Retroviral capture of c-erbB proto-oncogene sequences: rapid evolution of distinct viral genomes carrying mutant v-erbB genes with different transforming capacities.

The evolution of oncogene-transducing retroviruses was followed by studying the genomes of five new, erbB carrying retroviruses. These viruses, isolated from cells of one chicken infected with Rous Associated virus 1 (RAV-1), had captured c-erbB sequences as a consequence of RAV-1 integration into the host genome. Their genome structures were distinct; however, their v-erbB genes had sustained identical 5' and 3' deletions and the v-erbB-env junctions were identical at the nucleotide level. The results therefore strongly suggest that all five viruses originate from the same capture event. Sequence analyses of the v-erbB genes from three of these viruses revealed that one of them had undergone no further mutation and lacked detectable capacity to transform cells, therefore probably representing an 'early' form of transducing virus. The two other v-erbB genes contained distinct mutations and differed in their potential to induce fibroblast- and erythroblast transformation; they therefore probably represent later derivatives of the virus that captured the erbB oncogene. The data suggest that the initial retrovirus rapidly underwent many alterations after capture of c-erbB sequences, already in the RAV-1 infected bird as well as during subsequent in vitro isolation procedures. The changes involve both major rearrangements of the genome as well as point mutations that activated the erbB oncogene.

Perturbations of plant leaflet rhythms caused by electromagnetic radio-frequency radiation.

The minute-range up and down rhythms of the lateral leaflets of Desmodium gyrans has been studied when exposed to electromagnetic radiation in the radio-frequency (RF) range. The RF radiation was applied as homogeneous 27.12 MHz fields in specially-designed exposure cells(and in some cases as non-homogeneous radiation of 27 MHz, amplitude modulated by 50 Hz, in front of commercial diathermy equipment). All fields were applied as pulses. We report effects in the leaflet rhythms such as temporary changes in the amplitude, period, and phase. The radiation could also cause temporary or complete cessations of the rhythms. The lowest dose (8 W/cm2) used was still effective.

Measurement of the intracellular pH of Propionibacterium acnes: comparison between the fluorescent probe BCECF and 31P-NMR spectroscopy.

The intracellular pH of the Gram-positive skin bacterium Propionibacterium acnes was determined using the pH-sensitive fluorescent probe 2',7'-bis-(2-carboxyethyl)-5-(and-6)carboxyfluorescein (BCECF). The probe was introduced into the bacteria using the membrane-permeable acetoxymethyl ester BCECF-AM. The intracellular pH of the bacteria was determined by establishing a relation between the fluorescence ratio 505/450 and pH using the ionophore nigericin. To verify the intracellular pH determined using BCECF, the results were compared with those obtained using 31P-NMR spectroscopy. The effects of different external pH values and glucose addition upon the intracellular pH were examined using BCECF and 31P-NMR. Good correlation was obtained between the two techniques. Propionibacterium acnes maintained a pH gradient, inside alkaline, in the external pH range 5.0-7.4, which inverted when the pH was > 7.5. At external pH > or equal to 8.5, the intracellular pH was close to the external pH. Glucose exposure did not affect the intracellular pH. Rapid, transient intracellular acidification and alkalinization brought about using NaHCO3 and NH4Cl, respectively, could be detected using BCECF. A limitation encountered when using BCECF was BCECF leakage, which could significantly affect the results if not taken into account.

Primary gastric non-Hodgkin's lymphoma. A retrospective clinico-pathological study.

