness (IoS) of transit times of the forced expiratory flow/time curve have been compared with maximal mid-expiratory flow (MMEF), flow at 75, 50, and 25% of vital capacity (V75, V50, and V25), and the ratio of forced expiratory volume in 1 second (FEV1) to vital capacity (VC) in 51 asthmatic children judged on clinical grounds to be in remission. In 19 children all eight indices were normal. Of the remaining 32, MTT was normal in 5, V50 in 11, V75 in 12, and FEV1/VC and MMEF in 13 each. MTT was significantly more sensitive in detecting abnormality when other indices were normal, and abnormalities in MTT were of significantly greater magnitude when present. In 12 patients with at least one abnormal index given salbutamol aerosol, MTT was not more sensitive than MMEF, V25, FEV1/VC, or V50 in detecting change.
Vital capacity and forced expiratory volume in 1 s were measured in 387 healthy European and 16 non-European children aged from five to 11 years. These measurements were correlated with height, sitting height, arm span, weight, lean weight and age in various combinations. Suitable linear regression equations are presented from which these lung volumes may be predicted with confidence limits down to approximately +/- 400 ml and +/- 200 ml respectively.
The alpha- and the beta-tubulin genes of the hypotrichous ciliate Euplotes octocarinatus were isolated from a size-selected macronuclear DNA library. The alpha-tubulin gene is located on a 1,587 bp macronuclear DNA molecule and the beta-tubulin gene on a 1,524 bp macronuclear DNA molecule. Sequencing revealed that all the cysteine residues of the two genes are encoded by the common cysteine codons UGU and UGC and none by an UGA codon. This is in contrast to the genes of E. octocarinatus sequenced so far, where some of the cysteines are encoded by the opal codon UGA. The tubulin genes end like other Euplotes genes with a TAA. They do not contain introns. The last codon for an amino acid in the alpha-tubulin gene is a GAA which codes for glutamic acid. This is in contrast to what has been reported for most alpha-tubulin genes, but it supports for other hypotrichous ciliates. No evidence for the existence of more than one type of alpha- and one type of beta-tubulin genes could be obtained.
Three macronuclear genes encoding putative nuclear protein kinases of the ciliate Euplotes octocarinatus syngen 1 were isolated and sequenced. All three deduced gene products share significant properties with a group of recently identified nuclear serine/threonine protein kinases named Ndr. The three predicted proteins contain the twelve conserved catalytic subdomains of protein kinases and 22 near universally-conserved amino acids residues that are characteristic of serine/threonine protein kinases. In addition, there is an approximately 30 amino acid-peptide insertion between subdomains VII and VIII that contains a potential nuclear localization signal. Sequence analysis suggests that expression of the Eondr2 gene requires a + 1 programmed translational frameshift for its translation. Comparison of the deduced EoNdr2 with other known Ndr protein kinases implies that a + 1 ribosomal frameshift occurs at the motif AAATAA.
THE ASSESSMENT PROTOCOL for EXCELLENCE in PUBLIC HEALTH process was carried out in the state of Washington to assess local health department capacity and to identify their self-perceived strengths and weaknesses. Staff from 24 of the 32 local health departments in Washington completed organizational capacity assessments. Fifty percent or more of the health departments identified the following eight indicators as strengths: legal authority, public policy and implementation, budget development, financial reporting and administration, audit, financial documentation, organization and structure of program management, and policy board procedures. Seven indicators were identified as weaknesses by 50% or more of the respondents: legal counsel, mission and role, data collection and analysis, planning and development, evaluation and assurance of community health assessment, community health assessment and planning, and community health policy. The results of the assessment highlight the traditional organizational and service delivery strengths of the local health departments and point out weaknesses in their ability to assess community health and to develop communitywide health policy.
