Detection and partial characterization of a new murine T-lineage antigen: evidence for its enzymatic modulation.

A molecule with an apparent molecular weight of 21,000 daltons was found to be a predominating differentiation antigen of peripheral rat T lymphocytes. This antigen is also expressed on the cell surface of thymus lymphocytes though partially in masked form. The observation that these determinants could be unmasked by neuraminidase treatment in vitro suggests that enzymatic processes may play an important role in antigenic transformation along the differentiation pathway of T axis lymphocytes. Since this structure was found to be displayed also by peritoneal exudate macrophages, it is referred to as T lymphocyte-macrophage antigen. The finding that an identical antigen could be identified in the mouse suggests its more common biological significance.

Isoelectric focusing of viral polypeptides in urea. A methodological study on poliovirus.

The applicability of isoelectric focusing in urea to the analysis of viral polypeptides is reported, using poliovirus as a model. Experimental techniques for the dissociation of virus particles, for isoelectric focusing in urea-containing polyacrylamide gels (rods, flat bed and slab gels) and for pH measurement and two-dimensional analysis are described and their results, as applied to poliovirus polypeptides, discussed. Special attention is given to problems of recovery of all of the proteins and the incidence of artifacts. The influence of reagents, dissociation conditions, focusing procedures and preparation and storage of virus material on the occurrence of charge modifications of the polypeptides has been investigated. Some recommendations are made for the application of the method to other viruses of particles.

Isoelectric points of polypeptides of standard poliovirus particles of different serological types and of empty capsids and dense particles of poliovirus type 1.

The isoelectric points of polypeptides of standard and dense poliovirus particles and of empty capsids have been determined by isoelectric focusing in urea and by two-dimensional analysis. Comparing virus strains belonging to the three serological types of poliovirus, differences in the pI of some, but not all of the structural polypeptides are found. The pI of polypeptides of dense particles and of empty capsids are identical with those of standard particles. Polypeptide VPo present in empty capsids has a pI between those of VP4 and VP2.

[Spontaneous body experiencing.].

The author, in collaboration with a group of colleagues is in the process of perfecting a new psycho therapeutic approach called Spontaneous Body Experiencing, The author works from the body without a preconceived notion of man. Rather he creates enabling conditions for the revelation of body secrets, for life to once again become a process, to express itself according to its own laws. Jie then sets forth the concepts basic to his approach such as touch which is viewed simply as a presence for the other; spontaneous body experiencing in which he describes his method as well as the resultant physical manifestations; the organismic process seen as a series of events linked to one and other and progressively involving the total organism in the form of movements having certain characteristics such as in voluntarism, their uniqueness from one individual to another, and their progressive globalization; the body-memory or energetic structure which is restrained movement. The author concludes with a description of the beneficial effects of this new therapeutic approach.

Derivatives of the insect moulting hormone for affinity chromatography, and their biological activities.

Several derivatives of the arthropod moulting hormone have been synthesized which were coupled to AH Sepharose 4B yielding about 2 mumole ligand per g wet gel. As an indication of the suitability of the ligands for biological work the puff inducing capacity of their methyl esters was tested. The methyl ester of inokosterone-C-26-carboxylic acid possesses the highest biological activity; lower activities were obtained with the esters of ecdysterone-C-6-CM-oxime and ecdysterone hemisuccinates. Therefore, inokosterone-C-26-carbocylic acid should be a useful ligand for affinity chromatography of ecdysone recptors from insect tissues.

Antibodies against human CMV-induced "early" antigens detected by immunoblotting are not exclusively associated with active infection.

We investigated the correlation between the prevalence of antibodies against human cytomegalovirus (HCMV) "early" proteins and the state of infection. Two hundred and twenty-eight HCMV-IgG-positive (HCMV late-antigen ELISA) sera, drawn from subjects with active and non-active HCMV infections, were tested for specific IgG antibodies against HCMV "early" antigens by a Western blotting micromethod. Applying virus strain AD-169, we found antibodies to HCMV "early" antigens in 59% of all cases, irrespective of origin. Subjects with an elevated risk of cytomegalic inclusion disease (renal transplantation patients, AIDS patients) revealed significantly broader immune reactions when compared with healthy HCMV-antibody carriers. However, in follow-up studies we detected one patient who lacked an immune response to HCMV "early" antigens during active infection and subsequent convalescence. Our results indicate that active HCMV infection is not necessarily associated with the formation of antibodies against "early" antigens. Moreover, these antibodies may persist during non-active infection.

Development of obesity in transgenic mice after genetic ablation of brown adipose tissue.

Brown adipose tissue, because of its capacity for uncoupled mitochondrial respiration, has been implicated as an important site of facultative energy expenditure. This has led to speculation that this tissue normally functions to prevent obesity. Attempts to ablate or denervate brown adipose tissue surgically have been uninformative because it exists in diffuse depots and has substantial capacity for regeneration and hypertrophy. Here we have used a transgenic toxigene approach to create two lines of transgenic mice with primary deficiency of brown adipose tissue. At 16 days, both lines have decreased brown fat and obesity. In one line, brown fat subsequently regenerates and obesity resolves. In the other line, the deficiency persists and obesity, with its morbid complications, advances. Obesity develops in the absence of hyperphagia, indicating that brown fat deficient mice have increased metabolic efficiency. As obesity progresses, transgenic animals develop hyperphagia. This study supports a critical role for brown adipose tissue in the nutritional homeostasis of mice.
