Cf plasma desorption mass spectrometry of a polycyclic peptide antibiotic, nisin.

The polycyclic peptide antibiotic, Nisin, has been analysed by plasma desorption mass spectrometry using two different sample preparation techniques and two versions of the commercial plasma desorption mass spectrometer, and a prototype with high resolving power. The spectra obtained allow identification of a major component and two minor analogues. Extensive fragmentation is observed in samples prepared by the electrospray technique, whereas only ions indicating the molecular weight are produced when the sample is adsorbed on nitrocellulose.

Molecular approaches to genome analysis: a strategy for the construction of ordered overlapping clone libraries.

Here we describe progress on a series of molecular techniques designed to bridge the gap between genetic and molecular distances in mammals. This is an essential step in the molecular cloning of genes defined by mammalian mutations, and in the molecular analysis of large regions of mammalian genomes. We summarize approaches for the physical and molecular analysis of genetic distances and describe the experimental, statistical and computational basis of a new approach to create ordered libraries of overlapping clones from large genomes.

A mouse gene family associated with a major submaxillary-gland glycoprotein.

A gene family encoding msp36 (mouse submaxillary protein 36), a major product of the mouse submaxillary gland, was shown by Southern analysis and genomic cloning to contain approx. 10 related genes. Heteroduplex mapping has elucidated the structure of one of the genes. Close physical linkage of four of the genes in this family has been demonstrated by the use of specific oligonucleotide probes.
