[Modification of granulocyte function by lymphocytes].

Results of investigations are presented which are to show the impact of lymphocytes on granulocytophagocytosis . Lymphocytes in leukocyte suspensions are damaged by irradiation and by anti-human lymphocyte globulin. A slightly negative impairment of the phagocytic granulocyte function seems to be detectable. These findings are not unequivocal and not significant.

[Simultaneous studies of granulocyte phagocytosis, serum immunoglobulins, acute phase proteins and the 3d complement component in patients with malignant tumors].

In a clinical study granulocyte phagocytosis, serum levels of IgG and IgA and of the acute phase proteins ceruloplasmin, alpha-1-antitrypsin, transferrin, and of the 3rd component of complement (C3) are simultaneously determined in tumor patients and compared with healthy probands of the same age and with patients being under general surgical treatment. There was found a negative correlation between granulocyte phagocytosis and the IgG level and a positive correlation between phagocytosis and the C3 level in each group. Despite of some significant differences between all groups of probands with respect to the measured parameters no new essential information for clinical use could be received, thus the determination of granulocyte phagocytosis remains the only recommended method.

[Modification of granulocyte phagocytosis by extracts from tumor antigens].

The results with the granulocyte phagocytosis test adding various tumor antigen preparations are represented. In comparison with clinically healthy probands and patients having benign diseases there could not be proved any malignancy and organ specificity of granulocyte phagocytosis in patients with malignant tumors of different stages and various localization of organs.

Flow cytometric measurements of phagocytosis. I. A methodical and comparative study.

A flow cytometric technique is described for measurement of phagocytic activity in human leucocytes using FITC-labelled latex particles (0.77 micron). Cell suspensions separated from peripheral blood by dextran sedimentation were hemolyzed to avoid particle uptake by erythrocytes. The leucocytes were incubated for various time intervals with a standardized solution of 2 x 10(7) FITC-labelled latex particles per 2 x 10(5) leucocytes in 0.6 ml medium. Relative fluorescence per cell was measured on the basis of 10(5) cells, and a simple technique has been described to evaluate data characteristic for different steps of phagocytosis and to discriminate between phagocytizing and non-phagocytizing cells. In comparison, rice starch phagocytosis has been measured microscopically. A positive correlation between both methods has been found in 11 patients with different disease-related phagocytic capacity.

[In-vitro studies of granulocyte phagocytosis in patients with malignant tumors].

Investigations are presented about granulocyte phagocytosis with rice starch in patients with clinically curable malignant tumours and with those treated palliatively. A significantly reduced granulocyte efficiency of phagocytosis indicating a bad prognosis of the disease can be found in those patients being in the advanced tumour stage.

[The value of the granulocyte phagocytic test in oncology follow-up care].

This work represents the results of the granulocyte phagocytic test in tumor effected individuals by observing the course over a period up to three years. In tumor patients, which could be operated radically, a normal phagocytosis activity of granulocytes indicates the undisturbed course. In patients with progressive carcinoma growth the average of phagocytosis is lowered obviously. These results revealed information for the prognosis of malignant diseases and the nonspecific accompanying diseases.

[The normal state of granulocyte phagocytosis].

There are demonstrated results of the granulocyte phagocytosis with rice starch with healthy patients. The early phase of the phagocytosis seems to be more sensitive on changes of the unspecific defence. There were not found any significant differences between men and women. Patients who are older than 75 years have not a significant lower phagocytosis than younger ones.

Quantitative changes of some rat proteins by short and long time carbon tetrachloride administration.

In the last few years, the immunological quantitation of human plasma proteins has proved a useful diagnostic routine method and is used in many laboratories. It is conceivable that complex estimations of serum proteins provide valuable and addition information on substance activities also in toxicological animal experiments. To test the validity of this concept, we have estimated the serum concentration of four proteins (IgG, transferrin, alpha2-acute phase protein = alpha2-AP, very low density lipoprotein = VLDL) in carbon tetrachloride-treated rats. The serum concentrations of the examined proteins are changed significantly within the first 3 d after onset of treatment. IgG and transferrin are elevated during the whole period of treatment, VLDL falls after only 1 d of treatment, and alpha2-AP shows a biphasic course. Our results are comparable with the findings described for human hepatitides. Furthermore, age-dependent and chrono-biological influences were observed.

Biorhythms of four serum proteins of the laboratory rat: IgG transferrin alpha 2-acute phase protein and a VLD lipoprotein.

Four serum proteins (IgG, alpha 2-macroglobulin or alpha 2-acute phase protein, transferrin and a VLD-lipoprotein) of the rat have been examined wit respect to quantitative alterations reflecting biorhythms. All four proteins show distinct biorhythms. VLDL and alpha 2-AP seem to undergo a common circadian rhythm whereas the circadian rhythms of IgG and transferrin are superimposed by an infradian and ultradian rhythm, respectively. The examined proteins are apparently regulated by different ways possibly dependent in each case on the biological function. By means of chronobiological investigations perhaps further functional characterization of serum proteins are possible and alterations of the biorhythm may give additional information for pharmacological or toxicological research.