Prognostic factors and treatment results were analysed in 72 consecutive patients with primary gastric lymphoma treated between 1970 and 1985. There were 37 patients in stage IE, 17 in IIE, 3 in IIES and 15 in stage IV. Histopathological re-evaluation and classification according to the TNM system were performed. We found that disseminated disease (stage IV), serosal penetration (T3), involvement of adjacent organs (T4) and extensive abdominal lymph node involvement (N3) were poor prognostic factors. Neither histological malignancy grading, nor the appearance of lympho-epithelial lesions were significantly associated with relapse-free survival. Forty-six patients with 'limited localized' disease (stage IE, IIE, N3 excluded) received potentially curative treatment (surgery, radiotherapy, chemotherapy or combinations thereof), of whom 85% remained relapse-free. Thirty-four patients did only get local treatment (surgery and/or radiotherapy) with curative potential, the relapse-free survival rate was 85%. We conclude that primary gastric lymphoma stage IE and IIE (N3 excluded) is often a truly localized disease that can be cured with local therapy.

Low level laser versus placebo in the treatment of tennis elbow.

The effect of low level laser (GaAs) on lateral epicondylitis was investigated in a double-blind, randomized, controlled study. Thirty patients were assigned equally to a laser (n = 15) or a placebo laser (n = 15) group. All patients received eight treatments and were evaluated subjectively and objectively before, at the end of, and four weeks after treatment. Patients also completed a follow-up questionnaire on an average of five to six months after treatment. A significant improvement in the laser compared to the placebo group was found on visual analog scale (p = 0.02) and grip strength (p = 0.03) tests four weeks after treatment. In this study low level laser therapy was shown to have an effect over placebo; however, as a sole treatment for lateral epicondylitis it is of limited value. Further studies are needed to evaluate the reliability of our findings and to compare laser to other established treatment methods.

Comparison of changes in testosterone concentrations after strength and endurance exercise in well trained men.

Changes in the testosterone concentrations after single sessions of endurance and strength training were measured in seven well trained men, experienced in both forms of training. Both training sessions were rated as hard to very hard on the Borg scale. Blood samples for testosterone measurements were taken before, immediately after, and 2, 4 and 6 h after the training sessions as well as the next morning. The mean testosterone concentration increased 27% (P less than 0.02) and 37% (P less than 0.02) during the strength and endurance training session, respectively. Two hours after the training sessions the mean testosterone concentration had returned to the pre-training level and remained at that level for the length of the observation period. There were no significant differences in the changes in testosterone concentration after strength and endurance training but there were large differences in the testosterone response at the level of the individual. A high correlation (r = 0.98; P less than 0.001) for individuals was found between increases in testosterone concentration after strength and after endurance training. It was concluded that the changes in mean testosterone values followed the same timecourse after single sessions of strength and endurance training of the same duration and perceived exertion. The interindividual differences in testosterone response may be of importance for individual adaptation to training.

Near-UV-induced radicals in Propionibacterium acnes, studied by electron spin resonance spectrometry at 77 K.

Suspensions of Propionibacterium acnes were UV irradiated and the induced radicals were measured at 77 K by electron spin resonance (ESR) spectrometry. Two types of radical were formed during irradiation and stabilized in the frozen suspensions. The relative yield of each radical was studied as a function of irradiation wavelength. The first radical, which was a singlet with a peak-to-peak width of 20 G, was insensitive to the deoxygenation of the samples and to the exchange of solvent water by heavy water. The action spectrum was similar to the absorption spectrum of NADPH. The second type of radical was not formed in deoxygenated samples and the shape of the ESR spectrum was characteristic of the superoxide radical. This radical was only formed at wavelengths below 340 nm.

Tropistic responses of Avena seedlings in simulated hypogravity.

Results from investigation of the gravitropic responses of the Avena coleoptile under simulated weightlessness are presented. The tests were conducted using the flight hardware, identified by NASA as the Gravitational Plant Physiology Facility (GPPF), designed to support the Spacelab experiment, GTHRES. Weightlessness or so-called microgravity-conditions were simulated by the use of a so-called somersault clinostat. The plants were gravitropically stimulated with different g-forces and stimulation durations on a centrifuge. Timelapse video pictures of the plants were taken during the last hour prior to the stimulation and in a period of 3 hours after. The gravitropic responses of the plants were analyzed with the aid of an image analyzing system. Dose-response curves for stimulation forces 0.2, 0.4, 0.6, 0.8, and 1.0 g with different stimulation times ranging from 2 to 250 min were achieved. The results show that the reciprocity between force and stimulation time may be valid for small stimulation doses (lower than 5 g min), but not for larger doses. Furthermore, the threshold values (i.e. the smallest stimulation the plants are able to detect) found by extrapolation of the dose response curves, are less than 30 g s. The results are discussed with respect to an accepted space experiment, GTHRES, where the corresponding experiments will be carried out in the same apparatus under true microgravity conditions.

