Chronic arsenic toxicity from drinking tubewell water in rural West Bengal.

Hepatic damage caused by chronic exposure to arsenic has been frequently described. Here we report on 13 patients from West Bengal, India, who consumed large amounts of arsenic in drinking water. An epidemiological investigation of the study area showed evidence of chronic arsenical dermatosis and hepatomegaly in 62 (92.5%) of 67 members of families who drank contaminated water (arsenic level, 0.2-2 mg/l). In contrast, only six (6.25%) of 96 persons from the same area who drank safe water (arsenic level, <0.05 mg/l) had non-specific hepatomegaly, while none had skin lesions. Hepatomegaly occurred in all the 13 patients who were studied in detail, although five had splenomegaly. Biopsy of samples of liver from these patients revealed various degrees of fibrosis and expansion of the portal zone that resembled non-cirrhotic portal fibrosis (NCPF).

Evaluation of two compounds as possible radioprotectors.

Radioprotecting effectiveness of S-[2-(diethylamino)ethyl] 4-methylbenzothiohydroximate hydrochloride (Dev-B-9) and S-[2-(diethylamino)ethyl]alpha-keto 4-methyl benzothiohydroximate hydrochloride (Dev-B 43) were evaluated by both survival studies in mice exposed to 10.5 Gy whole body gamma-irradiation and 24 hours deoxycytidine (CdR) excretion in urine of rats following 5 Gy irradiation. The drugs were tested after intraperitoneal (i.p.) administration. The drugs rendered significant protection in sublethal (5 Gy) irradiation but could not protect against 10.5 Gy exposure.

Evaluation of radioprotectors by the Na+ influx study in RBC of lethally irradiated rats.

Sodium homeostasis in adult male albino Sprague Dawley rats has been examined 24 hours after exposure to 11 Gy whole-body gamma irradiation. Rate of influx of 22Na in red blood corpuscles (in vitro) of irradiated rats significantly increased and was modified by the administration of some radioprotective drugs prior to irradiation. Solcoseryl and AET (200 mg/kg) gave excellent protection and the combinations of 5-HTP with AET or MPG rendered better protection than when used alone.

Protection with combinations of hydroxytryptophan and some thiol compounds against whole-body gamma irradiation.

Protection of mice (as measured by survival at 30 days) against whole-body gamma exposure by prior treatment with combinations of hydroxytryptophan (HT) and some thiol drugs, is reported here. The combination of HT and aminoethylisothiuronium bromide hydrobromide (AET) was much more effective than HT alone against the effects of 10.5 and 12.5 Gy of gamma ray exposure. A combination of HT and beta-mercaptopropionylglycine (MPG) gave marked protection against a 10.5 Gy exposure but not against 12.5 Gy. The combination of HT with cysteine or cysteamine also provided reasonable protection against 10.5 Gy. The combination of HT and cystamine rendered a low level of protection against 10.5 Gy, but again it was better than HT alone. The thiol drugs by themselves in the doses used in combination were ineffective in rendering protection after whole body gamma ray exposure.

Role of some radioprotectors on 59Fe uptake in bone marrow after whole-body gamma irradiation to mice.

59Fe uptake in the femur has been studied at four time intervals up to 10 days in mice receiving pretreatment with hydroxytryptophan (HT), AET, MPG and combination of HT and AET or MPG and exposed to 6 Gy whole-body gamma irradiation. MPG or 20 mg/kg dose of AET pretreatment has failed to improve upon the 59Fe uptake depression induced by the gamma irradiation to mice at any of the time intervals. AET (200 mg/kg), HT + AET (20 mg/kg) and HT + MPG treatments though have failed to prevent the initial 59Fe uptake depression induced by the irradiation, have helped a speedy recovery in the iron uptake so that normal level of uptake is obtained on the 7th day. Pretreatment with the HT dose also helps in a speedy recovery as judged on the 7th day and a normal 59Fe uptake is reached in these animals by the 10th day.
