Proliferation of glycogen accumulating organisms induced by Fe(III) dosing in a domestic wastewater treatment plant.

To meet the effluent requirements given for the sensitive receiving body, the Southpest Wastewater Treatment Plant of Budapest, Hungary uses a combined activated sludge-biofilter system with chemical precipitation for P removal. Causes of the proliferation of glycogen accumulating organisms (GAOs) observed in the unaerated/oxic activated sludge unit of this system were investigated both in full-scale and in lab-scale experiments combined with a detailed analysis of the microbial communities. Concentration profile measurements throughout the 8-stage activated sludge unit indicated anaerobic conditions in the first two unaerated reactors and low orthophosphate level (< 1 mg l(-1)) in all of the stages that could not be attributed to the influent quality, but to Fe (III)-dosing to the returned activated sludge. Microbiological analysis revealed the presence of GAOs from the GB group in the Gammaproteobacteria and occasionally tetrad-forming organisms from Actinobacteria, and the absence of Rhodocyclus-related polyphosphate accumulating organisms (PAOs) in the activated sludge samples. Comparative lab-scale studies carried out in two identically arranged UCT-systems with staged anoxic reactors also confirmed that Fe (III)-dosing may result in phosphorus deficiency of the microbial niche, leading to the suppression of growth and EBPR activity of PAOs and to the proliferation of GAOs.

Enhanced nitrogen removal in the combined activated sludge-biofilter system of the Southpest Wastewater Treatment Plant.

In 1999 the existing activated sludge unit of the Southpest Wastewater Treatment Plant was supplemented by a two-stage biofilter system aiming for nitrification and post-denitrification. In this arrangement excess biomass of the filters is wasted through the activated sludge unit, facilitating backseeding, and recirculation of the nitrate-rich effluent of the N-filter serves for decreasing the methanol demand of the DN-filter and for saving aeration energy at the same time. The paper reports on the development of an ASM1-based mathematical model that proved to be adequate for describing the interactions in the combined system and was used to compare the efficiency of different treatment options. Full-scale results verified that backseeding may considerably improve performance. However, nitrification ability of the activated sludge unit depends on the treatment temperature and, if unexpected, can be limited by insufficient oxygen supply. The upgrading possibilities outlined may serve as a new perspective for implementation of combined activated sludge-biofilter systems.

Implementation of glycogen accumulating bacteria in treating nutrient-deficient wastewater.

Activated sludge treatment of nutrient-deficient wastes may lead to severe slime formation and consequent biomass separation difficulties. The purpose of this paper has been to show that bioreactor arrangement essentially influences the manner of biological excess carbon removal. In a comparative lab-scale experiment two differently arranged activated sludge systems were operated simultaneously: an aerated CSTR with an aerobic selector and an aerated CSTR with an anaerobic selector. The seed derived from an anaerobic/aerobic activated sludge plant of a winery. The model wastewater contained wine, sugar and acetic acid as organic carbon sources and lacked nutrients regarding both N and P, similarly to the influent of the full-scale plant. During the 52 days of the experiment the SVI values of the fully aerated system increased up to 600-800 cm3 g(-1) whereas those of the anaerobic/aerobic system remained below 250 cm3 g(-1) The SVI values showed a strict correlation with the amount of extracellular polysaccharides. In the anaerobic/aerobic experimental system, the high (40% of MLSS) intracellular polysaccharide content of the seed could be maintained. Besides the analytical data, also the microscopic observations of the biomass structure referred to the presence of glycogen accumulating organisms in both the lab- and full-scale anaerobic/aerobic systems.

Evaluation of respirometric data: identification of features that preclude data fitting with existing kinetic expressions.

The use of respirometric for from the evaluation of intrinsic biodegradation kinetic parameters for single organic compounds is discussed. Emphasis is placed on the preliminary assessment of the data set to determine whether it is suitable for kinetic parameter estimation. Careful preliminary examination of the data avoids attempting parameter estimation with unacceptable data. Furthermore, the use of unbiased respirometric data helps ensure that the estimated parameters truly reflect the intrinsic kinetics for biodegradation of a single substrate by the culture tested. Both experimental and theoretical oxygen uptake curves are used to illustrate how various conditions can limit the utility of a given data set. The effect of substrate inhibition, dual or multiple substrate limited growth, inaccuracies in the initial conditions assumed for curve fitting, and the use of poorly acclimated cultures are discussed. Techniques are presented which allow identification of whether a data set is unsuitable and should not be used for parameter estimation. In addition, experimental procedures which can help avoid the collection of aberrant data are discussed.

