Clinical immunological study of T lymphocytes and EA rosette forming cells in tonsils.

Tonsillar E and EA rosette forming lymphocyte subpopulations were studied in 120 tonsillectomized patients. T cell ratios was usually lower in the tonsils than in the blood. EA binding cells were studied with indicator systems of human or rabbit antibody sensitized red cells, respectively. Poorly sensitized human RBCs (EArabbit) bind much better to tonsillar cells than to blood lymphocytes and so this system proved to be specially suitable to study tonsillar EA binding cells. Increase in E and EA rabbit rosette forming cell frequencies were found with the age of patients. Decrease in percentage of T cells and EA rabbit rosette binding cells were found with high frequency of acut tonsillitis and with clinical sysmptoms of chronic local inflammation. The variation of these lymphocyte subpopulations with the local tonsillar inflammatory processes suggest a considerable clinical immunological role of local T cells and of this portion of Fc receptor postive lymphocytes.

[Study of C3 and C4 complement components of synovial fluids in connective tissue diseases (author's transl)].

C3 and C4 levels and the relative concentration of C3 degradation products were measured in fifty pairs of synovial fluids and sera of patients who suffered from different connective tissue diseases. Inverse correlation between C4 level and C3 breakdown products were found in synovial fluids of different groups. Compared to the other groups of patients, the highest increase of C3 degradation was found in RA. patients and it occurred in the SF all of these patients. In arthrosis cases we failed to find any degradation of C3. Of the seven investigated ankylosing spondylitis only two showed some degree of activation of C3 in the SF. Our results suggest that the occurrence and degree of degradation of C3 complement component is a valuable laboratory parameter in the study of connective tissue diseases.

Differential binding activity of EA receptor bearing human lymphocyte subpopulations.

Two types of EA rosettes were studied in rheumatoid arthritis patients and control subjects. Heterologous (rabbit) and homologous (anti-D) antibody sensitized human red blood cells (EAr and EAh, respectively) were used for examining the EA binding capacity of human lymphocytes. Both control and rheumatoid arthritis blood samples yielded significantly lower EArRFC levels as compared to the percentage of EArRFCs, the difference being even more expressed with control lymphocytes. No difference, however, was observed in the case of tonsillar lymphocytes, as equal rosette levels were reached in both systems used. Introduction of immunosuppressive therapy resulted in a temporary dissociation of the two rosette values EAr and EAh binding lymphocytes were differently affected by aggregated human IgG, only the latter ones being inhibited, contrasting with the equal inhibitory action of purified Fc fragments. Reducing the density of anti-D antibodies on the surface of erythrocytes, the EAh rosette count dropped to the level of EAr RFCs. The data suggest an activity-depending Fc receptor distribution resulting in different binding capacity for more or less sensitized indicator RBCs.

Synaptosomal fractions of cat cerebral cortex after hypo-osmotic and detergent treatment.

Different synaptosomal fractions were prepared by subcellular fractionation from cat cerebral cortex. The seperated nerve ending fractions differed in both density and osmotic sensitivity. Synaptosomal ghosts were obtained by hypo-osmotic treatment. In the synaptosomal ghost fractions the synaptic areas seemed to be intact morphologically. The morphology of the "synaptic triad" (pre- and postsynaptic membranes and the synaptic cleft) was not affected even by treatment with Triton X-100 non-ionic detergent (0.6%). The solubilizing effect of different concentrations of the detergent was checked in the supernatants and in the insoluble residues. The homogeneity of various subcellular fractions and the effect of the osmotic and detergent treatments are discussed.

Phototherapy increases platelet 3H-imipramine binding in patients with winter depression.

The density of platelet 3H-imipramine binding sites (Bmax) was investigated in seven inpatients with winter depression before and after incandescent light treatment and in seven healthy volunteers within a 7-day interval without light exposure. A significantly lower mean Bmax value was found in the depressed patients compared to the controls. After light treatment, parallel to a marked clinical improvement, Bmax increased in each patient and reached or even exceeded the mean values of the controls. These results indicate that in patients with winter depression the decreased Bmax value is state-dependent. The incandescent light treatment has beneficial effect in winter depressives and can influence Bmax values.

