Dy-DTPA-BMA as an indicator of tissue viability in MR imaging. An experimental study in the pig.

The aim of this study was to investigate whether dysprosium (Dy) induced signal intensity (SI) loss in infarcted tissue in MR imaging. Myocardial infarction was induced in 12 pigs and Dy-DTPA-BMA (1.0 mmol/kg b.w.) was administered i.v. to 6 pigs 4 hours after occlusion and allowed to accumulate in the infarctions for 2 hours. Dy was analysed by inductively coupled plasma atomic emission spectrometry in infarcted and non-ischaemic tissue samples. The remaining 6 pigs, not administered contrast medium, served as controls. The infarctions demonstrated a high SI in the proton density- and T2-weighted sequences in both groups (ex vivo), although the Dy-DTPA-BMA group demonstrated a 3-fold greater concentration of Dy in infarcted compared with non-ischaemic myocardium. The lack of SI loss after Dy accumulation indicates that susceptibility effects are minor or absent in infarcted myocardium.

MR imaging of double-contrast enhanced porcine myocardial infarction. Correlation with microdialysis.

MR imaging was performed to investigate whether Gd-DTPA-BMA-induced contrast enhancement of myocardial infarction is counteracted by Dy-DTPA-BMA. Myocardial infarction was induced in 5 pigs. Microdialysate probes were inserted in ischemic and nonischemic myocardium. Gd-DTPA-BMA (0.3 mmol/kg b.w.) and Dy-DTPA-BMA (1.0 mmol/kg b.w.) were administered i.v. 4 hours post occlusion. The microdialysate was collected every 10 min and measured for Gd and Dy using inductively coupled plasma atomic emission spectrometry. The pigs were sacrificed 2 hours after administration of contrast media. The concentration of both contrast agents was 3 times higher in infarcted myocardium than in nonischemic myocardium. The infarctions displayed high signal intensity in spin-echo sequences ex vivo. This lack of detectable susceptibility effects from Dy may be caused by loss of cell membrane integrity in infarcted tissue as shown by our microdialysate and biopsy data.

Combination of gadolinium and dysprosium chelates as a cellular integrity marker in MR imaging.

A combination of gadolinium (Gd) and dysprosium (Dy) chelates was investigated as a potential marker of cell-membrane integrity by means of a double-contrast effect in MR imaging. Blood samples with varying hematocrit (Hct) levels containing intact or lysed cells were used as model systems. With intact cells, the agents were assumed to be distributed solely extracellularly and the highest Hct studied (69%) was assumed to mimic the ratio of extracellular to intracellular water in tissue. The combined effect on image intensity of Gd (in a concentration corresponding to 0.2 mmol/kg b.w. in humans) and Dy (0.6 mmol/kg b.w.) applied simultaneously was a marked difference in signal intensity between samples with intact and lysed cells in both the T1- and T2-weighted spin-echo images with a corresponding increase in the contrast-to-noise ratio. This was the result of a T1 reduction caused by Gd with a negligible Dy susceptibility effect in areas with lysed cells. On the other hand, the Dy susceptibility effect (i.e. reduced apparent T2) dominated in areas with intact cells. Thus, the combination of Gd and Dy may serve as a marker of cell-membrane integrity in MR examinations.

MR imaging of gadolinium-DTPA-BMA-enhanced reperfused and nonreperfused porcine myocardial infarction.

To investigate whether Gd-DTPA-BMA-enhanced MR imaging permits differentiation between reperfused and nonreperfused myocardial infarction, myocardial infarction was induced in 12 domestic pigs. In 6 pigs, Gd-DTPA-BMA, 0.3 mmol/kg b.w. was administered i.v. 60 min after the occlusion. In 6 other pigs, the infarctions were reperfused 80 min after the occlusion, followed by injection of Gd-DTPA-BMA after 20 min of reperfusion. Radiolabeled microspheres were used to confirm zero-flow during the occlusion period and reperfusion in the infarcted myocardium. All pigs were killed 20 min after injection of contrast medium, and the hearts were excised and imaged with MR. The Gd concentration was measured in infarcted and nonischemic myocardium by ICP-AES. In the reperfused hearts, the infarctions were strongly highlighted, corresponding to a 5-fold higher Gd concentration in infarcted vis-à-vis nonischemic myocardium. In the hearts subjected to occlusion without reperfusion, there was only a rim of enhancement in the peripheral part of the infarctions.

