alpha-L-Fucosidase in cystic fibrosis: analysis of skin fibroblasts and liver.

The lysosomal enzyme alpha-L-fucosidase has been examined by thin layer gel and column isoelectric focusing in skin fibroblasts and liver from patients with cystic fibrosis and controls. All three common phenotypes of the enzyme were observed in both control and CF fibroblasts. When individuals of the same alpha-L-fucosidase phenotype were compared, no major differences between the isozyme profiles of cystic fibrosis patients and controls were detected in either fibroblasts or liver tissue.

The consistency of a class of coercive child behaviors across school settings for individual subjects.

This study addressed the problem of the intraindividual consistency of a class of coercive child behaviors across two school settings. Observational data collected on eight coercive behaviors of 53 boys in classrooms and on the playground. Previous Guttman scale analyses had shown that these eight coercive behaviors constituted a response class. The rates of the eight coercive responses in the response class were rank-ordered separately for each of the 53 subjects in each of the two settings. The intraindividual rank-order correlations were all positive, and 37 were statistically reliable. This number of significant correlations was further shown to be significantly greater than chance. It was concluded that there is a substantial amount of cross-situational consistency if intraresponse class analyses are done for individuals.

Intracellular retention of lysosomal enzymes in cystic fibrosis.

Multiple leakage of lysosomal enzymes from cystic fibrosis fibroblasts into the extracellular space has been suggested to occur but was not observed in these experiments. Both extracellular and intracellular levels of enzyme activity were within the normal range. The release of lysosomal enzymes from individual cell lines was seen to be affected both by intrinsic properties of each cell line and by many external factors.

Response class: a Guttman scale analysis.

This study addressed the problem of response class as it relates to coercive child behavior. Guttman scale analyses were used to determine if there were regular, unidimensional progressions across eight coercive response categories; that children who performed high probability coercive behaviors would also tend to perform all of the lower probability coercive behaviors. Rank-order correlations were done to determine if there was consistency in the response class across settings. Two samples of boys were observed in two school settings, classroom and playground. One sample was a derivation sample of 26 boys, the second a replication sample of 27 boys. Reproducibility coefficients for the derivation sample were .92 for the classroom Guttman scale analysis and .93 for the playground analysis. For the cross-validation sample the reproducibility coefficients were .94 in both settings. Rank-order correlations across settings and done in terms of the number of subjects performing each of the coercive responses were .971 (df = 6, p < .001) for the derivation sample and .996 (df = 6, p < .001) for the cross-validation sample. It was concluded that the eight coercive responses observed appeared to constitute a response class, i.e., the ordering the responses was transitive across subjects, and that the coercive response hierarchy was stable across school settings for groups of children.

Treatment of a patient with clinical features of both eosinophilic gastroenteritis and polyarteritis nodosa with oral sodium cromoglycate.

A young patient who suffered from allergic respiratory disease presented with an illness characterized by abdominal pain, diarrhea, and weight loss. Investigations revealed radiological and pathological features consistent with a diagnosis of eosinophilic gastroenteritis. Because of extensive gastric ulceration, treatment with corticosteroids was withheld and in its place oral sodium cromoglycate therapy given. The patient's intestinal symptoms rapidly responded to this treatment and remained controlled for a period of five months. The patient subsequently developed a systemic illness with features of polyarteritis nodosa.

Responses to noradrenaline of portal vein strips from normotensive and spontaneously hypertensive rats.

1 Responses to noradrenaline of isolated portal vein strips from 5-7 week and 15-17 week Wistar Kyoto (WKY) and spontaneously hypertensive rats (SHR) have been examined. 2 15-17 week SHR tissue exhibited a greater sensitivity (lower ED50) to noradrenaline but only at reduced Ca2+EXT levels. 3 pA2 values for the alpha-adrenoreceptor antagonists WB 4101 and prazosin were significantly lower in 5-7 week SHR consistent with some change at the adrenoreceptor level. 4 Veins from SHR relaxed more rapidly in CA2+-free saline following removal of noradrenaline than did veins from WKY. 6 D600 is a competitive antagonist of the Ca2+EXT-dependent component of the noradrenaline response but K1 values were not significantly different in SHR and WKY.

Selective inhibition of flagellar activity in Chlamydomonas by nickel.

Flagellar activity in the biflagellate chlorophyte Chlamydomonas reinhardtii is selectively inhibited by Ni2+ or by treatment with Ca2+-chelating agents. Inhibitions of swimming speed, geotaxis, phototaxis, and pattern swimming result from qualitative and quantitative losses in the activity of individual flagella and in the coordination of activity between the 2 flagella of each cell. Addition of Ca2+ (a) prevents inhibition and (b) restores normal flagellar activity in inhibited cells. Mg2+ is partially effective in reversal of inhibition. Other ions do not cause similar inhibition or reversal of nickel inhibition. The characteristics of inhibition and reversal suggest that the primary target for nickel is a component of the flagellar apparatus, and that this component uses Ca2+ to perform its normal function in the regulation of flagellar activity. A 2nd target for nickel is a Ca-requiring process specific to phototaxis (and not involved in the photophobic response).

Malignant myoepithelioma of the parotid gland.

Myoepithelial cells are a significant component of most types of salivary gland neoplasms. A small but increasing number of case reports have also shown that pure myoepitheliomas form a distinct class of neoplasms with unique histological features. Previously reported cases have been either benign or, at most, locally aggressive. We present here a case of a malignant myoepithelioma of the parotid gland, confirmed by electron microscopy, that was locally aggressive and eventually metastasized. Review of this case and of existing reports indicates that myoepitheliomas exhibit a wide range of biological behavior that seems to correlate with their histological appearance.