Glucocorticoid-induced apoptosis and treatment sensitivity in acute lymphoblastic leukemia of children.

Sensitivity of leukemic blasts to steroid therapy is one of the prognostic factors in acute lymphoblastic leukemia (ALL) in children. We examined the number of steroid receptors and the increase in the apoptotic index in peripheral blast cells after administration of prednisolone monotherapy in 21 children with ALL. A new diagnostic method was established based on determination of the apoptotic index in peripheral blood lymphoblasts to evaluate the steroid sensitivity of leukemic cells during the first day of therapy. The increase in apoptotic ratio, analyzed by morphologic and/or flow cytometric studies, was most expressed in the first 6 hours of treatment. The apoptotic ratio showed a good correlation with the clinical response. The number of steroid receptors (gcRs) on the blast cells was also examined, but it proved to be less informative than the in vivo steroid response itself.

Soluble interleukin-6 receptor in plasma and in lymphocyte culture supernatants of healthy individuals and patients with systemic lupus erythematosus and rheumatoid arthritis.

The soluble IL-6 receptor (sIL-6R) occurring in various body fluids of healthy persons and patients with various diseases is an agonist since its complex with IL-6 binds to gp130 making IL-6 receptor negative cells responsive for IL-6. The generation as well as the functional role of soluble IL-6 receptor is poorly understood. We measured the sIL-6R levels by ELISA sandwich technology in sera and in supernatants of lymphocyte cultures without and after incubations with dexamethasone. Our results indicate, that the sIL-6R levels in sera of patients with inactive systemic lupus erythematosus (SLE) and active rheumatoid arthritis (RA) were higher than those of the control group, active SLE and inactive RA. In vitro dexamethasone treatment stimulated generation of sIL-6R in both healthy persons and in active SLE, however it strongly suppressed sIL-6R in both RA groups. At mRNA level, we found that in SLE both the mRNA coding the cell-bound and an alternatively spliced variant corresponding to soluble IL-6R transcript increases, however the strong decrease of sIL6R protein in RA was not found at mRNA level.

Retinoic acid enhances histamine content and H1 receptor expression in human neuroblastoma cell line Paju.

BACKGROUND: A human neuroblastoma cell line (Paju) was induced by retinoic acid (RA) to differentiate into neuron-like cells. MATERIALS AND METHODS: We studied the expression and the possible role of histamine receptors H1 and H2 in retinoic-acid mediated differentiation by semiquantitative RT-PCR. We studied the effect of exogeneously added RA on the morphological change of the human neuroblastoma cell line and the differentiation was followed by vimentine, glial fibrillary acidic protein (GFAP) and neurofilament (NF) immunostaining. We monitored the change of the histidine decarboxylase (HDC) expression and the histamine content during the RA treatment by immunoblot and flow cytometry methods. RESULTS: Our data showed that H1 and H2 histamine receptors are present on Paju cells. Ten nM RA markedly increased the H1 receptor expression of these cells, while the H2 expression was unchanged. CONCLUSION: In the RA-treated Paju cells, the histamine content increased compared to the untreated cells, suggesting that neuroblastoma-derived histamine is involved in the regulation of RA-induced in vitro differentiation by H1 receptors.

[Functional analysis of immune complex-rich serum fractions in autoimmune diseases by in vitro C3 activation of fresh blood].

A method, measuring the activity of circulating immune complexes, based on C3 conversion of fresh human plasma by means of 3 per cent PEG precipitable immune complex-rich sera fractions is reported. Sera of patients suffering from autoimmune diseases of the connective tissue as well as of healthy controls were investigated. In case of standardised protein concentration of immune complexes, significantly low C3 conversion frequency of the isolated fractions has been observed in the control group as compared to the examined patients (rheumatoid arthritis with and without extraarticular manifestations, or juvenile chronic arthritis, or systemic lupus erythematosus). PEG fractions of RA with extraarticular manifestations induced the significantly highest C3 splitting. It is assumed that immune complexes will show a distinct interaction with the complement system also in vivo, and are therefore directly implicated in the systemic course of the disease process.