Measurement of magnetic susceptibility and MR contrast agent concentration.

This paper describes an MR imaging method for determining magnetic susceptibility constants of solutions containing paramagnetic contrast agents. The method's validity is demonstrated on Gd(DTPA) and Dy(DTPA) water solutions. The method can be used for measurement of the volume magnetic susceptibility or concentration of contrast agents in biological tissues.

Superparamagnetic iron oxide for liver imaging. Comparison among three different preparations.

OBJECTIVES: The effects of differently sized superparamagnetic iron oxide (SPIO) particles as liver contrast agents were evaluated by relaxation analysis and magnetic resonance imaging in normal rabbits. METHODS: We performed relaxivity measurements in agarose gels; T1 was measured by saturation recovery. Rabbits were injected with SPIO particles to evaluate hepatocellular localization and magnetic resonance appearance. RESULTS: Small (30 nm), medium (300 nm) and large SPIO particles (3,500 nm) reduced the T2 of liver by 50%, 40% and 15%, respectively, and the T2 of spleen by approximately 60%, 65%, and 25%, respectively, at 1 mg Fe/kg intravenous injection. Both small and medium SPIO particles decreased the T1 of spleen by approximately 35%, with no effect on liver T1. Magnetic resonance imaging showed decreased signal intensity ratios (SIliver/SImuscle) by approximately 80% and 60% for small and medium SPIO particles, respectively. Iron oxide (positive Perls blue staining) was observed in Kupffer cells after injection of medium and large SPIO particles, and also in hepatocytes after injection of small SPIO particles. CONCLUSION: The liver contrast effect seemed to be related to cellular distribution; the widely distributed small SPIO particles were most effective.

Interferon treatment of neuroendocrine tumour xenografts as monitored by MRI.

The neuroendocrine-differentiated colonic carcinoma cell line (LCC-18) was transplanted to 29 nude mice (Balb/c). The purpose of the present study was to establish an animal model that would allow monitoring with magnetic resonance imaging (MRI) of changes induced by interferon (IFN) therapy and to evaluate whether the therapeutic response, as expressed by changes in MR signal characteristics and tumour proliferation activity, could be modulated by different IFN dosages. IFN did not seem to have any obvious antiproliferative effect on the LCC-18 tumour cell line transplanted to nude mice and no convincing treatment-related changes in rho values or T1 and T2 relaxation values were observed. The animal model was probably unsuitable for demonstration of IFN effects.

MR imaging in cerebral gliomas. Tissue component analysis in correlation with histopathology of whole-brain specimens.

A comparative analysis between MR examinations and histopathologic whole-brain sections regarding tumour components was performed in 5 brain specimens from patients with malignant glial brain tumours. All cases were examined with MR imaging in vitro and in 2 cases a close comparison with the MR examinations in vivo was also possible. The most homogeneous hypercellular area in malignant gliomas, giving the highest tumour grade, was not visualised on MR imaging as an isolated entity, either in vitro or in vivo. The most conspicuous tumour component, reflecting the heterogeneity of malignant gliomas, was necrosis. This feature was best depicted in the T2WI. In 4 of 5 cases, distant tumour spread of benign-looking tumour cells was found in areas visualised as normal on T2WI, outside the margins of the peritumoural oedema. In 2 cases, estimation of water content was performed immunohistochemically and a close correlation was found in each case between peritumoural and periventricular hyperintensity on T2WI and areas of pallor on the haematoxylin-eosin-stained whole-brain sections. These areas corresponded to microscopical oedema. MR imaging reflects underlying heterogeneous histopathology in malignant gliomas. The degree of malignancy of the lesion as a whole can thus be assessed by MR imaging. However, the method does not allow malignant gliomas to be correctly delineated.

