The usage of infrared, ultraviolet, luminescence both in infrared and ultraviolet for anatomical purposes was studied and the films were transformed with image computer analysis system. Infrared turned out to be exceptionally useful for examination of deep structures. Ultraviolet and especially luminescence in ultraviolet enables tracing the elements placed on the surface. Image computer transformation makes possible the spatial assessment of selected details and it broadens the exploratory possibilities.
In this work usefulness of latest techniques in anatomical studies was assessed. The following methods were used: injection, Pickworth's method, image computer analysis with the usage of four systems: BVS 6471, BVS 6472, cytochromics, imtron 2000. Particular linear programs were analyzed and discussed. Several techniques were used simultaneously to reach difficult accessible structures.
Observations of periventricular matrix vascularization were performed. Following technics were used: injection Pickword's method, infrared, image computer analysis. Various transformations were applied; linear and mathematical morphology. The examined material consisted of 200 fetuses aged 4-8 months of gestation. Subependymally localized and sinusoidally extended vessels were observed. Arteries reaching the ventricular lumen were seen as anatomically and functionally terminal, dendroidally ramificating only at the periphery. The connections between vessels of periventricular matrix and those of surrounding white matter were evidentiated.
Identification hilar cells in fetus and adult gonads two sex and dysgenetic gonads were based on histochemical and immunohistochemical methods. The hilar cells were detected in 40% of fetus gonads (ovarian testis), two time more in adult and dysgenetic gonads. The attention was put to their connection with nervous system. On the bases of specific markers there was established their mesenchymal origin, ability to response to gonadotropin induction and synthetization steroid hormones, mainly androgens. Hilar cells in dysgenetic gonads and postmenopausal women may be responsible for clinical manifestation of virilisation.
The aim of the work was to examine internal vascularization of telencephalon. The brains of 200 fetuses aged 3-8 months of fetal life were examined. Various techniques were used: injection, infrared, Pickworht's method, image computer transformation. Linear and non-linear transformations (mathematical morphology) were also used. The five vascular layers of telencephalon were distinguished: cortical, of white matter, striatal, germinal and of choroid plexus. Their morphology and changes occurring in fetal period were estimated. It was observed that subcortical and paraventricular regions belong to hypovascular areas. White matter vessels were seen as rectilinear without any ramifications. After 6 months of fetal life the most intensive development of inner cerebral vessels occurs. Two age groups were distinguished. The first one comprises the period of 4th to 6th month and is characterized by the existence of long medullary and abundant germinal matrix vessels. The second group (over 7th months) may be described as intensive development of cortical vessels.
Application of mathematical morphology in morphologic sciences was tested. Two aspects were evaluated, cognitive and utilitarian on analyzing enigmatic objects in TK and searching for particular elements in angiologic photos taken in infrared. The results prove many application possibilities of the method. Combination of modern techniques: infrared, Pickworths method, TK, and image transformation delivers many new data. Mathematical morphology selects interesting details and rejects redundant information.
Comparative susceptibility testing of 428 strains of obligate anaerobic bacteria belonging to genera Propionibacterium, Arachnia, Actinomyces, Bacteroides, Prevotella, Porphyromonas, Anaerorhabdus, Fibrobacter, Fusobacterium, Peptostreptococcus and Clostridium to metronidazole and tinidazole was performed. The study of the susceptibility of anaerobic bacteria was carried out by the method of serial dilution in Brucella agar according to Finegold and Sutter (1972). Strains of B. fragilis species, B. fragilis group, other Bacteroides spp.. Fusobacterium spp. and Clostridium spp. were susceptible to both chemotherapeutics in clinically attainable concentrations. Of particular significance is the greater susceptibility of these bacteria to tinidazole. Taking into account this activity, tinidazole is a superior agent in the treatment of anaerobic infections. Both chemotherapeutics were not active against, rarely isolated from severe infections. Gram-positive anaerobic rods from genera Propionibacterium. Actinomyces and Arachnia and were partially active against peptostreptococci.
For evaluation of usefulness of natural fluorescence of clinical materials in UV radiation as rapid diagnostic method of infections with anaerobes, 405 samples of pus, bloody-purulent fluids, blood, wound secretions, dressings and other materials were investigated. Occurrence of red-brick UV fluorescence of clinical materials was compared with results of culture aimed at isolation of non-sporeforming anaerobes from "B. melaninogenicus group (P. melaninogenica, P. intermedia and P. saccharolytics). Significant correlation red-brick fluorescence of clinical materials resulting from UV irradiation with presence in these materials of anaerobes such as P. melaninogenica, P. intermedia and P. asaccharolytics was detected. Investigation of clinical materials with application of fluorescence in UV radiation lasts only 1-2 minutes and together with preparation and microscopical inspection which is Gram-stained--only 15-20 min. Positive results of this test may constitute a basis for rapid, preliminary identification of the etiologic factor and for direction of chemotherapy in the early period of infection.
