Association between atopy and variants of the beta subunit of the high-affinity immunoglobulin E receptor.

The beta-subunit of the high-affinity IgE receptor (Fc epsilon RI-beta) on chromosome 11 is maternally linked to atopy, the state of enhanced IgE responsiveness underlying allergic asthma and rhinitis. We have identified a common variant of Fc epsilon RI-beta, lle181Leu within the 4th transmembrane domain. Leu181 shows significant association with positive IgE responses in a random patient sample. Amongst 60 unrelated nuclear families with allergic asthmatic probands, Leu181 is identified in 10 (17%), is maternally inherited in each, and shows a strong association with atopy. Our data indicate that Fc epsilon RI-beta, subject to maternal modification, may be the atopy-causing locus on chromosome 11q.

Inheritance of Strain Instability (Sectoring) in the Commercial Button Mushroom, Agaricus bisporus.

The button mushroom, Agaricus bisporus, is a commercially important cultivated filamentous fungus. During the last decade, the button mushroom industry has depended mainly on two strains (or derivatives of these two strains). Using one of these highly successful strains (strain U1) we examined the phenomenon of strain instability, specifically, the production of irreversible sectors. Three "stromatal" and three "fluffy" sectors were compared with a healthy type U1 strain and with a wild-collected isolate. Compost colonization and fruit body morphology were examined. The main objective of this study, however, was to examine the meiotic stability of the sectored phenotype. Single basidiospores were isolated and subjected to a grain bioassay in which the ability to produce sectors was measured. Our results were as follows: (i) basidiospore cultures obtained from a wild-collected isolate showed no tendency to produce sectors; (ii) approximately 5% of the basidiospore cultures obtained from healthy type U1 strains produced irreversible sectors in the grain bioassay; (iii) the five primary sectors examined produced basidiospore cultures, half of which produced normal-looking growth in the grain bioassay and half of which produced some degree of sectoring; and (iv) the one sectored isolate that represented the F2 generation gave ratios similar to the 1:1 ratio observed for the F1 cultures.

Retrotrapezoid nucleus glutamate injections: long-term stimulation of phrenic activity.

In chloralose-urethan anesthetized, paralyzed, vagotomized, glomectomized, and servo-ventilated cats we examined the effects of 10 nl of glutamate (10 mM, 100 mM, and 1 M) injected unilaterally over 60 s into the region of the retrotrapezoid nucleus (RTN). Seven 10 mM glutamate injections produced no consistent effects on the amplitude of the integrated phrenic nerve signal, respiratory cycle duration, or blood pressure. Ten 100 mM injections consistently increased integrated phrenic amplitude significantly from a baseline average of 31 +/- 2% (SE) of maximum to a peak response average of 50 +/- 3% of maximum. This effect was long lasting (45.6 +/- 8.6 min). Blood pressure responses were variable. Seven 1 M glutamate injections consistently decreased integrated phrenic amplitude significantly from a baseline average for all injections of 29 +/- 3% of maximum to a peak average of 20 +/- 5% of maximum. Respiratory cycle duration and blood pressure responses were variable. Prior injection into the RTN of 10 nl of 100 mM kynurenic acid attenuated the subsequent response of the integrated phrenic amplitude to injection of 10 nl of glutamate at the same site. Comparison of glutamate (10 nl, 100 mM) injected over 60 s vs. 30 ms showed that the prolonged increase in phrenic activity was related to the longer-duration (60-s) injections and that RTN single units were stimulated for up to 5 min on average after the 60-s injection with one unit showing an increase in firing rate over 40 min. After the 30-ms injection, integrated phrenic amplitude and RTN unit mean firing rate were increased for the first two breaths and at 5 min after the injection. We conclude that glutamate injected into the RTN increases local single-unit firing rate and the amplitude of the integrated phrenic activity. Injections made over 60 s result in prolonged phrenic stimulation and, in some cases, in RTN single-unit firing rate.

Differentially regulated immediate early genes in the rat uterus.