Deletion of 14q in non-Hodgkin's lymphoma.

6 patients with non-Hodgkin's lymphoma [3 with small cell lymphocytic lymphoma of B-cell type (SL), and 1 each with follicular centroblastic/centrocytic, centroblastic, and immunoblastic lymphoma] and with the acquired cytogenetic abnormalities del(14) (q22) or del(14) (q24) are described. An evaluation of these 6 cases and 41 other lymphatic neoplasms with 14q deletion known from the literature revealed that 37 had a breakpoint in bands q22 to q24. The deletions occur significantly more often in lymphomas of SL morphology and in the leukemic counterpart, chronic lymphocytic leukemia, than in other types of lymphatic malignancies (p less than 0.001).

Rhythmic biological systems under microgravity conditions.

Rhythmic phenomena in biology cover a wide frequency spectrum. In Space, the rhythms will encounter microgravity conditions which can, therefore, be a valuable tool for their understanding. A review and discussion of important effects of gravity/absence of gravity on biological systems will be given. Convection will be emphasized as a mechanism which is drastically reduced in Space. Microgravity might also affect the coupling between individual oscillators in a multi-oscillatory system. The environmental interference with rhythms will be discussed with a simple feedback as a starting point. Model simulations will be presented and clinostat and microgravity-conditions will be discussed in a specific case, viz. the gravitropical system of plants which can show sustained oscillations.

A major part of platelet-derived growth factor purified from human platelets is a heterodimer of one A and one B chain.

Platelet-derived growth factor, PDGF, purified from human platelets is a disulfide-bonded dimer consisting of two homologous polypeptide chains denoted A and B; it has not been known whether it is a heterodimer or a mixture of homodimers. We present here evidence that a major part of PDGF has a heterodimer structure. A highly homogeneous, 31-kDa PDGF was purified in the presence of protease inhibitors and shown to contain both chains by means of immunoprecipitations with peptide antisera specific for the A and B chains, respectively. The susceptibility of PDGF to mild acid treatment and its chromatographic behavior in reversed-phase high performance liquid chromatography and immobilized metal ion affinity chromatography, as compared to A and B chain homodimers, is consistent with a heterodimer structure. Analysis of PDGF purified according to our routine, large scale procedure revealed the major part to have a heterodimer structure. In addition, B chain homodimers were also found. With the demonstration that a major part of PDGF purified from human platelets occurs as a heterodimer, all three dimeric forms of PDGF have been identified. The following nomenclature to distinguish the various forms is suggested: PDGF-AA, a homodimer of A chains; PDGF-AB, a heterodimer; PDGF-BB, a homodimer of B chains; PDGF, any dimeric form of A or B chains.

Expression of multiple growth factors in a human lung cancer cell line.

U-1810, a human large-cell lung cancer line, was found to express a PDGF-like growth factor. 35S-cysteine labelling and immunoprecipitation revealed the synthesis and secretion of a 31-kDa PDGF-like protein. Serum-free conditioned medium contained PDGF-receptor-competing and mitogenic activity when tested on human fibroblasts. Whereas the receptor-competing activity was fully neutralized by anti-PDGF antibodies, the mitogenic activity was only partially affected. We therefore probed U-1810 mRNA with a panel of growth-factor DNA clones. We found expression of the genes for PDGF A- and B-chains, TGF-alpha, TGF-beta and IGF-II but not EGF or IGF-I. U-1810 cells lacked specific binding sites for PDGF but showed specific binding of EGF and expressed EGF-receptor transcripts. Thus, U-1810 is an example of a human tumor cell line that expresses multiple growth factor genes; in the intact tumor the corresponding growth factors may operate in autocrine stimulation of the tumor cells as well as in paracrine growth reactions (i.e. stroma recruitment).

