[Stoichiometric protein-lipid interactions in liver microsomal membranes measured by the induction-resonance energy transfer method].

The stoichiometry of proteins and lipids in the microsomal membranes of the liver were studied by the induction-resonance energy transfer method. The energy was transferred between the fluorescent tryptophane residues of membrane proteins and fluorescent probes (pyrene and 4-dimethylaminochalcone) located in the microsomal lipid bilayer. It was assumed that the rate of protein fluorescence is correlated with protein mass. All microsomal proteins were arbitrarily divided into two groups, i.e. proteins located at a distance of more ("peripheral" proteins) and less than 30 A from the lipid bilayer surface. The former proteins made up to 14-24% of total microsomal protein, while microsomal ghosts--less than 10%. In the microsomes the bulk of other proteins is located at an average distance of about 19-26 A from the lipid bilayer surface, while the grosts--at about 4-9 A. Treatment of the ghosts with pronase results in approximation of the protein mass locus with the lipid by about 5 A. In this way the generation of ghosts and their treatment with pronase cause a loss of proteins predominantly from the peripheral parts of the microsomal membrane.

[Inhibition of lipid peroxidation by melanoprotein granules].

The effects of melanoprotein granules of the eye pigment epithelium and synthetic melanin on lipid peroxidation in rat liver microsomes and azolectin liposomes were studied. It was shown that lipid peroxidation in the microsomes and liposomes induced by ascorbate or the Fe2+ + ascorbate system is effectively inhibited by the melanoprotein granules and synthetic melanin. The enzymatic lipid peroxidation in the microsomes induced by NADPH and NADPH + Fe2+ is insensitive to the effect of the melanoprotein granules, but is inhibited by synthetic melanin. It is assumed that melanin is mainly responsible for the inhibitory action of the melanoprotein granules.

[Conformation of cytochrome b5 in soluble form and incorporated into artificial membranes].

It was shown that 35% of the peptide groups, which form the right alpha-helices, are presented in the soluble fraction of cytochrome b5. The incorporation of cytochrome b5 into egg lecithine liposomes increases the number of alpha-helices up to 51%. On the contrary, the cytochrome incorporation into microsomal lipid liposomes slightly increases the degree of spiralization. The reduction of cytochrome b5 haem slightly decreases alpha-helices in the soluble haemoprotein as well as in the one incorporated into the artificial membrane irrespective of its phospholipid content.

[Covalent binding of codeine hydroxylation products to albumin and microsomal membranes].

It was shown that incubation of rat liver microsomes with [3H] codeine in the presence of 3 mM NADPH and 0.5% albumin is accompanied by covalent binding of codeine radioactive metabolites to albumin and microsomes. At low concentrations of the microsomal protein in the samples the number of metabolites bound to the membranes is greater than those bound to albumin. At increasing concentrations of the microsomal protein the binding occurs mainly at the expense of albumin. The animal induction with phenobarbital increases the number of bound metabolites. An injection of 3-methylcholantrene does not affect the degree of binding. All typical inhibitors of cytochrome P-450, i.e. SKF-525A, methyrapone, octylamine and CO, inhibit the binding. The data obtained suggest that the metabolites which bind to the macromolecules are generated by cytochrome P-450. It is assumed that the appearance in the blood of covalently bound albumin-hapten conjugates formed in the cytochrome P-450--hydroxylase system can induce the immune response. Hense, one more protective system of the organism becomes involved in the decontamination of low molecular weight compounds.

[Conformation and thermal stability of soluble and liposomal forms of cytochrome P-450].

