Cytokeratin polypeptide in cultured human squamous cell carcinoma as a possible marker for keratinization.

A human cancer cell line (ZK-1) has been established from a well-differentiated squamous cell carcinoma of the tongue. The cytokeratin polypeptides pattern of ZK-1 cells consists of four major polypeptides with molecular weights between 46 and 58 kilodaltons (kd). Antibodies raised against the purified 58 kd cytokeratin filament from cultured ZK-1 cells were shown to be specific by one- and two-dimensional polyacrylamide gel electrophoresis, immunofluorescence, and immunoelectron microscopy. Immunoprecipitation studies showed that the antibody reacted mainly with the 58 kd cytokeratin. The distribution of the 58 kd cytokeratin in both sparse and confluent cultures was analyzed by the indirect immunofluorescence technique. In both cases, it appeared that fibrillar arrays extended throughout the cytoplasm running over the nucleus and toward cell-to-cell boundaries. Thick bundles of filaments were seen in confluent cultures, especially in large and flattened keratinized cells. Characteristically, reactions were also seen in the intercellular boundaries, appearing as "dots." Electron microscopy using immunoperoxidase techniques indicated that the 58 kd cytokeratin was localized in tonofilaments, tonofilaments attached to desmosomes, desmosomal plaques and membrane-coating granules.

A case of pleomorphic adenoma of the trachea.

An unusual tumor of the trachea was found in a 76-year-old woman. The tumor resembled an adenoid cystic carcinoma in having many cystic spaces, but the fairly uniform tumor cells gradually merged with the myxomatous or hyalinous stroma, and foci of squamous metaplasia and glycogen-rich adenoma-like components were also observed. S-100 protein was demonstrated immunohistochemically in the tumor cells. This tumor was diagnosed as a pleomorphic adenoma (benign mixed tumor) of the trachea.

A prospective study of congenital cytomegalovirus infection in Japan.

A prospective study on congenital cytomegalovirus (CMV) infection was carried out in Sapporo, Japan. Only 77 (6.2%) of 1233 pregnant women were seronegative in the first trimester. In such a highly immune population, 11 (0.5%) of 2070 newborn infants were found to have congenital CMV infection. None of them showed any clinical abnormalities during the period of observation (3-24 months), except for a cyst of septum pellucidum in one case. The incidence of congenital CMV infection in the first pregnancies was 1.1% (9/816) and in subsequent pregnancies 0.2% (2/1254). The difference was statistically significant. Three mothers of infants with congenital CMV infection were already seropositive in the first trimester. 26/30 (86.7%) infants, born to mothers with active CMV infection during pregnancy, had started to excrete CMV with urine before the age of 12 months. In contrast, none of the 4 infants born to seronegative mothers excreted CMV.

An odontogenic myxofibroma related to an embedded third molar of the mandible. Report of a case.

A case of odontogenic myxofibroma involving the angle of the mandible is reported. This case arose in the area closely associated with the pericoronal portion of the embedded third molar. When the mandible resected was histopathologically examined, the tumor mass containing various shapes of epithelial cell nests and amorphous calcified bodies in the myxomatous fibrous tissue was found to be formed in the localized area well-demarcated by the enamel of the embedded tooth. These findings strongly suggest that this lesion is of odontogenic origin.

Establishment of a human cancer cell line from carcinoma of the gingiva.

A new human epithelial cell line (MK-1) has been established from a well differentiated squamous cell carcinoma of the gingiva. It has been subcultured 90 times during the past 23 months. The population doubling time of the MK-1 cell line was 28 h. A colony forming ability with 18% plating efficiency was shown. Chromosomal analysis revealed an aneuploid human male karyotype with 22 marker chromosomes. The model chromosome number was 75. Scanning electron micrographs of MK-1 cells showed numerous microvilli and intercellular bridges. Transmission electron micrographs of MK-1 cells showed epithelial characteristics with desmosomes, tonofilaments and membrane-coating granules. Hetero-transplantation of cultured MK-1 cells produced tumors, the histology of which was similar to that of original ones. The ultrastructure of the tumors showed the presence of abundant tonofibrils and keratohyalin granules. This MK-1 cell line is a model useful for the study of keratinization of squamous cell carcinomas.

[Effect of predosing with Y-12,141 on its antiallergic activity. --Studies on anti-allergic agents(IV)-- (author's transl)].

Y-12, 141 (0.1 approximately 0.3 mg/kg) given i.v. 1 min before the antigen challenge inhibited both the 48-hour passive cutaneous anaphylaxis (PCA) and active anaphylactic bronchoconstriction of rats mediated by IgE-like antibody. The anti-PCA activity of Y-12, 141 (0.3 mg/kg i.v.) given 1 min before the antigen challenge decreased significantly by predosing with Y-12, 141 (1 approximately 3 mg/kg i.v.) 60 min before the antigen challenge. The induction of tachyphylaxis in the PCA test appeared to depend on the doses in internal between predosing and second dosing. The anti-PCA activity of disodium cromoglycate (DSCG) at a dose of 1 mg/kg i.v. decreased significantly by predosing with DSCG (10 mg/kg i.v.) 30 min before the antigen challenge. Cross-tachyphylaxis between Y-12, 141 and DSCG was observed in the PCA test. The tachyphylaxis to Y-12, 141 was more readily induced in the PCA than in the bronchoconstriction. The inhibitory activity of Y-12, 141 (0.3 mg/kg i.v.) given 1 min before the antigen challenge in the PCA and bronchoconstriction was not affected by predosing with Y-12, 141 (1 mg/kg p.o. or s.c.) for 7 days.

Histological and microradiographical studies of human mandibular condyle.

Twenty-one clinically normal human mandibular condyles were examined by histologic and microradiographic methods obtaining some measurements. The histologic structures of condyle were quite different between the child group and other aged groups. The difference in the surface articular zone composed of the fibrous tissue was not so great in all age groups, while there was quite a difference in the structure in the remaining zones. It was clearly demonstrated by microradiogram that the osteons of the subarticular bone plate were more prominent in aging. Abnormal structures of the condyle were revealed in some individuals of the adult and old age groups, which were assumed probably to be the reactive remodelling against the functional changes of the temporomandibular joint.
