Aldehyde content of collagen from liver of rats with collagen-like syndrome.

Collagen was obtained from liver tissue of rats with hydralazine-induced collagen-like syndrome. Bacterial collagenase was used to solubilize the collagen, and the aldehyde content of this material was measured using N-methyl benzothiazolone hydrazone. The aldehyde content was decreased in the in the collagen from livers of rats with collagen-like syndrome. The changes in hepatic collagen may be analogous to the effect of aging on collagen, wherein collagen cross-linking is strengthened although qualitative changes in cross-linking may lower the measurable aldehydes.

Collagen metabolism in patients with myocardial infarction.

The healing of myocardial infarction is accompanied by an increased accumulation of collagen in the heart muscle. Indexes of collagen metabolism (serum hydroxyproline, hydroxylysine and collagen-like protein concentration) were measured in 60 patients with myocardial infarction and 30 healthy blood donors. Determinations were carried out 1, 2, 3 days after admission and between the 10th-14th and 21st-25th day of the illness. It was found that hydroxyproline and hydroxylysine decreased in the first days of the disease, and subsequently increased in the course of the healing process accompanied by an increased collagen-like protein level. The changes did not depend on the total protein concentration.

[Differences in serum zinc and copper concentrations between a group of health subjects and a group of patients].

A review of the results published up to now of the determinations of serum zinc and copper levels showed that the criteria of selection of the groups of healthy subjects were lacking precision and rather arbitrary. The authors determined serum concentrations in 112 of healthy subjects fulfilling the individual criteria. No significant differences were found between the concentrations of these metals in the serum in this group of subjects and the mean concentration of these metals reported in the investigations quoted in the available literature. Serum copper concentration in patients was significantly higher than in healthy controls, while serum zinc concentration in the patients was not significantly different from that determined in healthy subjects.

Investigations upon collagen metabolites in blood serum and urine of patients with acquired valvular heart disease.

In 45 patients suffering from acquired valvular heart disease (a.v.h.d.) and in 20 controls blood serum levels of copper (Cu2+), ceruloplasmin (Clp), hydroxyproline (Hypro), hydroxylisine (Hyliz), collagen-like protein (Col-p), and daily urine excretion of Cu2+, Hypro, Hyliz and glycosamino-glicans (GAG) were determined. The comparison of the obtained mean values has shown that blood serum levels of Cu2+, Clp, Hypro, Hyliz, Col-p in the studied patients were statistically significantly higher than those found in controls, and the values of daily urine excretion of Cu2+, Hypro, Hyliz, GAG behaviour were similar. There were no significant differences between active and inactive processes and between infective and rheumatic endocarditis excluding GAG urine excretion. These elevated collagen degradation products provide evidence for disturbed collagen metabolism and confirm our previous biochemical, histopathological and ultrastructural examinations of connective tissue in patient with a.v.h.d. They also indicate that general collagenopathy might be the primary cause of a.v.h.d.

Metabolism of hydralazine by rat hepatocytes.

The metabolism of hydralazine and its pyruvic acid hydrazone was studied in a medium containing isolated adult rat hepatocytes. Hydralazine undergoes conversion to 3-methyltriazolophthalazine, while hydralazine pyruvic acid hydrazone is not metabolized in rat hepatocytes.

Zinc and copper in human milk.

Studies of zinc and copper contents of breast milk were performed in 34 mothers during their lactation period until 5 days postpartum. They experienced no problems during pregnancy and delivery. All women had full term healthy infants, 22 were multiparous and 12 primiparous. The highest zinc concentration was found in the colostrum, and subsequently the concentration declined. Copper content was stable during the 5 days of lactation. Significantly lower zinc concentration and higher copper concentration were found in the serum of lactating women 3 days post partum, when compared with non-lactating women. Factors including lactation history, age of lactating women, serum levels of zinc and copper did not affect the contents of these trace minerals in breast milk.

