The effects of quipazine on 5-HT metabolism in the rat brain.

Since quipazine is a potent 5-HT agonist in peripheral organs, its possible stimulatory effects on serotoninergic receptors in the rat brain were investigated. Quipazine administration (10 mg/kg, i.p.) induced a significant decrease in the synthesis and turnover rates of serotonin in the brain stem as well as in the forebrain. It is not likely that these changes were mediated by a negative feed-back mechanism triggered by a direct action of quipazine on central 5-HT postsynaptic receptors. Indeed, in contrast to LSD and 5-methoxy-N,N-dimethyltryptamine, this compound failed to activate the 5-HT sensitive adenylate cyclase in colliculi homogenates of newborn rats. However, quipazine exerted direct effects on serotoninergic terminals. It inhibited competitively the reuptake process in synaptosomes (Ki=1.38 X 10(-7) M) and stimulated the K+ evoked release of newly synthesized 3H-5-HT in slices of the brain stem. Injected in vivo in a dose which affected 5-HT uptake and release, quipazine did not modify MAO activity. However, this activity was non-competitively inhibited by high concentratin of the drug in vitro (Ki=3.0 X 10(-5) M). These actions are very likely indirectly responsible for the stimulation of central 5-HT receptors.

Effect of suckling on plasma prolactin and hypothalamic monoamine levels in the rat.

In lactating rats that were separated from their pups for 8 hours, suckling induced a rapid rise in plasma prolactin levels. Correlatively, it induced a depletion in dopamine (DA) and serotonin (5-HT) Concentrations within the hypothalamus as early as 5 min after the onset of suckling. This depletion lasted as long as the stimulus was maintained. A parallel increase in the level of the metabolite of 5-HT, 5-hydroxyindolacetic acid (5-HIAA), was observed. Under these conditons, the ratio of 5-HIAA/5-HT is augmented. This ratio is a good index of turnover of the transmitter. Suckling affected neither the content of noradrenaline (NA) in the hypothalamus nor the levels of any amine tested in the cerebral cortex. When pups were separated from their mother for 24 hours, suckling no longer resulted in changes in either plasma prolactin levels or hypothalamic DA, 5-HT, or 5-HIAA concentrations. We have concluded that the activation of a discrete system of 5-HT containing neurons is associated with the triggering effect of suckling on prolactin release.

[Muscular work and its hyperemic phase in the patient with arteritis].

Already users of a method of functional assessment of the muscular circulation in the patients with peripheral artery disease by Xenon 133, the authors propose a radical modification of the performance of the exercise test. A quantified exercise test leads to a new and more objective interpretation of the radio-active tracer elimination curve. This new method permits the measurement of the total quantity of blood which passed during the hyperhaemic stage and its time distribution. The aim of this test is to form an opinion on the tenacity and the efficiency of the supplementary mechanisms when there is an occlusion of the arterial axis.