Estrogen stimulates cellular proliferation in the luminal epithelium, stroma, and smooth muscle of immature rat uterus. Progesterone administered concurrently with estrogen blocks the stimulatory effect of estrogen specifically in the epithelium, whereas progesterone administered alone stimulates proliferation in the endometrial stroma and myometrium. The present studies determined the effects of estrogen and progesterone on expression of the growth-associated, immediate early genes c-fos, c-jun, and jun-B in the luminal epithelium of the uterus. Hormonal effects were quantitated by Northern analysis of RNA extracted directly from the uterine luminal epithelium. Estrogen stimulated c-fos and jun-B expression, but repressed c-jun mRNA levels in the epithelium. In contrast, when whole organ RNA extracts were analyzed, estrogen increased mRNA levels for all three genes. Although progesterone administered alone showed no effect on mRNA levels in either epithelial or whole uterus extracts, it did attenuate the estrogen-induced increase in c-fos mRNA by 50% in whole uterus extracts and by 23% in epithelial extracts. The estrogen-induced increase in epithelial jun-B mRNA was not affected by progesterone pretreatment. Thus, in the immature rat uterus, no simple correlation exists between cellular proliferation and increased expression of the genes studied. However, progesterone completely blocked the repressive effect of estrogen on epithelial c-jun, suggesting a link between decreased c-jun expression and induction of cell proliferation in the uterine luminal epithelium. Estrogen repression of epithelial c-jun expression was hormone specific and sensitive to antiestrogen blockade. After estrogen treatment, epithelial c-jun mRNA decreased with a rate similar to its half-life, as determined in primary cultures of rat uterine cells. These results suggest that estrogen, acting through its receptor, directly represses transcription of c-jun in the uterine epithelium. Differences in hormonal regulation of immediate early genes between epithelial and nonepithelial uterine tissues probably results from tissue-specific transactivating factors that control the expression of these genes.

Serum IgG antibody responses to Shigella invasion plasmid-coded antigens detected by immunoblot.

Serum IgG antibody responses to Shigella invasion plasmid-coded antigens (Ipa) from 58 Shigella flexneri, S. sonnei, and S. dysenteriae infected Swedish patients were investigated by immunoblot technique. Intense responses to most components of Ipa (Ipas A, B, C, D, and VirG-virulence determinant on SalI fragment G of the plasmid) were evident in sera from S. flexneri infected patients. The strongest response was to Ipa B and the weakest, to Ipa D. In contrast, there were weaker responses to Ipas A, B, C, and VirG but none at all to Ipa D in sera from S. sonnei infected patients. After absorption of the Ipa-positive sera by Ipa expressing strains of S. flexneri and S. sonnei, most IgG antibodies to components of Ipa were removed in sera absorbed by S. flexneri, but IgG antibodies to Ipas--especially to Ipa D--were only slightly reduced in sera absorbed by S. sonnei, suggesting that Ipa D in S. sonnei may not be exposed on the S. sonnei cell surface.

Comparative metabolism of SC-42867 and SC-51089, two PGE2 antagonists, in rat and human hepatocyte cultures.

1. The metabolism of SC-42867 and SC-51089, two PGE2 antagonists, was studied in cultured rat and human hepatocytes. Both compounds possess an 8-chlorodibenzoxazepine moiety, but differ from each other by the nature of the side chain connected to the nitrogen atom. SC-42867 and SC-51089 and their in vitro metabolites were separated by reversed-phase hplc. The major metabolites of both compounds were identified by mass spectrometry (ms) analysis. 2. SC-42867 was metabolized on the tricyclic moiety only. Oxidative N-dealkylation with opening of the oxazepine ring was the major metabolic pathway obtained in rat hepatocytes. The metabolic profile obtained in cultured human hepatocytes was comparable with that of cultured rat hepatocytes. However, the compound was metabolized to a much lower extent by the human cells. 3. SC-51089 was extensively metabolized by both cultured rat and human hepatocytes. Human cells metabolized this compound quite differently than cultured rat hepatocytes. Aromatic hydroxylation with consequent glucuronidation and sulphation were the main metabolic pathways observed in cultured human hepatocytes. Oxidative N-dealkylation with opening of the oxazepine ring and consequent glucuronidation was the major metabolic pathway observed in rat hepatocytes. Further metabolism occurred, in contrast with the human hepatocytes, mainly on the side chain. 4. The present in vitro results are compared with data of previous in vivo studies performed in rat.

