Application of colour laser microscope for observing living biological specimens.

Pollen-mother cells from Lilium longiflorum, human metaphase chromosomes and human spermatozoa, were observed under a colour laser microscope. Helical mitochondrial strands were clearly evident in the middle piece of human spermatozoa. The technique enables living specimens to be conveniently and effectively examined.

New micro-glass-tube leukocyte adherence inhibition assay assessing cell adherence of mononuclear cell subpopulations defined by monoclonal antibodies.

A new micro-glass-tube leukocyte adherence inhibition (LAI) assay which is appropriate for detecting delayed type hypersensitivity in vitro has been developed for human leukocytes. Enumeration of adherent cells is replaced by a cellular radioimmunoassay determining antibody binding of the monoclonal reagents, OKT4, OKT8 and OKM1, to glass-adherent cells, fixed by glutaraldehyde or formaldehyde. An LAI reactivity to purified protein derivative of tuberculin (PPD) was detectable in donors giving a positive PPD skin test with OKT4 reagent, but not with the other two reagents.

Fibrous structures of human chromosomes observed by scanning electron microscopy.

Human chromosomes and the nucleus from cultured HeLa cells and lymphocytes were observed stereographically by scanning electron microscopy. Specimens were heated in an acetic acid solution after treatment with diluted McIlvain's buffer solution at pH 5.6. The cytoplasmic matrix adhering to the chromosomes was successfully removed, and the chromosomes were observed three-dimensionally in their original position without any significant mechanical distortion. The interphase nucleus was spherical in shape and the nuclear membrane could not be seen on its surface. The chromosome fibres were 25-35 nm in diameter and showed bumpy, looping, tortuous and knotty appearances at high magnification.

Evaluation of surgical treatments for the major lesion of bilateral silicotuberculosis.

The present communication deals with the follow-up study of 24 patients with bilateral silicotuberculosis in whom only unilateral operation was carried out for major lesions. The operative procedure consisted of pulmonary resection, thoracoplasty or combined operation such as cavernostomy, intracavitary filling of a pedunculated muscle flap and thoracoplasty. The follow-up period ranged 1 year to 12 years and 5 months. The results of surgical treatment for unilateral major lesions and their effect on the contralateral minor lesions were clinically assessed by alterations in the chest x-ray findings and tubercle bacilli in sputum. In 16 of 24 patients (67%) alleviation was obtained, whereas no change occurred in 3 (13%) and aggravation in 5 (21%). The surgical treatment for unilateral major lesion brought about 41% of improvement in the contralateral minor lesions. Contralateral minor lesions remained unchanged in 46% of patients and aggravated in 14%. This shows a value of the surgical threatment for bilateral silicotuberculosis. It should be emphasized that surgical treatments more aggressive than have been heretofore practiced can be employed.

Scanning electron microscopic observation on intracellular structures of ion-etched materials.

Ion-etching technique on the cracked surface of biological material may give a plastic visualization of intracellular structures under the scanning electron microscope, because membraneous structures in the cell are generally etch-resistant and the cytoplasmic matrix is easily ion-etched. Mild ion-etching using low voltage was applied to the cells of the pancreas. Nuclear pores were clearly disclosed but were enlarged slightly in the process of etching. Endoplasmic reticulum with ribosomes, Golgi apparatus, mitochondria, some filamentous structures and crystalline inclusions (B cell granules) were also effectively disclosed by ion-etching technique. It is necessary, however, to compare the etched specimens carefully with non-etched ones to determine whether given structures observed are intrinsic ones or artifacts caused by etching.