Studies were performed in the years 1986-1990 and dealing with 405 clinical materials originating from infections within the abdominal cavity, maxilla, reproductive organ, post-operative wounds as well as dressings, extremities, blood, bile and other types of material and usefulness of analysis by gas chromatography of presence of lower fatty acids in materials for rapid and preliminary diagnostics with strictly anaerobic bacteria was confirmed. Presence in chromatograms of isobutyric, butyric isovaleric, valeric, isocaproic and caproic acid, and partly of propionic acid, is a good indication of infection with strictly anaerobic bacteria. Moreover, presence of succinic acid may suggest presence of anaerobic infection, similarly as presence in chromatograms of three or more various lower fatty acids. Rapid, approximative diagnosis of anaerobic bacteria by application of gas chromatography is worth of broader application. Investigation of clinical material by this method lasts only 1-2 hours and obtained results my present a basis for preliminary diagnosis of the etiological factor and for direction of chemotherapy, which is very important in most early period of treatment of infections.
The long colic nerves were investigated in 50 fetuses aged 13 to 21 weeks. The long colic nerves arise from the pelvic nerves, pelvic plexuses, hypogastric nerves, and intermesenteric plexus. The nerves may take origin from two or three sources, and in most cases they arise from the pelvic nerves. They ascend along the sigmoid and descending colons reaching the left portion of the transverse colon.
Muscle spindles were investigated in 19 human fetuses, aged 11 to 17 weeks. First spindles were observed in 11th week. The number of muscle spindles increases with advancement of the development being highest in the omohyoid muscle.
The muscle spindles were counted in 16 sternocleidomastoid muscles of human fetuses aged 12-18 weeks. The greatest number of muscle spindles were noted in the middle of the sternocleidomastoid muscle.
Sensibility (MBC) of 97 strains of non-sporing anaerobes separated from clinical materials coming from oral cavity to glutaric aldehyde (Koch Light Lab) has been examined. The experiments have been carried out by the suspension method adapting it properly to the examinations of anaerobes. 72 hours' cultures in enriched thioglycolate broth containing 10(9) of living cells of anaerobes in 1 ml have been used as inoculum. MBC results were read off after 7 days' incubation at temperature of 37 degrees C. From the total number of 97 strain 7 (7%) strains were sensible to the concentration of 156 to 311 micrograms/ml. In MBC ranges equal to 312 to 624 micrograms/ml 17 (17%) strains perished. Most of the strains namely 51 (53%) strains were damaged by the concentrations of 625 to 1249 micrograms/ml. The remaining 22 (23%) strains exacted to utilize glutaric aldehyde of the concentrations within the range of 1250 to 2500 micrograms/ml. It has been noticed that the strains from Leptotrichia buccalis species were the most sensitive ones. In the remaining of anaerobes under examinations the MBC values were similar to each other.
Sensibility (MBC--Minimal Bactericidal Concentration) of 97 strains of non-sporulated anaerobic bacteria separated from clinical material from oral cavity to Sterinole (Polfa) and cetylpirydine chloride has been examined. Experiments have been carried out by means of suspension method adapted appropriately to the tests on non-sporulated anaerobes. 72 hours' cultures of strains containing 10(9) of live bacterial cells in 1 ml have been used as inoculum. MBC readings have been performed after 7 days' incubation at temperature of (37 degrees C) 310 degrees K. From the total number of 97 strains under examinations 34 strains were sensible to low concentration of Sterinole equal to 7.8 to 15.5 micrograms/ml. Further 19 strains exacted MBC within 15.6 to 31.1 micrograms/ml and further 40 strains perished in the antiseptic concentration of 31.2 to 62.4 micrograms/ml. The remaining 4 strains exacted concentration of 62.5 to 125 micrograms/ml to be perished. Among 97 strains 29 strains were perishing in low concentrations of cetylpirydine chloride equal to 7.8 to 15.5 micrograms/ml. Successive 25 strains were damaging by the antiseptic concentration equal to 15.6 to 31.1 micrograms/ml. Further 28 strains exacted utilization of concentration equal to 31.2 to 62.4 micrograms/ml while in case of the remaining 15 strains MBC values achieved the level of 62.5 to 125 micrograms/ml. Strains from Leptotrichia buccalis and then Gram--positive anaerobic bacteria were the most sensitive ones both to Sterinole and cetylpirydine chloride while Gram--negative anaerobic bacteria and anaerobic micrococcus were less sensitive.