MR imaging of cerebrospinal fluid dynamics in health and disease. On the vascular pathogenesis of communicating hydrocephalus and benign intracranial hypertension.

The CSF flows in the aqueduct and at the foramen magnum were examined in 5 patients with communicating hydrocephalus (HC) and in 10 with benign intracranial hypertension (BIH) as well as in 5 healthy volunteers. As compared to normal individuals, the aqueductal flow in HC was about 10 times larger and the cervical flow was half as large. In BIH the CSF flows were not different from those of normal volunteers. The decreased arterial expansion as reflected in the reduced cervical flow in HC may be due to pathologic changes in the arteries and paravascular spaces. The large aqueductal flow in HC reflects a large brain expansion, causing increased transcerebral mantle pressure gradient and ventricular dilatation. In BIH there is a normal brain expansion (aqueductal flow) and consequently no ventricular dilatation. It is argued that BIH be caused by an obstruction on the venous side, as opposed to the vascular alterations in HC, which are on the arterial side.

A functional anatomical analysis of central pathways subserving the effects of interleukin-1 on stress-related neuroendocrine neurons.

Systemic administration of the cytokine interleukin-1 (IL-1) results in increased secretion of ACTH and corticosterone in rats. The available evidence suggests that the acute effects of IL-1 are exerted ultimately at the level of the hypothalamus to increase corticotropin-releasing factor (CRF) secretion into the hypophyseal portal circulation, and hence the central drive on the pituitary-adrenal system. However, the route(s) and mechanism(s) by which circulating IL-1 gains access to central mechanisms governing pituitary-adrenal output remain poorly understood. In this study, we show that intravenous injection of IL-1 beta provokes time- and dose-dependent increases in the expression of the immediate-early gene c-fos, in identified CRF and oxytocin-producing cells of the paraventricular nucleus of the hypothalamus (PVH). Several cell groups known to be involved in central visceromotor regulation also displayed comparable time- and dose-related activation to systemic IL-1, including the bed nucleus of the stria terminalis, the central nucleus of the amygdala, the lateral parabrachial nucleus, and cell groups of the dorsomedial and ventrolateral medulla. Activation of circumventricular organs, which have been hypothesized to serve as central monitors of circulating IL-1, required doses roughly an order of magnitude above those required to activate CRF neurons in the PVH. Combined immunohistochemical and retrograde tracing experiments revealed many IL-1-responsive cells in the nucleus of the solitary tract and the ventrolateral medulla to be catecholaminergic and to project to the region of the PVH. Discrete and unilateral interruption of ascending catecholaminergic projections from the medulla attenuated IL-1-stimulated increases in Fos immunoreactivity and CRF mRNA in the PVH on the ipsilateral side. Disruption of descending projections from circumventricular structures associated with the lamina terminalis did not affect IL-1-mediated Fos induction in the PVH. We conclude that medullary catecholaminergic projections to the PVH play either a mediating or a permissive role in the IL-1-induced activation of the central limb of the hypothalamo-pituitary-adrenal axis.

Dependence on T1 of the echo amplitudes from multiple spin-echo sequences with equidistant echoes: simulation studies.

Transverse relaxation times were estimated from numerical simulations on spin systems using multi-echo spin-echo MRI protocols. The influence of T1 on the echo amplitudes via stimulated echo components was studied. The resulting effects on T2 estimates from the Carr-Purcell (CP), Carr-Purcell-Meiboom-Gill (CPMG), and Phase-Alternating-Phase-Shift (PHAPS; combination of CP and CPMG), multiple echo schemes were examined. Protocols with either spatially selective or nonselective refocusing pulses were studied. An intravoxel static field inhomogeneity of 0.1, 1, and 10 ppm was stimulated. The dependence on T1 of the T2 estimates was notable for T1 values below approximately 800 msec for all protocols. The PHAPS scheme provided rather accurate, but underestimated, T2 values when selective refocusing was used. With nonselective refocusing, PHAPS T2 values were overestimated and demonstrated a pronounced dependence on magnetic field inhomogeneity. In general, long T2 values were erroneous with the PHAPS protocol. The results indicate that a CPMG protocol structure provides a more robust method for T2 estimations than the PHAPS protocol.