[Pingyangmycin-induced chromosome damage in lymphocytes of laryngeal cancer patients].

Lymphocytes from 28 untreated laryngeal cancer patients and 23 healthy controls were cultured in vitro and exposured to pingyangmycin (bleomycin A5), a clastogen. The lymphocytes were arrested in metaphase and analyzed. The total chromosome aberration rate, mean chromatid break rate per cell and cell aberration rate were 1.98% +/- 0.05%, 0.57% +/- 0.35%, and 42.8% +/- 12% respectively for laryngeal cancer patients. However, for healthy controls these values were 0.94% +/- 0.04%, 0.28% +/- 0.12%, and 27% +/- 12% respectively. Statistical analysis showed there are significant differences between the two groups. The data indicate that under our experimental conditions chromatid break rate 0.40 can be considered to be a borderline value, 0.80 hypersensitive value. For any individual, if the chromatid break rate is 0.40 or more, one should be ranked as having cancer risk. If 0.80 or more, then, highly cancer risk.

[Studies on morphological superstructure of inactivated rabies virus and uninactivated rabies virus].

Rabies is widely epidemic in human and animals. It is a very dangerous disease at present. In 1919, semple has shown that antigenicity was not destroyed by chemicals. After that inactivated rabies vaccine was widely used in human being up to more than seventy years. But the afferration of morphological superstructure between inactivation and uninactivated, we have not see any report on the world.

[The protection of gut mucosa with early enteral feeding in burned rabbits].

The mechanisms of the protection of gut mucosa with early enteral feeding in burns were investigated. A 25% TBSA burn rabbit model, in which a gastrostomy, and a blind segment of the jejunum were formed before the thermal injury. 39 rabbits were randomly divided into three groups: A. burn and feeding with the nutritional fluid within 30 minutes postburn through gastrostomy. B. same as A except feeding with cold water instead of nutritional fluid; and C. no burn and fed freely. The results showed that burn injury caused an increase in mucosal oxygen consumption, intestinal uric acid formation and MDA (malondialdehyde) production, and decrease in submucosal blood flow. However, these were significantly improved by early enteral feeding after burn injury. The changes in the mucosa of the jejunal blind segment were similar to that of the rest of jejunum. It indicated that the effect of enteral feeding might be a systemic as well as a local one. It is concluded that the early enteral feeding can maintain the mucosal function and structure of the gut, as evidenced by a decrease in the mucosal hypermetabolism, improvement in submucosal blood flow and reduction in gut injuries as a result of reperfusion.

[Effects of Chinese herb decoction sizunzituang on metabolism and nutrition in burnt guinea pigs].

Sizunzituang is a decoction prepared by boiling water extraction of codonopsis pilosulae, Acractylodes, poria and glycyrrhiza. Male burnt (30% TBSA) guinea pigs weighing 380-420 grams with a pre-formed gastrostomy were studied. The animals were divided into two groups, both of them were fed with pellet chow. One received sizunzituang, while the other group served as controls. The treated group was found to eat more food, with better nitrogen balance, higher body and ileum mucosal weights, and higher nitrogen contents of the liver and ileum mucosa than control group. However, the body and carcase weights as well as gastrocnemius muscle nitrogen contents in the treated group were reduced significantly as compared with those in the control group. In addition, Sizunzituang can also regulate the secretion of hormones and visceral water contents. The above findings indicate that the decoction may be beneficial in improving the metabolism, nutrition, and visceral organ functions after a burn injury.

[The roles of macrophage in immune dysfunction following severe thermal injury].

