Urinary oligosaccharides of fucosidosis. Evidence of the occurrence of X-antigenic determinant in serum-type sugar chains of glycoproteins.

Urine of a fucosidosis patient contained a large amount of fucosyl oligosaccharides and fucose-rich glycopeptides. Six major oligosaccharides were purified by a combination of Bio-Gel P-2 and P-4 column chromatographies and paper chromatography. Structural studies by sequential exoglycosidase digestion and by methylation analysis revealed that their structures were as follows: Fucalpha1 leads to 6GlcNAc, Fucalpha1 leads to 2Galbeta1 leads to 4(Fucalpha1 leads to 3)GlcNAcbeta1 leads to 2Manalpha1 leads to 3Manbeta1 leads to 4GlcNAc, Galbeta1 leads to 4(Fucalpha1 leads to 3)GlcNAcbeta1 leads to 4Manalpha1 leads to 4GlcNAc, Galbeta1 leads to 4(Fucalpha1 leads to3)GlcNAcbeta1 leads to 2Manalpha1 leads to 6Manbeta1 leads to 4GlcNAc, and Galbeta1 leads to 4(Fucalpha1 leads to 3)GlcNAcbeta1 leads to 4Manalpha1 leads to 6Manalpha1 leads to 6Manbeta1 leads to 4GlcNAc. In additon, the structure of a minor decasaccharide was found to be Galbeta1 leads to (Fucalpha1 leads to)GlcNAcbeta1 leads to Manalpha1 leads to [Galbeta1 leads to (Fucalpha1 leads to)GlcNAcbeta1 leads to Manalpha1 leads to]Manbeta1 leads to 4GlcNAc.

Oligosaccharides of human milk. Structural studies of two new octasaccharides, difucosyl derivatives of para-lacto-N-hexaose and para-lacto-N-neohexaose.

Two new octasaccharides were isolated from human milk of a nonsecretor Lewis a+b- individual. Structural studies by sequential enzymic degradation and by quantitative methylation analysis revealed that their structures were as follows: Galbeta1 leads to 3 (Fucalpha1 leads to 4) GlcNAcbeta1 leads to 3Galbeta1 leads to 4 (Fucalpha1 leads to 3) GlcNAcbeta1 leads to 3Galbeta1 leads to 4Glc, Galbeta1 leads to 4 (Fucalpha1 leads to 3) GlcNAcbeta1 leads to 3Galbeta1 leads to 4 (Fucalpha1 leads to 3) GlcNAcbeta1 leads to 3Galbeta1 leads to 4Glc. The core portions of these sugars, which are newly found linear hexasaccharides, are named para-lacto-N-hexaose and para-lacto-N-neohexaose, respectively.

Structure of a carbohydrate moiety of a unit A glycopeptide of calf thyroglobulin.

The structure of the carbohydrate moiety of a unit A glycopeptide of calf thyroglobulin was studied by glycosidase digestion, acetolysis, and methylation analysis. Based on the results, the whole structure of a unit A glycopeptide with the largest carbohydrate moiety was determined as Man alpha 1 leads to 2Man alpha 1 leads to 6(Man alpha 1 leads to 2Man alpha 1 leads to 3)Man alpha 1 leads to 6(Man alpha 1 leads to 2Man alpha 1 leads to 2Man alpha 1 leads to 3)Man beta 1 leads to 4GlcNAc beta 1 leads to 4GlcNAc leads to Asn-peptide.

The carbohydrate structure of a glycopeptide released by the action of plasma kallikrein on bovine plasma high-molecular-weight kininogen.

