Effects of vanadate on glycolytic enzymes and malic enzyme in insulin-dependent and -independent tissues of diabetic rats.

The effects of sodium orthovanadate (0.6 mg/ml in drinking water) on hexokinase isozymes, pyruvate kinase and malic enzyme in liver and kidney of control and alloxan diabetic rats were studied and compared. Vanadate treatment of diabetic rats normalized hyperglycemia and almost completely restored the differentially altered enzyme profile of liver (a tissue that underutilizes glucose in diabetes) and kidney (a tissue that overutilizes glucose during diabetes). Vanadate therapy, however, could not restore the depressed plasma insulin level of diabetic rats. The study clearly indicates that vanadate can effectively normalize many metabolic abnormalities even at a low insulin level in both insulin-dependent and -independent tissues of diabetic rats.

Modulation of IgM to IgG class switch by protein A.

The Fc binding property of soluble protein A (SpA) from Staphylococcus aureus has been utilized to form IgG-SpA complexes which enabled an increase or decrease of IgG from the host depending on the dose of SpA administered. When 5 micrograms SpA was administered the IgG-SpA complexes were rapidly catabolized and, hence, low plasma IgG levels were observed. In contrast 25 micrograms SpA resulted in a significant increase in the IgG level in the host plasma. Based on these observations, the present investigation attempted to study the effect of IgG depletion/increase on the primary and secondary B-cell response to sheep erythrocyte (SRBC) antigen in Balb/c mice. Introduction of 5, 10 and 25 micrograms SpA at the time of the primary antigenic challenge inhibited both the primary IgM and the secondary IgM and IgG responses in a dose-dependent manner. Administration of 5 and 10 micrograms SpA at the time of the secondary antigenic challenge enabled the host to maintain the otherwise depressed secondary IgM response equivalent to the normal primary response. In contrast, 25 micrograms SpA at the time of the secondary antigenic challenge inhibited both the IgM and IgG PFC responses. These results extend our understanding of the mechanism of switch in immunoglobulin class expression during antigen driven maturation of the B-cell response.

Effect of zinc on immune functions and host resistance against infection and tumor challenge.

The effect of zinc treatment on immune function and resistance against infection and tumor challenge was studied in mice. Swiss albino mice were treated with zinc acetate (3 mg/kg body weight) in one or two intraperitoneal injections. Various immune function assays were performed in treated animals. Zinc treatment to normal animals caused potentiation of T-lymphocyte and macrophage functions. Zinc treatment was also found to increase host resistance against Candida albicans and Semliki Forest virus infections. Increased resistance against endotoxin shock and Ehrlich's ascites tumor challenge was also observed in zinc treated animals. It can be stated from this study that zinc treatment potentiates the cell mediated immunity and host resistance against infection and tumor challenge.

Incidence of phthirapteran infestation upon the buffaloes of Dehradun (India).

Out of 373 buffaloes examined in different localities of Dehradun, 60.58% have been found infested with Haematopinus tuberculatus. Incidence of infestation was higher on adults than on youngs. The lice index was highest in January and lowest in June. The adults and nymphs preferred neck, back, ribs, shoulder, belly, thigh, hind, flank, head, hind legs, tail, fore legs and fore flank in this order of decreasing frequency. On the other hand eggs were found most heavily concentrated on neck and back regions.

Incidence of Mallophaga on poultry in Dehradun (India).

The order of abundance of different mallophagan species on poultry birds in Dehradun (India) has been found to be Menopon gallinae greater than Menacanthus cornutus greater than Menacanthus stramineus greater than Goniocotes gallinae greater than Goniodes dissimilis greater than Lipeurus caponis greater than Lipeurus lawrensis tropicalis greater than Goniodes gigas. The intensity of these species upon 1249 birds has been recorded by coding system. The correlation between the monthly incidence of different species and environmental temperature as well as R.H. has been recorded. The degree of association between four most commonly occurring combinations has also been analysed.

Effect of vanadate administration on polyol pathway in diabetic rat kidney.

Effect of oral administration of sodium orthovanadate for three weeks on polyol pathway in renal cortex and medulla was studied in control and alloxan diabetic rats. An enhancement in aldose reductase in cortex and medulla and sorbitol dehydrogenase in cortex was observed in alloxan diabetic rats. Despite depressed insulin secretion, vanadate treatment to diabetic rats counteracted hyperglycemia, normalized elevated enzyme activities and glucose level, prevented medullary sorbitol accumulation and markedly checked increase in kidney weight. These results show that vanadate causes marked improvement in renal hypertrophy and has an antidiabetogenic effect on polyol pathway in diabetic kidney.

