Ring 18 mosaicism in identical twins.

Male identical twins with r(18)/normal mosaicism are reported. Twin 1 has the characteristic manifestations of the r(18) syndrome, but twin 2 shows a normal phenotype. Cytogenetic study of cultured lymphocytes revealed that the proportions of r(18) are 19.7% and 19.2%, respectively. However in the fibroblast cultures, the ring is observed in 51% of cells in twin 1 and in 0% in twin 2. The mechanism of the occurrence of the discrepant phenotypes in the twins is discussed.

A new malformation syndrome of long palpebral fissures, large ears, depressed nasal tip, and skeletal anomalies associated with postnatal dwarfism and mental retardation.

Five unrelated patients with a previously unrecognized mental retardation malformation syndrome are presented. Clinical features common to them include moderate mental retardation, postnatal dwarfism, susceptibility to infection in infancy, and peculiar craniofacial dysmorphia characterized by long palpebral fissures, high-arched and abnormal eyebrows, heavy and long eyelashes, large ears, short nasal septum and/or depressed nasal tip, and cleft palate. Other anomalies are stubby fingers, deformed vertebra and other bone and joint anomalies, and abnormal dermatoglyphics. The absence of familial occurrence and of consanguinity suggests some environmental causation, but the possibility of an autosomal dominant or X-linked mode of inheritance remains. Based upon our five patients and other five of Niikawa et al, we propose this syndrome as a new disease entity.

Complete amino acid sequence of mouse liver metallothionein-II.

The complete amino acid sequence of thionein-II, one of the two major mouse liver thionein components, was determined. The main fragmentation of thionein-II, which consists of 61 amino acid residues, was accomplished by digesting the S-[14C]-carboxymethylated protein and the cyanogen bromide-treated oxidized protein with trypsin. The peptides obtained by papain digestion of S-[14C]carboxymethylated thionein-II were used to align the major tryptic peptides. The sequence was determined by a combination of automated and manual Edman degradation techniques. Remarkable structural homology is observed in mouse thionein-I, mouse thionein-II, and thioneins from man and horse.

Acute cadmium intoxication in inbred mice: a study on strain differences.

Male C3H, BALB/c and DBA/2 mice were injected subcutaneously with a single dose of 30 mumol CdCl2/kg. Most of the C3H mice died within 15 h, while all of DBA/2 survived for at least 48 h. At 6 h after Cd-injection, histology revealed severe hemorrhage, fatty change, zonal hepatocyte necrosis and congestion of C3H livers, while these findings were rare in the other strains even at 48 h post-injection. The rate of metallothionein (MT) induction at 6 h was significantly lower in C3H mice than in the other two strains.

Purification and some properties of diamine oxidase from pig kidney and human placenta.

1. A homogeneous preparation of pig kidney diamine oxidase was obtained by polyacrylamide gel electrophoresis and immunodiffusion. Its molecular weight was estimated as 170,000 by Sephadex G-200 column chromatography and as 220,000 by SDS-polyacrylamide gelelectrophoresis. After mercaptoethanol cleavage of disulfide bonding, the molecular weight of the subunit was 130,000. 2. Little contamination was observed in the preparation of human placenta by SDS-polyacrylamide gel electrophoresis and immunoelectrophoresis and the molecular weight was calculated as 30008000 and 280,000 by the extrapolation from the calibration curve of Sephadex G-200 column chromatography and SDS-polyacrylamide gel electrophoresis, respectively. The subunit was 170,000. 3. By immunodiffusion, immunoelectrophoresis and inhibition test of the enzyme activity using a heterologous antiserum, a good cross-reactivity was observed between the anti-human diamine oxidase serum and pig diamine oxidase. 4. The activities of the two enzymes were not almost inhibited by homologous and heterologous antisera.

In vitro cellular responses to autologous tumor extract detected by inhibition of macrophage migration.

Indirect macrophage migration technique was employed to study in vitro cellular immune response to autologous tumor extract in man. Cellular hypersensitivity to autologous tumor extract was detected in 21 (26%) of 80 patients with various carcinomas preoperatively by means of the in vitro production of macrophage migration inhibitory factor (MIF). The relative frequency of MIF production in these patients varied inversely with progress of the diseases. The follow-up studies reveal postoperative decline of the cellular hypersensitivity after curative operation. These findings indicate that this assay can be applied for detecting cellular hypersensitivity to autologous tumor extracts in clinical situation.
