Double replica electroblotting for the simultaneous phenotyping of C6 and C7, and its application to the examination of bloodstains.

A new reliable and reproducible technique for the simultaneous determination of C6 and C7 types is presented, which employs double replica electroblotting after isoelectric focusing. It permitted clear discrimination of both C6 and C7 components, and the patterns were nearly comparable to those demonstrated separately. The population data obtained by this new technique fitted the genetic hypothesis. The present double replica electroblotting method was successfully applied to the combined phenotyping of C6 and C7 from bloodstains which were stored at room temperature for up to 4 weeks. The method is quite suitable for medicolegal examination of bloodstains particularly for the saving in the amount of sample.

Possible involvement of bcl-2 suppression in wild-type p53 gene-dependent cell growth repression in rat osteosarcoma cells.

We recently obtained 3 cloned cell lines demonstrating the p53 mutation from a lung metastatic nodule of a rat transplantable osteosarcoma. In this study, we applied wild-type p53 gene transfer to the rat osteosarcoma cells by lipofection to investigate the effects on cell growth, expression of genes such as waf1/p21, bcl-2, and bax, and nucleosomal DNA fragmentation due to apoptosis. Reconstitution of the p53 gene inhibits cellular growth, and this growth-suppressive effect is partly due to apoptosis involving bcl-2 gene suppression in this tumor type. This rat osteosarcoma model is similar in biologic behavior to human cases and thus is very suitable for further investigation of tumorigenesis and gene therapy for osteosarcoma.

Preoperative estimation of neural invasion in rectal carcinoma.

To reduce postoperative local recurrence in patients who undergo a nerve preserving operation for rectal carcinoma, preoperative estimation of neural invasion would be desirable. For this purpose we clarified the characteristics of rectal carcinomas with neural invasion and then evaluated the usefulness of matrix metalloproteinase 7 (MMP7) gene as an indicator for neural invasion. The study included 128 patients with primary rectal carcinoma. The histologic examination plus immunohistochemical study for S-100 protein was performed. A reverse transcription-polymerase chain reaction (RT-PCR) study for MMP7 using preoperatively obtained biopsy specimens was also performed in 32 patients. Neural invasion was recognized in 38 (29.7%) of 128 tumors. The tumors with neural invasion were characterized by infiltrative growth pattern, frequent lymphatic and venous permeation, frequent lymph node metastasis, and advanced stage of disease. The cases with neural invasion also showed a higher recurrent rate and worse prognosis. An RT-PCR study demonstrated that a significantly higher expression of MMP7 mRNA was recognized in neural invasion positive cases than in negative ones (p<0.05), indicating that MMP7 may be a good indicator for neural invasion of rectal carcinoma. Neural invasion would be an additional useful factor for providing information on malignant potential. The RT-PCR results of MMP7 gene may be useful for predicting which tumors may develop neural invasion.

[Practical value of LDH-X determination for the detection of sperm traces].

The examination of 38 stains collected in sexual offences provided spermatozoa-positive, LDH-X-negative results in 4 samples and spermatozoa-negative, LDH-X-positive results in 6 samples. The results suggest that besides the microscopic detection of spermatozoa the demonstration of LDH-X should be performed in medicolegal identification of seminal stains.

[C7 typing of blood stains using isoelectric focusing and immunoblotting].

By means of isoelectric focusing and immunoblotting C7 types were clearly demonstrated from bloodstains which had been stored at 37 degrees C for up to three weeks, at room temperature for up to six weeks and at 4 degrees C for over ten weeks. The C7 typing is practically useful in medicolegal individualization of unknown bloodstains.

Thoracoscopic limited resection of bronchogenic carcinoma in patients over the age of 80.

Video-assisted thoracoscopic surgery (VATS) has emerged as a minimally invasive therapy of bronchogenic carcinoma, when the diseases occur in patients unable to tolerate open thoracotomy who are 80 years old or older. We report on an 80-year-old man with poor pulmonary function and an 83-year-old man, who were treated with a complete VATS resection of bronchogenic carcinoma. Mean chest tube duration of the two patients was one day and mean postoperative hospital stay was 7 days with an uncomplicated postoperative course. Compared with patients with open thoracotomy, there were early mobility, less morbidity, and less mortality. We believe that when the indication for treatment of bronchogenic carcinoma in patients over the age of 80 with concomitant systemic disorders, a thoracoscopic limited resection should be considered.