Aortic dissection: diagnosis and management.

Acute dissections mandate surgery after the patient has been stabilized medically. Hypertension appears to be a prerequisite in the pathogenesis of aortic dissection, and control of ventricular contraction velocity may be prophylactic.

The distribution of lice (Phthiraptera) on poultry (Gallus domesticus).

The distribution of three amblyceran (Menacanthus stramineus, Menacanthus cornutus, Menopon gallinae) and five ischnoceran species (Lipeurus lawrensis tropicalis, Lipeurus caponis. Goniocotes gallinae, Goniodes gigas, Goniodes dissimilis) on poultry was determined. The preferred sites of these species on the host body were recorded.

A study on the B cell activity in protein deficient rats exposed to methyl isocyanate vapour.

The effect of MIC on the humoral immunity of the malnourished (protein deficient) subjects has been investigated. A single exposure of MIC (1.60 mg/l) on protein deficient rats showed no significant change in the body weight and mortality rate compared with the normal but the serum protein levels were found significantly low (P less than 0.01) in the protein deficient diet fed control (PDC) ones. Both PDC and protein deficient MIC exposed (PDMIC) rat showed diminished B-cell proliferation with the optimal dose of LPS, compared to the NDC and normal diet fed MIC exposed (NDMIC) group. Furthermore significant suppression (P less than 0.01) in the B-cell activation by LPS was observed in the PDMIC compared to PDC. The total IgM level in PDMIC was 43% less while 26% higher in NDMIC compared to NDC. The total IgG level in PDMIC and NDMIC was higher (20%) compared to NDC, while 25% less in PDC. The antigen specific B-cell immunity was affected in PDMIC, PDC and NDMIC. As the terminal differentiation process of B-cells were found equally affected in both PDC and NDMIC, it appears that MIC has no synergistic effect on the humoral immunity of the protein malnourished host.

Binding of thiocyanate and cyanide to manganese(III)-reconstituted horseradish peroxidase: a 15N nuclear magnetic resonance study.

Binding of thiocyanate and cyanide ions to Mn(III) protoporphyrin-apohorseradish peroxidase complex [Mn(III)HRP] was investigated by relaxation rate measurements (at 50.68 MHz) of 15N resonance of SC15N- and C15N-. At pH = 4.0 the apparent dissociation constant (KD) for thiocyanate and cyanide binding to Mn(III)HRP was deduced to be 156 and 42 mM, respectively. The pH dependence of the 15N line width as well as apparent dissociation constant for thiocyanate and cyanide binding were quantitatively analyzed on the basis of a reaction scheme in which thiocyanate and cyanide in deprotonated form bind to the enzyme in a protonated form. The binding of thiocyanate and cyanide to Mn(III)HRP was found to be facilitated by protonation of an ionizable group on the enzyme [Mn(III)HRP] with a pKa = 4.0. From competitive binding studies it was shown that iodide, thiocyanate and cyanide bind to Mn(III)HRP at the same site; however, the binding site for resorcinol is different. The apparent dissociation constant for iodide binding deduced from competitive binding studies was found to be 117 mM, which agrees very well with the iodide binding to ferric HRP. The binding of thiocyanate and cyanide was shown to be away from the metal center and the distance of the 15N of thiocyanate and cyanide from the paramagnetic manganese ion in Mn(III)HRP was found to be 6.9 and 6.6 A, respectively. Except for cyanide binding, these observations parallel with the iodide and thiocyanate ion binding to native Fe(III)HRP. Water proton relaxivity measurements showed the presence of a coordinated water molecule to Mn(III)HRP with the distance of Mn-H2O being calculated to be 2.6 A. The slow reactivity of H2O2 towards Mn(III)HRP could be attributed to the presence of water at the sixth coordination position of the manganese ion.

Platelet serotonergic mechanisms in ischaemic heart disease.

The uptake, basal content and efflux of serotonin (5-HT) from the platelets of patients of ischaemic heart disease (I.H.D.) was studied and compared with normal platelets. A significant increase in uptake and basal content of 5-HT as well as increase in 5-HT efflux was observed as compared to control. Our data indicate that I.H.D. is associated with platelet serotonergic dysfunction which seems to indicate that the platelets are preactivated in I.H.D.

