Hepatic lipid peroxidation and trace elements--nutritional status in streptozotocin-induced diabetic rats.

The aim of this experiment was to study the interrelationships between nutritional status of chosen trace elements (Cu, Zn, Fe) and hepatic lipid peroxidation in streptozotocin-induced diabetic rats. Both copper accumulation and disruption of iron storage were observed in livers of diabetic rats. MDA 0' (baseline) and MDA 30' (produced) levels measured in the liver were negatively correlated with blood glucose levels. MDA 30' levels correlated positively with iron concentration in the liver. It is supposed that the hormonal lability during experimental diabetes caused changes in metabolism of trace elements, and subsequently influenced the rate of lipid peroxidation.

Effect of fat and retinol loading on serum triglyceride and retinol levels in patients with ulcerative colitis.

Serum copper, zinc, selenium, magnesium, calcium, cholesterol, triglyceride and retinol contents were determined in 11 patients with ulcerative colitis and in 20 healthy volunteers. Lipid and retinol loadings were performed as well. No differences were found in serum copper, zinc, calcium and selenium levels between the groups. Serum magnesium levels were somewhat lower in the patients than in the volunteers. During the period of loading, cholesterol levels did not differ between the two groups at any time. Triglyceride levels except for two persons, were higher in patients. Triglyceride curves showed pathological courses in each case. Retinol concentrations in patients remained below the values observed in volunteers. Data suggest a disturbed utilization of fat and retinol in patients with ulcerative colitis.

[Incidence of food allergy and zinc deficiency in children treated for atopic dermatitis].

Among the 134 children who were admitted to hospital with atopic dermatitis, authors found 41, suffering from food allergy and 39, being hypozincaemic; the latter ones belonged to the group of patients with serious and extended ekzemic laesions. The lymphoblast transformation with aspecific mitogen in the blood of ten hypozincaemic patients with negative tuberculinplaster-reaction in 9 cases showed pathological decreased level, and after one month of zinc intake, 8 children's lymphoblast transformation became normal.

A review of the application of chiroptical methods to analytical chemistry.

The analytical applications of the two most important chiroptical methods, optical rotatory dispersions (ORD) and circular dichroism (CD), have been surveyed, emphasizing the methods corresponding to the profile of the Journal of Pharmaceutical and Biomedical Analysis. After a brief introduction of the ORD and CD methods, the advantages and drawbacks of the application of the two methods have been described, and compared, and the calibration of ORD and CD instruments have been given. The analytical applications have been divided as follows: the use of ORD and CD in identification studies, the direct determination of optically active substances, the determination of optically active compounds after chromophore group formation, determination of optically inactive substances via their products with optically active compounds. The difference chiroptical methods increasing the selectivity of measurements and the chiroptical titrations have been summarized. Stress has been laid on the applicability of selective chiroptical detectors (based on laser light, too) used in liquid chromatography and the future developments have been mentioned.

Longitudinal examinations in the course of dietotherapy of mentally retarded obese in-patients.

Mentally retarded obese in-patients were fed by low-energy diet (4.2-4.6 MJ) for 9 months. During this period, an average of 13 +/- 4.5 kg loss of body mass occurred in men and 16 +/- 2.7 kg in women. Anthropometric measurements were performed before starting the dietotherapy and in the ninth month. Changes of body fat could be followed well when calculated according to BMI. Less reliable results were obtained with skinfold thickness measurements, presumably due to body deformities. Results of clinical laboratory tests, which were carried out before starting the dietotherapy and in the fourth and seventh months, suggested that a low-energy-containing diet with balanced nutrient content and adequate protein intake did not impair protein metabolism, favourably affected serum triglyceride and cholesterol levels, but resulted in an unfavourable decrease in the HDL-cholesterol content.

The effect of alpha and beta adrenergic blockade on Ni induced coronary spasm in scalded rats.

After 20% total body surface scalding inducing 3d degree burn in rats the following parameters were studied in ebb and flow phases, as well as in burn disease, 2 hours, 2 and 7 days postburn: atomic absorption spectrophotometric study of the serum and myocardial Ni levels; the effect of Ni ions (10(-8)-10(-4) M/l) in vitro on total coronary resistance in the isolated perfused rat heart; the effect of alpha and beta adrenergic blocking agents (phenoxibenzamine, oxprenolol) on Ni-induced coronary vasoconstriction; electron cytochemical study of the ultrastructural changes of the myocardium 7 days after scalding. It was found that: During the first 2-3 hours after scalding or bleeding there was a significant elevation of serum Ni level which levelled off by the end of the first week. Total coronary resistance (TCR) increasing effect of Ni ions was significantly augmented during the first 2-3 hours after burn and bleeding (ebb phase). In the scalded group it decreased to the control value by the 2-3d day (flow phase) and increased again a week later (burn disease). In the scalded groups in vivo pretreatment with oxprenolol (10(-4) g/kg) completely inhibited the Ni induced coronary spasm. PBZ treatment in vitro (10(-5) M/l) had similar effect in the three postburn periods tested. Ultrastructural changes and intracellular Ni-dimetilglyoxim complexes could be detected. Oxprenolol treatment continuously applied from the 4th day postburn (2 X 0.5 mg/kg) prevented the development of myocardial damage, while the electron dense complexes containing Ni were still detectable. In view of our previous data we assume that the endogenous Ni increase in the serum after burn may be cardiopathogenic. The phasic changes of the sensitivity of the coronary vessels to Ni ion and the similarity of the effects of alpha and beta adrenergic blocking agents may be explained by the alterations in the neuroendocrine system and metabolic disturbances.