This secondary structure of soluble cytochrome P-450 and the one incorporated into liposomes from egg lecithin and microsomal lipids has been studied. Using circular dichroism and infrared spectroscopy, it was shown that about 60% of alpha-helices are presented in the structure of haemoprotein and the rest 40% have the structure of statistical coil. The binding of haemoprotein with the type II substrates--octylamine and diaminooctan, slightly increases alpha-helices in soluble cytochrome P-450. The type I non-polar substrates--hexane and cyclohexane--do not change the conformation of isolated enzyme. Cytochrome P-450 incorporated into the artificial membranes of phosphatidyl choline and microsomal phospholipid has almost identical secondary structure as does the soluble one. Data from circular dichroism suggest that the binding of the types I and II substrates to cytochrome P-450 incorporated into lecithin liposomes and microsomal lipid liposomes does not change the conformation of the polypeptide chain. The reduction of cytochrome P-450 haem increases the degree of alpha-spiralization by 10% for soluble haemoprotein and by 5% for the membrane-bound enzyme. The thermal stability of soluble and liposomal forms of cytochrome P-450 was investigated by circular dichroism technique. The effective values of enthalpy and the temperature transition of soluble cytochrome P-450 at pH 6.9, 7,6 and 7,9 are 78, 80 and 78 kcal/mol and 47,7 degrees, 45,2 degrees and 42,4 degrees, respectively. The enzyme incorporated into the phospholipid vesicles is much more stable. The cooperative transition of soluble cytochrome is clearly expressed in contrast to the one of the membrane-bound enzyme.

[Kinetics of formation of the enzyme-substrate complexes of cytochrome P-450 in rat liver microsomes].

A spectrophotometric study of the kinetics of binding of I and II type substrates to cytochrome P-450 from rat liver microsomes has been carried out. The rates of binding of the substrates of type I to a low spin form of the enzyme and those of substrates of type II to a high spin form are approximately the same. The interaction of type II substrates with a low spin form of cytochrome P-450 occurs at a 15--20-fold lower rate. The dependence of initial reaction rates on substrate concentration describing the Michaelis equation suggests that the reaction occurs in more than one step both for type I and II substrates. The data obtained allowed to develop a two-step scheme of reactions, according to which the substrate in the first step rapidly and irreversibly interacts with the binding centers, which can be located both in the microsomal membrane and in cytochrome P-450. At the second stage the substrate in transferred into the active center of cytochrome P-450, resulting in a formation of complexes with specific optical properties.

[Induction of cytochrome P-450 and the immune response by free and covalently bound to albumin phenobarbital].

There is a relationship between the induction of microsomal enzymes and xenobiotic-induced immunological response. Phenobarbital bound with albumin covalently loses the ability to induce the cytochrome P 450-hydroxylase system but acquires an ability to induce the lymphocytic immunological system. This indicates that the two protection systems of the body, hydroxylase and immune ones, can interact with low and high molecular xenobiotics. The cytochrome P 450-hydroxylase system of the liver and other tissues is intended for protection of the body from the action of low molecular hydrophobic compounds. The immunological system is responsible for the organism protection from high molecular compounds.

[Comparative study of effects of the tyrosine-copper(II) complex on xenobiotic hydroxylation and lipid peroxidation].

It has been found that NADPH-dependent hydroxylation of dimethylaniline, aniline, p- and o-nitroanisol and lipid peroxidation is inhibited by the tyrosine-copper (II) complex (low molecular weight analog of superoxide dismutase), which is indicative of a possibility of superoxide radicals formation in these reactions. The inhibition of the above-mentioned reactions with Tyr2-Cu2+ is less pronounced or absent, if cumole hydroperoxide is used as cosubstrate instead of NADPH. Differences in the Tyr2-Cu2+ complex effects on the cumule hydroperoxide-dependent xenobiotics hydroxylation and lipid peroxidation catalyzed by various forms of cytochrome P-450, e. g. microsomal, soluble and incorporated into liposomes, have been found. The data obtained suggest that the efficiency of the inhibitory effect of the Tyr2-Cu2+ complex depends on the type of cosubstrates (NADPH, cumole hydroperoxide) and substrates used as well as on the form of cytochrome P-450.

[Accessibility of microsomal membrane proteins for protease K].

The effect of protease K on the microsomal membrane proteins was studied. It was shown that treatment of the microsomal membranes by low concentrations of protease did not remove more than 35% of total membrane proteins. Treatment by higher protease concentrations removed about 50% of the proteins. The residual protein is accessible to protease after treatment of the membranes by phospholipase A2. A simultaneous treatment of the microsomal membrane by protease and phospholipase removed up to 80% of the membrane proteins. In this way about 30% of microsomal membrane proteins are protected by the phospholipid against the action of protease K.