[HIV seropositivity in promiscuous heterosexual, homosexual and bisexual men and patients voluntarily reporting for screening in Hungary between 1 January 1986 and 31 December 1989].

The cumulative seroprevalence was 5.21% among the homo/bisexual men in the four years. As well as in Budapest, we experienced the increase of seroprevalence in the country, too, after a decrease in 1988. The cumulative seroprevalence was 0.44% with men and 0.04% with women in the group of the voluntary reporters in the mentioned period. The same indexes were 0.07% for men and 0.019% for women in the group of the STD patients. Seroprevalence with men increased significantly in the last year. Hungary belongs to the low-incidence area among the European countries, even though the group of homosexuals can be considered moderately infected, seroprevalence is increasing in Budapest as well as in the country and heterosexuals are also affected.

Surface immunoglobulins of tonsil lymphocytes.

Human tonsillar lymphocytes bound 14C-labelled homologous IgG, IgA, and IgM at 5 degrees C and released them at 35 degrees C. B cells were found to be more active in this type of Ig binding. The amount of lymphocytes which could be labelled with fluoresceinated anti-human Ig-s decreased during preincubation of the cells at 35 degrees C. Intrinsic Ig-s (determinants of lymphocytes) were demonstrated by immunofluorescent staining preceded by preincubation of the cells at 35 degrees C for 60 min. The ratios of IgG, IgA, and IgM bearing cells in the human tonsillar lymphocyte population were 16.0, 8.0 and 7.4% respectively. The weak binding of Ig-s to the surface of lymphocytes is discussed with regard to the demonstration of surface Ig determinants of lymphocytes.

[Bouquet qualification of wines by gas chromatography. 1. Experimental method for gas phase analysis].

In the course of our investigations we tried to find a method for the bouquet qualification of wines by gas chromatography. For this purpose, instead of aroma research studies requiring complex equipment and laboursome precise laboratory work, we developed a method for head space analysis functioning with simple experimental instruments and being sufficiently fast for serial routine work. By the method developed, the head space of a great number of samples taken from 7 different wine-types have been successfully examined. Errors of measurement were checked by standard deviation analysis and the values obtained (3,23%) were found satisfactory. According to the above, our head space analysis may provide a satisfactory basis for elaborating a method for the instrumental bouquet qualification of wines.

[Bouquet qualification of wines by gas chromatography. 2. Development of measuring unit for bouquet qualification by determination of relationship of chromatographic peak areas and conditions of application].

In a previous paper we described a method by which routine head-space analysis of wines can be performed with sufficiently accurate results by means of gas chromatography. From the results thus obtained, with application of organoleptic data, chemical and classical analytical parameters of aromatic changes during seasoning and storing wines as well as mathematical-statistical data of the method elaborated, a mathematical relationship has been derived between gas chromatographic peak areas characteristic of different aroma components and the bouquet of wine. Reliability of the method has been verified by the fact that, in spite of the small number of samples, the bouquet of seven different types of wine could be discriminated in 95,45% probalility. The method allows arbitrary choice in the number of samples and in the degree of reliability required, in investigating the quality of a wine. For this purpose, another mathematical relationship has been set up to provide criteria for the distinction of the quality of bouquets. By this method, the bouquet of various types of wines or within an identical type, the bouquet of different vintages and/or bottlings can be determined.

Comparison of chemical and immunological characteristics of some commercially obtainable conjugates.

A method is suggested for the chemical and immunological characterization of commercially obtainable conjugates. 19 antihuman IgG; IgA; IgM and beta1A-beta1C conjugates of various origine were compared by means of the method. The most surprising characteristics of the samples were the great differences in that quality. Experiments carried out with conjugates of poor quality will produce doubtful or contradictory results. Reliable characterization of conjugates is an essential and important part of immunofluorescent examination.