Properties of the phase-alternating phase-shift (PHAPS) multiple spin-echo protocol in MRI: a study of the effects of imperfect RF pulses.

The effects of imperfect radiofrequency (RF) pulses on the echo amplitudes from the Carr-Purcell (CP), Carr-Purcell-Meiboom-Gill (CPMG), and the PHase-Alternating Phase-Shift (PHAPS; combination of CP and CPMG) multiple spin-echo schemes were studied. Properties of the PHAPS scheme for transverse relaxation time measurements was emphasized. Numerical simulations on non-relaxing spin systems were performed to assess the properties of selective (damped sinc shaped) and nonselective refocusing pulses in terms of effective spatial selectivity and generation of secondary echo signal. Analytical solutions of the Bloch equations were applied to study the generation and propagation of stimulated echo signal caused by nonideal 180 degrees phase reversals, and the results were used to analyse the numerical simulations in terms of primary and stimulated echo components. Finally, the simulated echo train patterns from the different MSE schemes were compared with MR imaging measurements. It was found that the underestimation of T2 values by the PHAPS protocol with selective refocusing pulses is mainly an effect of an "artificial" echo amplitude decay in the CP scheme, while the CPMG scheme produces a typical even-odd echo pattern (different from corresponding echo patterns in conventional high resolution NMR). Both effects are related to the flip angle error and phase dispersion along the slice selection direction from selective RF pulses, and are not significantly influenced by stimulated echo interference for nonrelaxing spin systems. However, the presence of stimulated echoes at the time of the primary echoes implies a dependence on T1 of the PHAPS echo amplitudes. In the CPMG protocol, different gradient schemes have been implemented to defocus stimulated echoes. However, the results indicate that there exists stimulated components that will not be affected by such gradients, and that the optimization of the RF refocusing pulses then remain the main objective.

MRI measurements of the dependence on T1 of the echo amplitudes using a multiple spin-echo scheme.

Analytical calculations using the Bloch formalism were performed to assess the dependence on T1 of the echo amplitudes for the Phase-Alternating Phase-Shift (PHAPS) multiple spin-echo protocol. Measurements in a 0.5 T MR imaging unit were performed to ratify the analytical results. Especially for low T2 values, the echo amplitudes were erroneous, with an increasing contribution from stimulated echo components with increasing T1. Apart from affecting T2 estimates, stimulated echoes generated a non-monoexponential signal decay of the echo trains. The results confirmed previous simulation studies as regards the dependence on T1 of T2 estimates from PHAPS.

The relationship between crown size and ring width in Pinus sylvestris L. stems: dependence on indole-3-acetic acid, carbohydrates and nitrogen in the cambial region.

Indole-3-acetic acid (IAA), carbohydrates, total nitrogen and amino acids in the cambial region and bark were measured at the top (10-year-old internode) and bottom (1.3 m) of the main stem of 50-year-old Pinus sylvestris L. trees, having different rates and longitudinal gradients of annual ring width formation. The trees were sampled during the most active period of wood production (June 23, July 15) and at the end of this period (August 23). Trees with a small crown and relatively slow growth rate (S-trees) were compared with trees in the same stand that had a large crown and fast growth rate (F-trees) as a result of thinning and fertilization. The effect of bottom pruning fast-growing trees (pruned F-trees) was also investigated. The F-trees had greater wood production than the S-trees at both the stem top and bottom. The difference was larger at the stem bottom, thus the relative decrease in ring width down the stem was steeper in the S-trees. The amount of sucrose and IAA per cm(2) in cambial region tissues was higher in F-trees than in S-trees. However, the differences in annual ring width between treatments and within trees were not reflected in the concentrations (expressed per gram fresh weight) of these substances, measured either in differentiating xylem, the cambium plus phloem, or in the cambial region as a whole. The concentrations of total nitrogen and amino acids were slightly higher in the F-trees than in the S-trees. Pruning the F-trees reduced wood production, particularly at the stem bottom, inhibited the springtime increase in starch, and decreased the amounts of sucrose and IAA per cm(2) in the cambial region. However, it was evident that the concentrations of sucrose, IAA, amino acid and nitrogen in the cambial region were not related to ring width. During June and July, the concentrations of these substances were generally higher at the stem bottom in the pruned F-trees than in the F-trees. The results provide evidence that it is the activity of the cambium rather than the availability of carbohydrates that determines the allocation of wood production along the tree stem. The results also indicate that tracheid production is not directly related to the IAA concentration in the cambial region.