Effects of aflatoxin on lymphoid cells of weanling rat.

Aflatoxin (AF), the hepatocarcinogenic food contaminant produced by the Aspergillus flavus group of fungi, is known to interact with various vital processes, including the immune function. Effects of long-term treatment of three dose levels of aflatoxin B1 (AFB1) on lymphoid cells of weanling rats were studied. AFB1 treatment caused a reduction in body weight gain, significantly (P less than 0.01) at the 700 microgram level. There was also a significant decrease in the weight of spleen and thymus in AFB1-treated animals in comparison to control. Similarly, AFB1 depleted cell populations of thymus and bone marrow and WBC and RBC counts. There was a marked reduction in the population and phagocytic capacity of macrophages due to AFB1 administration at dose levels of 350 and 700 micrograms kg-1 body weight. Macromolecular synthesis of DNA, RNA and protein in macrophages was affected, as there was significant inhibition in the incorporation of [3H]-thymidine, [3H]-uridine and [3H]-leucine. The hampered functioning of macrophages may be due to the cytotoxic action of AFB1.

Protection against carbon-tetrachloride-induced lymphoid organotoxicity in rats by protein A.

In a previous study we described protection of rats from carbon tetrachloride (CCl4)-induced hepatotoxicity by protein A (PA). In the present report we described protection against CCl4-induced lymphoid organotoxicity in rats by PA. Our data indicate that CCl4 administration produced a significant increase in the number of total leukocytes and polymorphs in blood, a significant decrease in leukocyte count in bone marrow, and a significant loss in weight of spleen and adrenals. In the animals receiving PA prior to and after CCl4, these values were found to be near those of controls. Histological examination revealed thymocyte depletion in thymic lobe and degenerative changes of tissue in spleen of CCl4-exposed animals. PA treatment to CCl4-exposed animals exhibited improvement and recovery from the damage caused by CCl4. These observations throw a new light on the field of CCl4 toxicity to lymphoid organs and their protection by PA.

Synthesis, biological evaluation, and quantitative structure-activity relationship analysis of [beta-(Aroylamino)ethyl]piperazines and -piperidines and [2-[(Arylamino)carbonyl]ethyl]piperazines, -pyrazinopyridoindoles, and -pyrazinoisoquinolines. A new class of potent H1 antagonists.

Some [beta-(Aroylamino)ethyl]piperazines and -piperidines and [2-[(Arylamino)carbonyl]ethyl]piperazines, -piperidines, -pyrazinopyridoindoles, and -pyrazinoisoquinolines have been synthesized and their H1-antagonistic activity studied in isolated guinea pig ileum. Quantitative structure-activity relationship analysis indicates that the hydrophobicity of the side chain of these compounds plays a major role in their activity while steric and electronic factors are of secondary importance. All these compounds act on a common receptor and appear to interact similarly with the receptor.

Protein A induced abrogation of cyclophosphamide toxicity is associated with concomitant potentiation of immune function of host.

This report confirms our previous observation that protein A (PA) of Staphylococcus aureus Cowan I reduces the cyclophosphamide (Cy) induced toxicity. PA treated animals recover quickly from the toxic effects of Cy. We have exhaustively studied the role of specific and nonspecific immunity in the protection of the animals. It was observed that PA helped the animals in the accelerated regeneration of leukocytes of blood (p less than 0.001) and different lymphoid organs like thymus (p less than 0.001), spleen (p less than 0.01) and bone marrow (p less than 0.01). Increased number and function of macrophages was also observed in PA (p less than 0.001) and PA+Cy (p less than 0.001) groups. PA, on one hand enhanced the cell mediated immunity while suppressed the humoral immunity as was assessed by increase in delayed type hypersensitivity response (p less than 0.001) and decreased in plaque forming cells (p less than 0.001), EAC-rosettes (p less than 0.001), hemagglutination (p less than 0.001) and hemolysin titre (p less than 0.05). On the basis of above observations we propose that the immunomodulatory activity of PA helped the animals to remain alive in two ways- (1) by early generation of the cells depleted by the Cy thus helping animals to repair the damaged immune system and fast clearance of the toxic metabolites of Cy (2) by temporarily suppressing the cells responsible for humoral immunity which are more susceptible to Cy metabolites.