A 31P-NMR study of Propionibacterium acnes, including effects caused by near-ultraviolet irradiation.

161.8 MHz 31P-NMR spectra were recorded from the light sensitive skin bacterium Propionibacterium acnes. The cells were grown anaerobically on synthetic phosphate-buffered Eagle's medium or on a complex yeast extract medium. The spectra showed a large accumulation of polyphosphates when grown on Eagles medium. A splitting of the inorganic phosphate peak indicated a difference between internal and external pH of the cells. Addition of glucose to the cell suspension gave rise to a change in the pH gradient across the cell membrane, as reported for other Gram-positive bacteria. A decrease in the polyphosphate peak was observed after addition of glucose. A lethal dose of broad-band near-ultraviolet light (corresponding to a 10% survival in a survival test), increased the amount of polyphosphates visible in the NMR-spectra. The addition of glucose to irradiated cells decreased the pH in the external solution, but no splitting of the inorganic phosphate peak could however be observed. 31P-NMR can, therefore, be used to study immediate near-ultraviolet-induced effects at the cellular level, at least in the case of P. acnes.

Fluorescence from pilosebaceous follicles.

Fluorescence studies were performed on the extrusions from pilosebaceous follicles. Pressure extractions produced follicle samples which showed fluorescence under Wood's light. The samples were then analysed in a fluorometer giving corrected excitation spectra. The structured emission spectra achieved were interpreted as being due to porphyrins produced by Propionibacterium acnes (P. acnes). Details in the spectra showed close resemblance to spectra from cultured P. acnes cells. The emission spectra showed distinct features in all the four subjects investigated (who were different with respect to age, sex, follicle sampling area, and tendency to acne) and dominant peaks due to at least three porphyrins were found. The concentrations of these porphyrins vary from case to case. Excitation spectra were recorded and supported the assumption that the fluorescent emission was partly due to coproporphyrins and metalloporphyrins in the samples. Free protoporphyrins did not seem to be present in the extrusions. The excitation spectra, in particular, vary from person to person but seem to be constant over time in one and the same subject.

Influence of the g-force on the circumnutations of sunflower hypocotyls.

Circumnutations of hypocotyls of sunflower (Helianthus annuus L. cv. Californicus) were studied under 1 g and 3 g conditions. Root mean square values of the hypocotyl deviation from the plumbline and period of the movements were determined from calculations of the autocorrelation functions of the movements. The amplitude and the period of the circumnutations increased under 3 g as compared to 1 g. A transition from 3 to 1 g or vice versa also caused changes in period and amplitude of the movements. The results are interpreted as a support for the idea that gravity influences the circumnutation parameters in this sunflower variety. A comparison is made with published results on the dwarf sunflower cv. Teddy Bear where the force influence is very small or negligible. Simulations of a model for circumnutations show movements which are in qualitative agreement with the experimental results, provided adaptation to g-levels is included in the model. Finally, the results are discussed with the recent Spacelab-experiment (SL1) as a background.

Trilateral malignant lymphoma: primary malignant B-cell lymphoma of the eyes and brain. Diagnosis by transvitreal retinochoroidal biopsy.

A 59-year-old woman with therapy-resistant bilateral involvement of posterior uvea and retina of some months' duration was examined. Diagnostic vitrectomy was inconclusive, but subsequent transvitreal retinochoroidal biopsy gave ample material for the immunohistopathological diagnosis of a malignant B cell lymphoma. Despite systemic and intrathecal cytostatics and ocular X-ray therapy, central nervous system involvement was observed 2 months later. The ocular lesions regressed after treatment and have not recurred.
