Thermal injury to bone. A vital-microscopic description of heat effects.

A titanium implant, the thermal chamber, allowing vital microscopic observations of bone tissue during heating and bone temperature measurements to be made simultaneously is described. The tissue reactions to a defined heat trauma may be followed and registered on slides and ciné film. The thermal chamber technique allows repeated observations of the same bone tissue compartment for indefinite follow-up period. In the present experiment, the chamber was inserted in the rabbit tibia and heating to 53 degrees C was applied for 1 min. During heating to the assigned temperature, the blood-flow velocity initially increased. At 53 degrees C the blood flow stopped in some vessels and became sluggish in others. 2 days after heating the blood flow in the pre-existing vessels had stopped. Gradually thereafter, all original vessels became replaced with ingrowing newly-developed vascular pathways. Fat cells seen before burning became resorbed and bone remodelling started 3-5 weeks after the heat injury. A temperature of 53 degrees C, i.e. below the denaturation point of alkaline phosphatase, caused an irreversible bone injury, after which healing occurred from the surrounding tissues.

Fatal snowmobile accidents in northern Sweden.

The number of snowmobiles registered in Sweden has increased rapidly in recent years. At the same time, there has been a proportional increase in the number of snowmobile accidents. In the present study all the 36 fatal accidents reported in northern Sweden in the period 1 July 1973 to 1 July 1981 have been scrutinized. Most of the fatalities were men, two thirds of them between 20 and 40 years of age. Most of the accidents occurred at weekends and during holidays, especially in the spring, when the traffic is heaviest. About two thirds of the fatal accidents occurred at night. Drowning and lacerations of the thorax and the head were the most common causes of death. Twenty-four of the 30 drivers killed were found to have been driving under the influence of alcohol. This suggests that alcohol was a major contributing factor to the accidents. Retrospective analyses of police, social, and medical records revealed that a high proportion of the deceased were habitual excessive drinkers. Preventive measures designed to reduce alcohol consumption while driving snowmobiles seem to be needed.

Is muscle structure influenced by genetical or functional factors? A study of three forearm muscles.

Relative fibre type composition and weights of 3 forearm muscles; the m. extensor carpi radialis longus (ECRL), m. extensor carpi radialis brevis (ECRB) and the m. brachioradialis (BR) from both the left and right arm were determined. Fibre type 1 muscle weight of the ECRB, which co-contracts posturally during gripping, was significantly greater in the right than the left forearm. Weights of type 2 fibres in this muscle were, however, similar in the left and right arm. Fibre type weights of both ECRL and BR were also similar in both arms. For all these muscles, intra-individual co-variation in relative fibre type occurrence was found. It is proposed that genetic factors may play a role in determining the relative fibre type composition at least as far as the upper extremities are concerned. However, functional demands are also important determining factors for development of muscle structure.

Genetic background of acute anterior uveitis.

To determine the hereditary pattern of acute anterior uveitis, we examined seven families-one family with four cases, one family with three cases, and four families with two cases of acute anterior uveitis. A total of 16 patients with uveitis and 28 relatives underwent ocular examinations and serologic determination of HLA antigens, protease inhibitor alpha 1-antitrypsin typing, and X-ray examination of sacroiliac joints. Sacroiliitis was found in 11 of 16 patients (68.8%) and incomplete Reiter's disease in seven of 16 (43.8%). HLA-B27 antigen was found in all patients with uveitis and in six of 21 (28.6%) relatives. HLA-Cw1 antigen was found in nine of 14 (64.3%) patients with uveitis, often in the same haplotype with HLA-B27 antigen. HLA-DR4 antigen was found in seven of 11 (63.6%) patients with uveitis. There was no significant association of the alpha 1-antitrypsin variant MZ with uveitis. We believe that a pleiotropic gene associated with HLA-B27 antigen with autosomal dominant inheritance, incomplete penetrance, and variable expressivity may determine susceptibility to acute anterior uveitis in linkage disequilibrium with HLA-Cwl antigen.

Filament systems in the Purkinje fibers of the heart.

The presence of a specialized cell system designed to conduct the electrical impulses for the sequential contraction of heart muscle is well known. This conduction system of the heart exhibits special morphological features and differs from the contractile myocardium in several respects, including cell form and size, intercellular contacts, and the absence of T tubules. This review is focused on another distinguishing feature, the specific filament systems present in the conduction cells. The conduction cells contain myofibrillar material with different histochemical, biochemical, and morphological properties compared with the contractile myocardium. The conduction cells also contain specific myofilament-polyribosome complexes, of unknown function, never seen in ordinary myocardial cells. Intermediate filaments, present in all eukaryotic cell types, are extremely abundant in conduction cells, especially in birds and larger mammals. Other filamentous organelles of the conduction cells include leptofibrils, microtubules, and microfilaments. The combined results from several investigations indicate that the filamentous systems maintain the structural integrity of the conduction cell bundles functioning as a cytoskeleton. Extracellular elements, such as the surrounding connective tissue sheath, may aid in this respect. Other functions have been ascribed to the filamentous systems of the conduction cells, but these are unproven.

Skeletin immunoreactivity in peripheral nerves.

By use of monospecific antibodies against the cow heart intermediate filament protein, skeletin, an antigenic relationship between skeletin and neurofilament protein of peripheral nerves is demonstrated. Crossreactivity is also demonstrated in the filament-containing Schwann cells. The results are consistent with the existence of several subclasses of related intermediate-sized filament proteins.

The development of the Purkinje fibre system in the bovine fetal heart.

In the bovine fetal heart, subendocardial bundles of cells could be distinguished from the main myocardial mass. Their morphological characteristics suggest that they represent bundles of Purkinje fibres. An intense fluorescence after incubation in antisera against the intermediate filament protein skeletin also supports this suggestion. Further, the bundles exhibited different histochemical reaction from the main myocardial mass. During development the histochemical pattern changed. Bundle cells in mitosis were observed. With increasing fetal age, binucleate cells were seen progressively more frequently. Our observations indicate that the Purkinje fibres differentiate along a line separate from the ordinary myocardial cells and that they acquire their adult characteristics gradually.

Transcellular aqueous humor outflow: a theoretical and experimental study.

When the aqueous humor passed through the transcellular channels of the inner wall endothelium of Schlemm's canal the flow was estimated to be very slow and viscous (Reynolds' number about 10(-3)). It was found to be accurate to treat this extreme flow situation with conventional hydrodynamic formulae without specific corrections. Four different principal configurations of the transcellular channels were investigated. The theoretical calculations yielded results in good agreement with those obtained from the experimental model studies. Where deviations occurred they were probably explained by a radial flow component, which was not incorporated in the theoretical formulae applied. A spherical channel configuration was considered most similar to the in vivo situation and this yielded experimental resistance values half those of previously applied theoretical methods. Using previous morphological data the present results indicated that less than 5% of the total resistance to aqueous humor outflow was located in the inner wall endothelium in normal human eyes, and about 10% in cynomolgus monkey eyes at normal intraocular pressures.

The reliability of histochemical fibre typing of human necropsy muscles.

The reliability of muscle fibre typing of post mortem specimens was investigated with special reference to the influence of time and temperature. In specimens stored at +4 degrees C, muscle fibre typing could be reliably performed up to at least ten and fifteen days post mortem for the masseter and biceps brachii muscles respectively. The corresponding figures for storage at room temperature were three and six days. The difference in the preservation of enzyme activity between masticatory and limb muscles might be related to the demonstrated difference in the fibre type composition and thus the enzyme content and energy sources.
