[Comparative effects of different inhibitors of phenylalanine hydroxylase and dihydropteridine reductase. In vivo and in vitro study in rats].

The effects of inhibitors of phenylalanine hydroxylase and dihydropteridine reductase have been tested in vivo and in vitro to induce classical phenylketonuria and the variant due to dihydropteridine reductase deficiency. In addition two inhibitors of intestinal absorption of phenulalanine were investigated. Blood, urine, and hepatic parameters have been measured and are discussed.

Faster estimation of reserve bilirubin binding capacity of serum from the neonate by thin-layer chromatography on sephadex.

Knowledge of the reserve bilirubin-binding capacity is useful in management of hyperbilirubinemia in the newborn. We describe thin-layer gel chromatography that permits measurement of this variable, with a 250-mul sample of serum, within 50 min. Single samples from four different infants can be analyzed on the same plate. The capacity of 40 sera from jaundiced and of 19 sera from nonjaundiced infants was measured. Results by thin-layer Sephadex gel chromatography and column Sephadex chromatography correlated well. Dithiothreitol, added to the standard bilirubin solution, protected bilirubin from oxidation for at least six days, which obviated the time consuming preparation of standard solution before such determination.

Effects of serum-free culture media on human liver and fibroblastic cells.

The morphological aspects and amino acid variations of human fibroblast and liver cell monolayer cultures were studied in serum free media. Under these conditions, the behaviour of the two cell types differed greatly. The morphological changes for the liver cells, as compared with the fibroblasts, appear more quickly and some of these changes are particular. Furthermore, extracellular amino acid variations are of lesser importance for the liver cells, during the 10 days following the suppression of serum, except for serine.

[Effect of cycloleucine on renal uptake of dibasic amino acids and cystine].

Cycloleucinead ministration (a synthetic amino acid) to rats produces a selective hyperaminoaciduria bearing on dibasic amino acids (lysine, arginine, ornithine) and cystine. Interference of cycloleucine with tubular reabsorption of these amino acids was studied "in vitro", using rat kidney cortex slices. When dibasic amino acids and cystine are at physiological concentrations, the data indicate that cycloleucine decreases their intracellular accumulation. However, effect of cycloleucine "in vitro" is not specific whatsoever its concentration; similar results are obtained with neutral amino acids such as alpha-alanine and valine. Inhibitory effects are not modified by the presence of several amino acids in the incubation medium. The results are interpreted as evidence for a dissociation between "in vitro" cellular accumu-lation and "in vivo" transepithelial transport.

Comparative study of extracellular amino acids in culture of human liver and fibroblastic cells.

Amino acid concentration are studied in the extracellular media of ten series of human fibroblast and liver cell monolayer cultures. These two cell types consume and produce ostensively the same amino acids. Among the nonessential amino acids, the most significant variations involve serine and aspartate which are decreased; alpha-alanine, glutamate, ornithine and proline are, on the contrary, increased. Among the essential amino acids, leucine, isoleucine and glutamine are preferentially decreased. The variations of some amino acids are correlated with the cell density. The interrelations which may exist between the variations of these different amino acids are discussed. Furthermore, the glycolytic acitivity of the cells studied is very high: 85% of glucose consumed is found in the form of lactate.

[Lipoprotein lipase activity in human milk; inhibition in vitro of the glucuro-conjugation of bilirubin (author's transl)].

The lipolytic activities of esterase, lipase and lipoprotein lipase were measured in 17 samples of human milk. Lipase and esterase activities were the same in all samples. However, lipoprotein lipase activity was increased only in samples provided from mothers whose infants had prolonged neonatal jaundice; after storage these samples inhibited the glucuro-conjugation of bilirubin in vitro and their concentration in non-esterified fatty acids was high. The mechanism of action of this enzyme is discussed in relation to the physico-chemical state of fat globules.

[Automated determination of serum BSP. Improvement of dialysis by sodium salicylate].

Because of the great difficulty of BSP dialysis in automatized determinations in serum the authors investigated the effect of sodium salicylate. They proved its good performance with aqueous and proteic solutions of BSP. About 70 mul of patient sera is used to perform the analysis with Technicon Autoanalyser. Comparison of data from about 220 serum evaluated by their proposed method and manual technique are studied.