Endogenous nickel release in injured patients: a possible cause of myocardial damage.

Atom absorption spectrophotometry was used for the study of serum nickel levels in 58 injured patients. From 27 autopsies an electron cytochemical examination of the myocardium was performed in 19 patients. In patients with severe burns and cerebral injury, the serum nickel content showed a significant increase in the early period after injury. In these cases direct traumatic or toxic damage to the central nervous system and, in cases of early death, cerebral oedema, were present. The ECG tracings revealed disturbances in repolarization in a large majority. Nickel-dimethylglyoxim complex precipitates were found in the myocardium of all patients who died. In addition to disorientation, increased serum nickel levels and abnormal ECG records were observed in septicaemia. On the basis of these observations and the experimental data, we suggest that one of the causes of posttraumatic myocardial damage may be endogenous nickel release as part of the homeostatic disequilibrium arising as a direct or indirect effect of the injury.

Copper(II) and nickel(II) ternary complexes of L-dopa and related compounds.

The stability constants of the mixed ligand complexes of L-dopa, L-tyrosine, L-phenylalanine, and dopamine with copper(II) and nickel(II) ions and with 2,2'-bipyridyl and 1,10-phenanthroline were determined pH-metrically at 25 degrees C and an ionic strength of 0.2 mol/dm3 (KCl). Spectral studies were made to establish the binding mode of the ambidentate L-dopa in the ternary complexes. In contrast with the aromatic (N,N) donor atoms, the (O,O) binding mode of L-dopa is particularly favored in its ternary systems with copper(II) and nickel(II); thus, even at physiological pH there is a very considerable formation of (O,O)-bound mixed ligand complexes containing a free amino acid side-chain. Numerous binary transition metal-L-dopa complexes and the ternary complexes formed with various B ligands have been evaluated from a coordination chemistry aspect, with regard to the possibility of their therapeutic application in the treatment of Parkinson disease.

[Supplying various age groups with certain essential trace elements].

Investigations revealed a very good correspondence of the intake of trace elements, calculated from per capita food consumption, with the analytically determined values of the full daily diet. It is above all the copper intake and, to a lesser degree, the zinc and iron intake, which deviate from the generally recommended values. The results obtained for the different age groups indicate the copper intake to be sufficient only with children up to six years old. The iron supply of adolescent girls and old people should be improved. As is well known, the utilization of trace elements is influenced by many dietetic and individual factors. Therefore, further investigations must be carried out to decide whether the analytically determined intake of trace elements guarantees an adequate supply or whether disturbances often occurring with a slight under-supply must be taken into consideration.

The importance of quantitative electron microscopy in studying hypertrophic neuropathies. A comparison between a case of Déjérine Sottas disease (HMSN III) and a case of the hypertrophic form of Charcot-Marie-Tooth disease (HMSN I).

The electron microscopic features and quantitative morphometric data of the sural nerve in Déjérine-Sottas disease (HMSN III) and in the hypertrophic form of Charcot-Marie-Tooth disease (HMSN I) are reported. Both forms are characterized by onion bulb formations, but they differ in: a) increased incidence of mucoid connective tissue in DS disease; b) higher incidence of demyelination in DS disease; c) uniformly small size of the remaining myelinated fibers in DS disease; d) larger and more developed onion bulbs in DS disease; e) clusters of 2-4 myelinated fibers in the core of onion bulbs of CMT disease; f) peripheral concentric lamellae with the typical aspects of the denervation bands of the unmyelinated fiber type in the onion bulbs of the CMT disease; g) non-involvement of unmyelinated fibers in DS disease. These differences permit two types of onion bulbs to be distinguished, and suggest a different pathogenesis. Electron microscopy, coupled with quantitative determinations, permits a deeper insight into the formation modalities of onion bulbs and may help in the diagnosis of the different forms of hypertrophic neuropathies.

Endogenous nickel release as a possible cause of coronary vasoconstriction and myocardial injury in acute burn of rats.

The aim of the present study was to test the hypothesis that endogenous nickel release may play a role in myocardial depression in the acute phase of burn shock in rats. Serum nickel concentration (Se-Ni) measured by atomic absorption spectrophotometry, the sensitivity of isolated perfused hearts to exogenous nickel chloride (0.01 mumol/l), and ultrastructural myocardial alterations, as well as subcellular localization of endogenous Ni by the dimethylglyoxim method, were analyzed in the first week after standardized third-degree full thickness skin thermal burn covering 20-25% of the body surface. In the first 2 days after burn a significant fivefold increase of Se-Ni concentration was observed which leveled off by the end of the first postburn week. The coronary vascular resistance increasing effect of exogenous NiCl2 grew significantly in the first 3 h after burn, decreased to control in the 2nd postburn day, and increased again in the second half of the week after burn. Both ultrastructural damages and nickel containing reaction products could be observed in the myocardium 7 days after burn. The experimental results suggest that endogenous nickel release may play a role in coronary vasospasm and consequent myocardial depression in the acute phase of burn shock in the rat.
