[Sea-blue histiocytes and storage diseases. 3 cases].

"Sea-blue" with prominent blue granules on Giemsa staining have been described in many diseases. The authors report three cases of storage diseases, in which these particular cells have been found. The significance and pathogenesis of "sea-blue" histiocytes are discussed.

Hepatoma after thorotrast. Morphological and autoradiographic data.

Two women, respectively given thorotrast 30 and 40 years ago, died of hepatoma. The spleen and liver of one and the liver of the other were studied by morphological, macro- and micro-autoradiographic methods. The part played by thorotrast in the neoplasic process and its effect on the mechanism of tumour induction are discussed.

[Pulmonary complications in "sniffers"].

When a sudden death occurs, a necropsy is of the upmost value, but sometimes it does not yield the anticipated benefit; in this situation histological study of the specimens may help the diagnosis. We describe two decreased "sniffers" with pulmonary lesions that could have explained their sudden deaths. The histological appearance is often not recognised and considerably worsens the prognosis in subjects inhaling certain drugs.

[Morphological, histochemical and biochemical changes induced by homochlorcyclizine in the rat].

Young Wistar rats have been treated with homochlorcyclizine and the morphological, histochemical, biochemical changes studied. Lungs contained few macrophages overloaded with phospholipids but the main variations occurred in liver. Histochemical reactions showed draught-board patterns due to selective phospholipid or glycolipid storage within a part of the lobule. The phospholipidosis induced by the drug presented an increase in phosphatidyl choline and acid phospholipids and some lysosomal enzymatic activities were increased also.

Effects of Pexid on liver cell cultures. Ultrastructural and histoenzymological studies.

The effects of different doses of Pexid on cultured liver cells have been studied by ultrastructural and histoenzymological techniques. Two types of abnormal lysosomal inclusions were seen: clear matrix inclusions with either homogeneous or tiny lamellar structures, and dark matrix inclusions with clear vacuoles and either random or myelin-like lamellar structures. These patterns correspond to storage of triglycerides, phospholipids and gangliosides, either singly or together.

Histoenzymological activities of Gaucher cells.

Seven samples from liver and/or spleen of patients with Gaucher's disease have been investigated by histoenzymological techniques. Unexpected results such as beta-glucosidase positivity, increase of other hydrolases, and increase of dehydrogenase activities are discussed.

Deoxyglucose uptake in pathological conditions.

In case of quick physiological variations or in pathological conditions, it is impossible to measure the deoxyglucose consumption by means of the deoxyglucose technique and we propose a method for the study of the deoxyglucose uptake by the cerebral tissue. It is simultaneously measured with the blood flow, 3 min following the intravenous administration of deoxyglucose (2-deoxyglucose-14C). Hypercapnia induces an increase in the flow but does not modify the deoxyglucose uptake and moderate hypoxia results in a decrease in the deoxyglucose uptake without modifying the flow. In pathological conditions such as ischemia or stricture, the blood flow and deoxyglucose uptake variations are not parallel. These results show that the 2-deoxyglucose uptake does not follow the blood flow and that it depends on the cell activity. It is therefore possible to use this method when the consumption studies cannot be implemented.

[Brain rat histoenzymological changes induced by microsphere injection during ischemia (author's transl)].

The cerebral thrombosis of the rat with 35 micrometer labeled microspheres gives infarcted areas easily seen. After 15 min, in these areas, there is no change in NADH diaphorase-, succinic-dehydrogenase-, mono-amino-oxidase-, glucose-6-phosphate-dehydrogenase-, isocitric dehydrogenase-, lactic-dehydrogenase-, ATPase, alpha galactosidase-, acid phospatase activity. After 2, 4, 6 h all these activities diminish in the neuropil but they are preserved in the neurones for diaphorase, succinic-dehydrogenase and mono-amino-oxidase. The margin of infarcted areas shows a strong staining for acid phosphatase. Before 2 h there is not enzymatic changes neither oedema. This experimental model seems trustly and could be developped.

[Histological and histoenzymological study of liver. I: In gallstone without treatment (author's transl)].

46 liver biopsies from gallstones and a control group have been studied by histological and histoenzymological technics. The most frequent injuries are in the portal tract, inflammatory infiltration and fibrosis; the most peculiar is a sinusoidal congestion. A brown pigment overload is seen in numerous livers. Lactic-, glyceric-, glutamic-, isocitric-, glucose-6-phosphate-deshydrogenase activities decrease but alkaline phosphatases-, transaminases activities are increased.
