Transformation of Acinetobacter calco-aceticus (Bacterium anitratum).

A highly efficient transformation system has been demonstrated in a strain of Acinetobacter calco-aceticus (Bacterium anitratrum). During mixed growth of various stable, unencapsulated, mutant strains, deoxyribonucleic acid (DNA) is liberated and fully encapuslated transformants can be isolated. Purified DNA preparations have been used to transform suitable recipient mutant strains for ability to synthesize capsules, ability to dispense with a growth factor requirement, and resistance to streptomycin. When the wild-type strain is deprived of its capsule, either by mechanical stripping or by mutation, the unencapsulated cells tend to form large clumped masses. A nonclumping mutant of an unencapsulated strain has been isolated. When ability to synthesize capsules is transformed into this nonclumping strain, the resultant cells no longer form chains, unlike the wild-type encapsulated strain. It appears likely that the occurrence of transformation during growth of mixed cultures, with glucose or gluconate as the carbon source, may be the result of osmotic rupture resulting from the inability of unencapsulated strains to oxidize triose phosphates as fast as they are formed. The finding of transformation in Acinetobacter may provide an additional useful organism for the study of this mode of genetic transfer since this strain grows well in a simple mineral medium containing a single oxidizable source of carbon. Furthermore, no special supplementary factors seem to be required for transformation to take place.

[Nutrition of pregnant women in rural and urban areas].

The purpose of the study was to gain information on the present state of nutrition of pregnant women in rural areas and towns, and on the level of their knowledge of adequate nutrition. The study covered 300 women in villages and 258 in towns. The selection of women was done randomly, the method of the study was interviewing of the women. The questions concerned consumption of milk, cheese, meat and its products, eggs, vegetables and fruit during the whole pregnancy, and separately the data on potato consumption were obtained. The women were questioned also about the read publications on rational nutrition of pregnant women. The answers were obtained from mothers aged 18-42 years in labour rooms, clinics for children or in homes. The answers were noted during interviews by experienced interviewers. It was found that 26.6% of pregnant women in towns and 16.2% in villages were not consuming daily milk or cheese, while 6.2% of these women in towns and 4.3% in villages consumed no milk at all. Only 41.1% of these women in towns and 19% in villages took meat and processed pork everyday. Most frequently the women consumed meat and processed pork 4 times weekly (39.6% in towns and 60.3% in villages). Eggs were consumed daily by only 13% of women in towns and villages, and they were eaten most frequently 4 times weekly (30.2% in towns and 45.7% in villages). In a week meat, processed pork and eggs were eaten 5 to 7 times by only 43.8% of pregnant women in towns and 21.3% in villages.(ABSTRACT TRUNCATED AT 250 WORDS)

The effect of prolonged administration of disulfiram alone or in combination with ethanol on some indices of lipid metabolism in the blood serum and hepatic tissue of the rat.

Disulfiram alone or in combination with ethanol was administered orally for six weeks. Disulfiram significantly elevated the triglyceride level in the blood serum and hepatic tissue and cholesterol level in the blood serum, and depressed the content of free fatty acids in the hepatic tissue. Given together with ethanol, disulfiram increased the concentration of triglycerides and cholesterol in the blood serum and depressed the cholesterol level in the hepatic tissue. Given separately or in combination with ethanol, disulfiram disturbed the lipid metabolism. The results may indicate that chronically given disulfiram may produce in man similar disturbance and owing to that may aggravate hepatic steatosis present in 60--90% of alcoholics, in this way intensifying the damage of this organ.

[Influence of chronic intoxication with cadmium chloride on the content of some lipids in lens and vitreous body of rabbits].

The experiments were carried out on 21 rabbits, divided into two groups. The first group consisted of 10 control animals, the second one consisted of 11 animals given per os 3 times per week for 3 months 1 ml of water solution of cadmium chloride in the dose of 0.35 mg for 1 kilogram of body weight. After the animals have been killed, the content of cholesterol, triglycerides and phospholipids in their vitreous was identified with enzymatic methods, applying appropriate tests. The content of free fatty acids was estimated with the Duncombe method. The obtained results were analyzed statistically, with t-Student test. It was determined statistically that the content of all tested lipids components in the vitreous of rabbits poisoned with cadmium chloride was significantly decreased in comparison with their content in control animals. The greatest decrease was found in the level of free fatty acids and triglycerides, smaller decrease in phospholipids and cholesterol. Significant decrease of the total cholesterol level was found in the rabbits' lens. However no changes were observed in any other lipid components of the lens. The article discusses the significance of the obtained results in relation to ecological problems.
