A decreased serum leptin level in patients with systemic sclerosis.

Serum leptin levels were determined in 31 women with systemic sclerosis and 24 age-matched healthy controls. Both groups were divided into premenopausal and postmenopausal subgroups. A decreased serum leptin was found in the patients with systemic sclerosis. The premenopausal patients and controls had higher serum leptin than those in the postmenopausal subgroups. Serum leptin correlated with body mass index in the patients with systemic sclerosis.

Free radical activity and antioxidant defense mechanisms in patients with hyperthyroidism due to Graves' disease during therapy.

Free radical-mediated oxidative stress has been implicated in the etiopathogenesis of several autoimmune disorders. We investigated the prooxidant-antioxidant status in order to evaluate the possible deleterious role of oxidative phenomena in patients with Graves' disease. Thirty patients with hyperthyroidism due to Graves' disease were investigated. Thirty age-matched healthy subjects were studied as a control group. Free radical activity indices, antioxidant defense systems, and thyroid and pituitary hormone levels were measured in fasting blood samples. Blood samples were taken before initiation of therapy and after attainment of euthyroid state. A significant increase in lipid peroxidation activity indices, i.e., conjugated dienes and thiobarbituric acid-reacting substances, was found in blood serum of the patients with untreated Graves' disease. These changes were accompanied by a decrease in plasma thiol and erythrocyte lysate thiol groups concentrations. Hyperthyroidism resulted in a marked increase in intracellular antioxidant enzymes, i.e., superoxide dismutase, catalase and glutathione peroxidase activities as compared to the controls. Extracellular anti-free radical scavenging systems potential, measured by glutathione reductase activity and total antioxidant status level, was found to be significantly decreased in untreated Graves' patients. Treatment with thiamazole resulted in normalization of the free radical and antioxidant activity indices. The obtained results indicate an enhanced generation of reactive oxygen species and impairment of cellular and extracellular antioxidant systems potential in patients with Graves' disease. The attainment of euthyroid state led to an improvement in oxidative stress indices and antioxidant potential parameters.

[The respiratory system of workers employed in the casting and processing of copper].

The aim of the study was to examine the influence of work in exposure to copper on the respiratory system. The study was carried out in 267 men aged (x +/- SD) 40.6 +/- 7.2 yrs, workers of a colour metal mill. 134 of them dealt with casting and processing of copper (the mean weighed concentration of copper on work posts was below 0.05 mg x m-3) and 133, not employed in a contaminated environment, comprised the control group. The subjects were divided into five groups according to the degree of energy expenditure at work and, additionally cigarette smoking was taken into account. All subjects underwent medical examination and spirometry. However the mean values of the ventilatory parameters in the compared groups did not significantly differ (p > 0.05), the multivarious regression analysis has shown a decreasing trend of FEV1 and FVC with relation to the exposition to copper and tobacco smoking. The frequency of chronic bronchitis as well as of radiological changes was not increased. The authors conclude: 1. At copper concentration not exceeding the MAC, and an advantageous microclimate, this exposition only slightly diminishes the pulmonary ventilation and does not cause other comprehensible pulmonary pathological sequelae, especially chronic bronchitis. 2. Tobacco smoking is a favourable factor for the occurrence of chronic bronchitis.

[Cognitive function in patients with systemic sclerosis].

Central nervous system involvement is seldom reported in patients with systemic sclerosis (SSc). Cognitive functions were determined in 21 patients with definite SSc and 42 healthy controls. Thyroid function was also measured in order to eliminate the effect of hypothyroidism on cognitive functioning. It was found that the SSc patients with normal thyroid function showed defective long-term and recent memory, learning ability, criticism, perception and visuo-perceptual skills, their simple reaction time was prolonged. Similar but less advanced cognitive defects were shown in the SSc patients with overt or latent hypothyroidism. The obtained results indicate that the central nervous system involvement is more common in patients with SSc than it has been reported earlier.

Effect of cimetidine and ranitidine on absorption of [125I]levothyroxine administered orally.

Female patients with a simple goiter were pretreated on 2 occasions (at an interval of 4 wk) with po placebo or 400 mg cimetidine (Cim) (Group A, n = 10), or with placebo or 30 mg ranitidine (Ran) (Group B, n = 10), 90 and 150 min, respectively, prior to the po gelatin capsules containing [125I]levothyroxine ([125I]LT4). A double-blind randomized study protocol was kept. Venous blood samples were taken at 15, 30, 45, 60, 75, 90, 105, 120, 150, 180, 210, and 240 min after po [125I]LT4 and the radioactivities in serum were counted. Similar [125I]LT4 radioactivities were found after placebo pretreatment in both groups: AUC 467 +/- 82 in Group A vs 459 +/- 109 in Group B. Cimetidine decreased the serum [125I]LT4 radioactivities: AUC371 +/- 72 (Cim) vs 467 +/- 82 (placebo) (P < 0.01), but Ran did not: AUC 477 +/- 132 (Ran) vs 459 +/- 109 (placebo) (P > 0.05).

[Subchronic toxic effect of the grain fungicide PF-70 on the electrolyte barrier of the gastric mucosa in rats].

The fungicide for corn PF-70 contains 21% of sulphur, 8.4% of mancozeb, and 0.6% of triadimefon . The aim of the study was to determine the effect of the fungicide on the gastric mucosa electrolyte barrier in rats. Mucosa lesion is known to be one of the risk factors of chronic peptic ulcer disease. The experiments were carried out on 80 rats of Wistar strain. The animals were divided into 5 groups--males and females, separately. The fungicide was given for 13 weeks, at the following doses: 100, 400, 1600, and 6400 mg of active substance per 1 kg of fodder. The gastric mucosa electrolyte barrier was examined by determining the transmural electric potential differences (PD) in mucosa before and after treatment with 150 mmlo of hydrochloric acid and 40 mmol of acetylsalicylic acid solutions. No changes in general condition of the animals were observed. An increase in fodder consumption was observed in males receiving the fungicide at doses of 1600 and 6400 mg per 1 kg of fodder. Females exhibited a decrease in body mass after intoxication at concentration of 6400 mg/kg fodder. A decrease in PD was found in all groups intoxicated with the fungicide, irrespective of the animal sex.

[Effect of long-term exposure to high manganese levels in the air of the work area on the content of various immunoproteins in the blood].

Fifty-four subjects chronically exposed to high manganese concentrations due to professional melting of iron-manganese alloys have been examined. The threshold limit value was exceeded 3 to 4-fold. The minimum length of employment in exposure was 5 years, whereas the average-17.7 years. Control groups were matched by age, length of employment and energy expense. Among the determined parameters, i.e. IgG, IgA, IgM concentrations in blood serum and C3 and C4 elements of the complement--only the IgM concentration was significantly lower, whereas the others were not significantly different from those in relevant control groups.