[Microsomal hemoprotein reduction by superoxide radical formed on NADPH-specific flavoprotein].

The superoxide radicals formed on NADPH-specific flavoprotein of liver microsomes can reduce cytochromes c, b5, and P-450. This reaction is inhibited under aerobic conditions by a low molecular weight analog of superoxide dismutase, e.g. the copper-tyrosine complex. The inhibitory effect of the complex is not observed under anaerobic conditions. Based on the results obtained a scheme of the electron transfer between the flavoprotein and haemoproteins involving superoxide radicals is proposed.

[Reconstitution of liver endoplasmic reticulum membranes from solubilized proteins and lipids by removal of detergent].

A method for membrane reconstitution from cholate-solubilized microsomal proteins and lipids by a removal of the detergent on a column with charcoal has been developed. A comparative study showed that the membranes reconstituted by a dialysis or absorption do not differ from each other in terms of membrane proteins incorporation into lipid vesicles and cytochrome P-450 reconversion into cytochrome P-450. A possibility of biomembrane reconstitution from membrane proteins and lipids solubilized by a non-ionic detergent Triton X-100 was shown. A removal of the detergent results in a formation of membranes, which are chemically close to the original ones but ultrastructurally very different from the latter. On the other hand, absorption or dialysis of cholate-solubilized proteins and lipids results in reconstituted membranes with asymmetrically arranged intramembrane particles located on the hydrophobic surfaces of the membrane halves. The number and size of these particles are similar to those of the original microsomal membranes.

[Effect of purified cytochrome P450 incorporation into liposomal membranes on its properties].

The physico-chemical properties and hydroxylase activity of three forms of cytochrome P450, i. e. purified soluble hemoprotein, purified hemoprotein incorporated into the liposomal membrane and microsomal cytochrome P450, were studied. Soluble cytochrome P450 binds type I substrates in a lesser degree than does its microsomal form. The incorporation of hemoprotein into phosphatidyl choline liposomes restores the ability of purified cytochrome P450 to interact with these substrates. The soluble and lipid-bound forms of cytochrome P450 do not differ in their thermal stabilities and protease digestion. The liposome-bound cytochrome P450 has higher dimethylaniline, aniline and p-nitroanisol hydroxylase activities as compared to its soluble form. The aniline hydroxylase activity of microsomal, proteoliposomal and soluble forms of cytochrome P450 is inhibited by the tyrosinecopper complex with NADPH or cumole hydroperoxide as cosubstrates. The inhibiting effect of the complex on other hydroxylase activities depends on the type of cytochrome P450 and the cosubstrates and substrates used.

[Alteration of the lipid composition of rat liver microsomes on reconstitution of the cytochrom P-450 system. Inactivation of the enzyme by lysolecithin].

The lipid composition of rat liver microsomes, their "ghosts" and the reconstituted membrane hydroxylation system was investigated. The phospholipid content and composition of the reconstituted membranes were found to depend on the reconstitution method. The reconstituted membranes contained besides phospholipids also cholesterol and gangliosides. The cholesterol/phospholipid ratio of the reconstituted membranes was the same as that of the initial microsomes. Lysolecithin was shown to enhance the conversion of the active form of cytochrom P-450 to the inactive cytochrome P-420.

[Isolation, analysis and characterization of microsomal ghost fractions].

A new method of isolating the microsomal ghost fractions has been developed. The membranes were purified from adsorbed protein, ribosomes and intravesicular content. The purified membranes contained 6 times less RNA than did the original microsomes. The phospholipid:protein ratio in the ghosts increased from 0.34 to 0.80. Protein electrophoretic pattern ghost was more homogenous than that of microsomes. SDS-polyacrylamide electrophoresis showed that the ghosts contain 11 fractions as compared to 21 in microsomes. A high degree of membrane purification did not affect the inactivation of microsomal enzymic systems. Practically all cytochrome P-450 in the ghosts was found in its active form. The activities of NAD(P)H-dependent systems of dimethylaniline and ethylmorphine demethylation, p-hydroxylation of aniline were well retained. The removal of non-membrane protein allows to obtain an electrone microscope picture of the lipid bilayer as a structural basis of microsomal membrane.