Fragment 1, released from bovine plasma high-molecular-weight kininogen by the action of plasma kallikrein, is a glycopeptide with approximately 3 mol of each of N-acetylgalactosamine, galactose and sialic acid in one molecule. All these sugars were in the T-1 fragment obtained by tryptic digestion of fragment 1. Sialic acid can be completely released from the T-1 fragment by sialidase digestion. When this sialic acid-free T-1 fragment was incubated with purified diplococcal endo-alpha-N-acetylgalactosaminidase, all remaining sugars were released as a disaccharide. By Smith degradation and beta-galactosidase digestion, the structure of this disaccharide was found to be Gal beta 1 leads to 3GalNAc. Methylation analysis of the trisaccharide released from fragment T-1 by alkaline borohydride treatment indicated that all the galactose was obtained as the 2,4,6-tri-O-methyl derivative. Based on this evidence, the complete structure of the carbohydrate moieties of fragment 1 was proposed as Sialyl alpha 2 leads to 3Gal beta 1 leads to 3GalNAc. In addition, small amounts of a tetrasaccharide, Sialyl alpha 2 leads to 3Gal beta 1 leads to 3(Sialyl alpha 2 leads to 6)GalNAc also occurred as a carbohydrate chain of fragment 1.

Purification and characterization of an endo-beta-galactosidase produced by Diplococcus pneumoniae.

An endo-beta-galactosidase acting on blood group A and B substances was found in the culture fluid of Diplococcus pneumoniae. The enzyme was purified 1000-fold, and its properties were studied in detail. The enzyme preparation, thus obtained, was practically free from various exoglycosidases, endo-beta-N-acetylglucosaminidase and proteases. The enzyme releases trisaccharides from blood group A and B active mucins purified from ovarian cyst fluid. The structures of the trisaccharides liberated from A and B active mucins were elucidated to be GalNAcalpha1 leads to 3(Fucalpha1 leads to 2)Gal and Galalpha1 leads to 3(Fucalpha1 leads to 2)Gal, respectively. The enzyme also hydrolyzes blood group A and B active oligosaccharides composed of type 2 chains, yielding the same products as in the case of ovarian cyst blood group substances. An H active mucin from ovarian cyst fluid, H active oligosaccharides, and A and B active oligosaccharides with type 1 chains were not hydrolyzed by the enzyme. Consequently, the enzyme catalyzes the following reaction, resulting in the degradation of blood type A and B determinants. (see article).

Chemical characterization and distribution of ABO blood group active glycoprotein in human erythrocyte membrane.

Occurrence and distribution of glycoprotein H substances in blood type O human erythrocyte membrance were studied using an alpha N-acetylgalactosaminyltransferase (A-enzyme) purified from milk of blood type A women. Erythrocytes of blood group O, nonsecretors and secretors alike, contain H substances that serve as a substrate for this transferase, and the H determinants seem to occur in all three erythrocyte membrane glycoproteins (periodic acid-Schiff (PAS)-1, PAS-2, and PAS-3). Structural studies of the A glycoproteins obtained by the transferase action, suggested that all erythrocyte H glycoproteins contain only type 2 antigenic determinant.

Partial purification and characterization of an endo-alpha-N-acetylgalactosaminidase from the culture of medium of Diplococcus pneumoniae.

The culture medium of Diplococcus pneumoniae contains enzymic activity that cleaves Galbeta1 leads to 3GalNAc from desialized human erythrocyte membrane glycoprotein. The enzyme was purified 180-fold by ammonium sulfate fractionation, gel filtration through a Sephadex G-200 column, and DEAE A-25 Sephadex chromatography. The purified enzyme liberates Galbeta1 leads to 3GalNAc from glycopeptides and glycoproteins with Galbeta1 leads to 3GalNAcalpha1 leads to Ser and Thr moieties. The optimum pH of this enzyme is 6.0. Using glycopeptides obtained by trypsin digestion of human erythrocyte membrane glycoprotein as a substrate, a Km of 0.20 mM (on the basis of the amount of Galbeta1 leads to 3GalNAc residues) was obtained. So far, the enzyme appears to have a strict specificity for Galbeta1 leads to 3GalNAcalpha1 leads to Ser and Thr structures, because no oligosaccharides larger than trisaccharides were liberated from porcine submaxillary mucin.