This experimental study in serious scald mice showed that the cytosolic free calcium ion concentration and the capacity of secreting TNF-alpha of macrophage (M phi) were obviously increased while the secretion of IL-1 was significantly decreased, and accompanied with simultaneous apparent rising of serum TNF-alpha in early postburn period. These indicate that M phi have been highly or even over activated after burn. But the ability of antigen presentation of M phi during this period was deeply suppressed rendering the obvious declination of activity and proliferation response of antigen pulsed lymphocytes. It might be concluded that the defective antigen presentation of M phi played a critical role in early immune dysfunction of severe scald mice, and the highly activated or even overactivated M phi by excessive secretion of TNF-alpha were the molecular basis of the alterations of M phi function.

[Plasma renin activity (PRA), angiotensin II (AII), atrial natriuretic peptide (ANP) and AII/ANP ratio in severely burned patients].

Plasma renin activity (PRA), angiotensin II (AII), atrial natriuretic peptide (ANP) and AII/ANP were determined by radioimmunoassay in samples of blood from 59 burn patients. The results demonstrated that PRA, AII, ANP, AII/ANP increased significantly postburn. The increased level of AII/ANP correlated positively with the extent of burn injury as well as the severity of postburn renal dysfunction suggesting that the imbalance of AII and ANP played an important role in the pathogenesis of postburn renal damage.

[Experimental study on cytoplasmic free calcium and antigen presentation function of peritoneal macrophage following severe scald].

A dynamic observation of cytoplasmic free calcium concentration (PM phi [Ca2+]i) and antigen presentation (AP) function was made in severely scalded mice. The results showed that [Ca2+]i of unstimulated PM phi in vitro increased significantly at 12, 24 and 72h postburn (PB), and the [Ca2+]i of PM phi stimulated by A23187 in vitro was also increased but not statistically significant at the same time points. These results suggested that PM phi was activated in vivo, even in a highly or over activated state. Severe scald caused the obvious depression of AP function of PM phi. This phenomenon, not only appearing very early but also persisting for a long time, might become the key factor of suppression of specific immune function PB. The changes in M phi activity in vivo might be the basic event of the M phi dysfunction after severe burns.

Dynamic changes of fibrinopeptides A and B in sera of burn patients and their effects on vascular endothelial cells.

In this study, dynamic changes of fibrinopeptides A and B (FPA, FPB) in sera of burns patients are determined with an HPLC method. On this basis detrimental effects of FPA and FPB on vascular endothelial cells (VEC) are observed in vitro. The main results are as follows: (1) FPA and FPB in 18 burn patients' sera increase at day 2 postburn, reach peaks at days 5-7 postburn, then decline after day 15 postburn, and return to the control levels at day 25 postburn. The fluctuations in FPA and FPB levels in burn patients' sera are parallel to the progress of burn illness. The measurements of FPA and FPB levels in burn patients' sera may be useful in evaluating patients' condition. (2) FPA and FPB are detrimental to the cultured VEC in vitro. Characteristics of their injurious effects are: specific, irreversible and dose dependent. FPA and FPB may play an important role in endothelial injury.

The isolation of Cryptococcus neoformans from pigeon droppings and serotyping of naturally and clinically sourced isolates in China.

This is the first report on the isolation of Cryptococcus neoformans from pigeon droppings in China and their serotypes. C. neoformans colonies which produced brown colonies on caffeic acid-cornmeal agar were found in Twenty-five out of thirty-six samples of pigeon droppings. Fifty-one colonies randomly picked from the positive samples were identified as C. neoformans by a commercially available kit for carbon source assimilation test and Christensen's urea agar. Forty (78%) out of the 51 strains were serotyped as A and 11 (22%) as AD. At the same time, seventeen out of nineteen clinical isolates were serotyped as A and 2 as B. There are three findings in our results. One is that only C. neoformans var. neoformans strains could be isolated from pigeon droppings, although the variety gattii strains were found in the clinical isolates obtained in the same geographic site in China. The second is that serotype A strains were most frequently seen in natural and clinical materials in the southeast part of China, and serotype AD strains were isolated in pigeon droppings but not in clinical materials. The third is that the coexistence of serotype A and AD cells of C. neoformans strains in same samples of pigeon droppings were observed.