[Prevention of atherosclerosis with the help of antioxidants].

Recent research findings have suggested a role for pharmacology as well as nutritional antioxidants in the prevention of atherosclerosis. Data from animal studies as well as culture experiments have shown that probucol, which has hypocholesterolemic and antioxidant properties, is able to prevent oxidative modification of low density lipoproteins. Such modification is now believed to play a major part in the initiation and progression of arterial lesions. Nutrients with antioxidant properties such as vitamins C and E, beta-carotene, and monounsaturated fatty acids (when they replace polyunsaturated fatty acids) can reduce the susceptibility of LDL to oxidation. Antioxidant therapy, if proven useful, should be considered an adjunct to lipid-lowering therapy in order to have the greatest impact on coronary heart disease.

[Assessment of anaerobic metabolism in central nervous system tissues in infants with intractable hypertension].

Analogously to the blood lactate levels in conditions caused by impairment of peripheral circulation the authors sought a correlation between the concentrations of lactate, glucose and lactate dehydrogenase in the cerebrospinal fluid in children with hydrocephalus and the values of intracranial pressure as an important factor impairing oxygen supply to central nervous system tissues.

[Zinc deficiency in children infected with Giardia lamblia].

The group of children infested with Giardia intestinalis consisted of 52 children aged from 3 to 10 years. The infestation was diagnosed on the basis of duodenal contents examination. The control group comprised 50 children in whom no infestation with intestinal parasites was found. Mean age of children in both groups was not different. Disturbances were found in zinc metabolism in children with Giardia intestinalis infestation. Mean serum zinc concentration in those children was 58% of serum zinc concentration in children in the control group. Similarly, zinc concentration in the erythrocytes of children in the infested group was lower and was 64% of the concentration in children in the control group. Elimination of zinc with urine in infested children was lower than in the control group, and was 36% of the level in non-infested children. Mean concentrations of serum zinc carriers, total protein, albumin fraction and transferrin were not differing between both studied group of children. Also the concentration of picolinic acid--a zinc absorption factor in the gastrointestinal tract--in duodenal contents was similar in both groups. The above mentioned results show that in children infested with Giardia intestinalis a decrease occurs of zinc absorption from the gastrointestinal tract which causes zinc deficiencies in the those children.

[Zinc deficiencies in children after discontinuation of breast feeding].

Zinc is a component or activator of many enzymes, especially enzymes participating in biosynthesis processes of nucleic acids and proteins. Thus the role of that microelement in the development of children is very important. The paper discusses clinical pictures of both mild and acute zinc deficiencies. Since the biological availability of zinc is much greater from mother's milk than from cow milk, thus during discontinuation of breast feeding the possibility is to be considered of occurrence of zinc deficiency.

[Selenic enzymes as factors protecting against oxygen stress].

Oxygen stress is an increase of activity of free oxygen radicals in a degree exceeding the possibility of their control by the antioxidative system of the organism. Since glutathione peroxidase was indicated as a selenium-dependent enzyme of the antioxidative system, many later studies did not confirm that function, indicating rather the participation of that protein in selenium transport. Lately, two other selenoproteins were discovered, one being a hepatic selenoprotein, participating in repairing of the damage caused by free radicals, the other being a blood serum selenoprotein which is supposed to act as a carrier. The participation of these selenoproteins in the antioxidative system of the organism is not well known.

[Malonic dialdehyde concentration in cerebrospinal fluid as an indicator of the intensity of lipid peroxidation processes in intracranial hypertension].

The authors of the study correlate malonic dialdehyde (MDA) concentration in the cerebrospinal fluid of infants with hydrocephalus with the values of intracranial pressure. The purpose of the study was verification of the usefulness of MDA concentration determination as an indicator of the processes of lipid peroxidation in CNS tissues in hydrocephalic children. The obtained results point out that the isolated MDA concentration is not a sufficiently precise marker of intracranial hypertension.