Doppler-derived pressure differences in normally functioning aortic valve prostheses. Studies in Björk-Shiley monostrut and Biocor porcine prostheses.

To determine the normal range of maximum and mean Doppler-derived pressure differences for mechanical and bioprosthetic valves in the aortic position, Doppler echocardiography was performed on 239 stable patients with normally functioning Björk-Shiley monostrut (BSM, n = 185) or Biocor porcine (n = 54) prostheses. The interval from aortic valve replacement to echocardiography was 3-9 days. Maximum and mean pressure differences were significantly greater in 21 mm than in 25 or 27 mm BSM prostheses. The pressure differences in 23 mm BSM valves did not diverge significantly from those in 21, 25 or 27 mm valves. The mean pressure difference did not exceed 30 mm Hg in any type or size of studied prosthesis. No significant differences were found in pressure gradients in comparisons between BSM and Biocor prostheses of corresponding sizes. The calculated velocity ratio for BSM prostheses was not significantly influenced by the valve size. We suggest that the normal range of Doppler-derived maximum and mean pressure differences determined in this study be adopted as reference in evaluations of aortic BSM and Biocor valve prostheses.

A comparison of two immediate-early genes, c-fos and NGFI-B, as markers for functional activation in stress-related neuroendocrine circuitry.

The promoter regions of the rat corticotropin-releasing factor (CRF), oxytocin (OT), and vasopressin (AVP) genes contain sequences similar to the cis-acting response element identified for NGFI-B, an immediate-early gene structurally related to the steroid hormone receptor superfamily. Combined immuno- and hybridization histochemical approaches were used to determine whether challenges that influence the synthesis and secretion of CRF, OT, and/or AVP result in altered expression in neurosecretory neurons of NGFI-B and another immediate-early gene, c-fos, which is widely used as a marker for functionally activated neurons. NGFI-B mRNA was found to be expressed at constitutively high levels in the telencephalon, but not in the endocrine hypothalamus, of unperturbed controls; basal levels of c-fos expression were uniformly low throughout the CNS. NGFI-B and c-fos mRNAs, and Fos protein, were induced with a similar time course and in similar neuroendocrine cell types in response to acute hypotensive hemorrhage (15% reduction in blood volume), intravenous injection of interleukin-1 beta (IL-1 beta; 1.87 micrograms/kg), chronic salt loading (7 d maintenance on 2% saline), and acute bilateral adrenalectomy. c-fos mRNA and Fos protein were readily demonstrable in afferent pathways that have been implicated as mediating the neuroendocrine responses in the three stress paradigms; these include medullary catecholaminergic cell groups in response to IL-1 beta and hemorrhage, and cell groups lining the lamina terminalis in response to salt loading. Challenge-specific induction of NGFI-B expression was detectable in these extrahypothalamic cell groups, though with a lesser sensitivity than that required to reveal NGFI-B induction in the hypothalamus, or c-fos expression in these related afferents. These results establish NGFI-B as a useful adjunct to c-fos, for revealing synaptic and/or transcriptional activation in the magno- and parvocellular neurosecretory systems. Differences in the sensitivity of the two markers in revealing functionally related activation in extrahypothalamic regions speak to general issues concerning the use of immediate-early genes in mapping functional circuitry in the CNS.