Responses of respiratory modulated and tonic units in the retrotrapezoid nucleus to CO2.

We hypothesized that the retrotrapezoid nucleus (RTN) contains both respiratory modulated (RM) and non-respiratory modulated (NRM) neurons which participate in the ventilatory response to increased CO2. We made extracellular recordings of the activity of 46 single units in the RTN of 9 decerebrate, paralyzed, ventilated cats (5 intact; 4 with carotid body and sinus ablation) under eucapnic (PCO2 = 34.2 +/- 3.5 mmHg; mean +/- SD) and hypercapnic (PCO2 = 47.4 +/- 3.4 conditions. To define a RM unit, we used the eta 2 statistic which is the ratio of the variance of the unit firing rate within respiratory cycles to that across respiratory cycles. We classified the units as RM (N = 17) if the eta 2 values in eucapnia or hypercapnia were > or = 0.25 and as NRM (N = 29) if the values were < 0.25. Overall, 19/46 units (41%) increased their firing rate with increased CO2, 5 decreased their firing rate, and 22 had no significant change in firing rate. Of 17 RM units, 8 (47%) increased their mean firing rate with hypercapnia from 7.6 +/- 3.9 to 23.2 +/- 6.8 spikes/sec. These included 5 inspiratory units, 2 inspiratory units that had an onset of firing in late expiration (Pre-I/I), and 1 expiratory unit. Seven of these also changed their discharge pattern (eucapnic eta 2 = 0.02 to 0.12; hypercapnic eta 2 = 0.34 to 0.79) Of 29 NRM units, 11 (38%) showed a significant increase in mean firing rate with CO2 stimulation from 19.8 +/- 7.2 to 31.3 +/- 8.2 spikes/sec. The RTN has RM units which change their discharge pattern and firing rate in response to increased CO2, as do units within the medulla and pons, and it has NRM units which are also responsive to increased CO2. These data indicate that some neurons of the RTN are involved in the central chemoreceptor response but they provide no direct evidence that chemoreception resides within the RTN.

Safety and immunogenicity of the live oral auxotrophic Shigella flexneri SFL124 in adult Vietnamese volunteers.

The live, auxotrophic dependent Shigella flexneri Y vaccine strain SFL124 with a deleted aroD gene was tested in 30 healthy adult male Vietnamese volunteers. A single dose of 2 x 10(9) live bacteria was given orally to 15 volunteers, whereas 15 received three doses every other day. None of the volunteers reacted with fever or diarrhoea and SFL124 was excreted by all for a mean of 2.8 (single dose) and 2.6 (three doses) days. A total of 27 of 30 (90%) and 26 of 30 (87%) responded with significantly (0.001 < p < 0.01) increased antibody-secreting cell (ASC) numbers against Shigella flexneri Y lipopolysaccharide (LPS) and invasion plasmid-coded antigens (Ipa). A faecal IgA antibody response to LPS and Ipa was seen in 20 of the 30 (67%) volunteers against both antigens. Serum antibody responses were seen in 23 of 30 (77%) against the LPS and in 17 of the 30 against Ipa. The three-dose schedule elicited only somewhat stronger immune responses than the single-dose schedule. A booster dose of 2 x 10(9) live bacteria was given to half of the volunteers in each group after 6 months, the other half received the same dose after 12 months. Following the booster at 6 or 12 months (i) the excretion of SFL124 was significantly shorter (p < 0.05) than after primary vaccination; (ii) the anti-S. flexneri LPS and anti-Ipa faecal sIgA titres were significantly higher (p < 0.05 to p < 0.01) than after primary vaccination; (iii) the anti-LPS and anti-Ipa ASC responses were significantly lower (p < 0.05) and of shorter duration than after primary vaccination, and (iv) the serum anti-LPS and anti-Ipa responses were significantly elevated (p < 0.05) and similar to those seen after primary vaccination. The results indicate that SFL124 is a safe, live vaccine strain with a negligible reactogenicity in adults living in a Shigella endemic area. SFL124 induces specific immune responses against LPS and Ipa with a mucosal memory lasting for at least 1 year.
