Preparation and assay of urogastrone, an inhibitor of gastric acid secretion.

1. Urogastrone, an acid gastric secretion inhibitor extracted from human urine, was assayed in the anaesthetized rat by circulating through its stomach a succinic-propionic buffer and recording electrometrically the secretion of acid in response to carbachol, and inhibition of carbachol secretion by urogastrone.2. Quantitative assay procedures are described requiring the administration of only one or two doses of urogastrone to each rat.3. An extraction procedure for urogastrone is described based on the use of zeocarb, a cationic polystyrene resin.4. The most purified samples contained 40-80 times the amount/milligram which was just detectable by the assay method.

Synthesis of cell constituents by methane-grown Methylococcus capsulatus and Methanomonas methanooxidans.

1. A study was made of the incorporation of carbon from [(14)C]methanol by cultures of Methylococcus capsulatus and Methanomonas methanooxidans growing on methane. 2. The distribution of radioactivity within the non-volatile constituents of the ethanol-soluble fractions of the cells, after incubation with labelled substrate for periods of up to 3min, was analysed by chromatography and radioautography. 3. Over 80% of the radioactivity fixed by Methylococcus capsulatus at 30 degrees C at the earliest times of sampling appeared in phosphorylated compounds, of which glucose phosphate constituted 60%. 4. Most of the radioactivity fixed by Methanomonas methanooxidans at 30 degrees C at the earliest times of sampling appeared in serine, malate, aspartate and an unknown compound(s) tentatively suggested to be folate derivative(s). At 16 degrees C, [(14)C]methanol was fixed predominantly into serine and the unknown compound(s). 5. Extracts of Methylococcus capsulatus contain an enzyme system that catalyses the condensation of formaldehyde and ribose 5-phosphate to give a mixture consisting mainly of fructose phosphate and allulose phosphate. No similar activity was detected in extracts of Methanomonas methanooxidans. A convenient method was developed for assay of this enzyme system. 6. The enzyme system catalysing the condensation of formaldehyde with ribose 5-phosphate is particle-bound in both Methylococcus capsulatus and Pseudomonas methanica and is unstable in the absence of Mg(2+). 7. Extracts of Methanomonas methanooxidans contain high activities of d-glycerate-NAD oxidoreductase, whereas extracts of Methylococcus capsulatus and Pseudomonas methanica contain negligible activities of this enzyme. 8. These results indicate that during growth of Methylococcus capsulatus on methane, as with Pseudomonas methanica, cell constituents are made by the ribose phosphate cycle of formaldehyde fixation. This contrasts with Methanomonas methanooxidans, whose assimilation pathway resembles in some features that of Pseudomonas AM1 growing on methanol.

The assay of gastrin using the perfused rat stomach.

1. A method is described for the bio-assay of gastric secretagogues using the perfused rat stomach in which the acid output can be readily and accurately quantified.2. This technique increases the sensitivity of the perfused rat stomach so that 10-20 ng of synthetic human gastrin I can be detected.3. It is possible to assay secretagogues, relative to a standard with fiducial limits of less than +/-30%.4. The use of this system is illustrated by assays comparing synthetic human gastrin I with pentagastrin, pure hog gastrin and gastrin added to plasma.

Nitric oxide as a modulator of medullary pathways.

1. The present study has employed in vitro electrophysiology and chemical neuroanatomy to determine whether nitric oxide (NO) could be considered a modulator at neurons involved in medullary cardiovascular control pathways. 2. The NO donor diethylamine-NO caused concentration-related depolarizations of the rat isolated nodose ganglion preparation that were sensitive to blockade by inhibitors of soluble guanylate cyclase. 3. Incubation of rat medulla oblongata sections (50 microns) with a polyclonal antibody to NO synthase (NOS) revealed a topographic network of NOS-containing neurons. Specifically, NOS-positive neurons were found throughout the entire rostral-caudal axis of the nucleus tractus solitarius (NTS) and also in the area postrema, dorsal motor nucleus of the vagus, nucleus ambiguous, caudal and rostral ventrolateral medulla, nucleus gigantocellularis and lateral paragigantocellularis. 4. These data indicate that NO has functional actions on centrally projecting rat vagal afferent neurons that ultimately innervate the NTS. Furthermore, neurons that contain the enzyme NOS and that are, therefore, capable of using NO are localized in medullary nuclei concerned with both parasympathetic and sympathetic cardiovascular control pathways.
