[Toxicologic evaluation of sucrose carbonic acid esters in basic subchronic feeding studies in rats. 2. Effect of sucroacetoglycerides].

Groups of male and female rats received a sucroacetoglycerides containing product (SAG) for 3 months at dietary levels of 0, 2.5, 5 and 10%. Food consumption was initially increased in females at all SAG-levels. After two weeks significant increases in food intake were observed in males and females fed 10% SAG throughout the feeding period. The serum analysis revealed significantly elevated activity in serum alkaline phosphatase at the highest SAG-level in males and females after 6 weeks and in females after 13 weeks. Histological changes related to the SAG-feeding were noted in the intestinal lymph nodes of male and female animals fed 10% SAG. The no-adverse effect level established in this subchronic feeding study was 5% SAG in the diet of rats, equivalent to a daily intake of 3.6 g/kg body weight in males and 4.0 g/kg in females.

Separate cloning and expression analysis of two protein components of 4-chlorobenzoate dehalogenase from Pseudomonas sp. C8S3.

The two protein components, II and III, of the 4-chlorobenzoate dehalogenase from Pseudomonas sp. CBS3 were cloned separately into Escherichia coli. Component II was obtained on plasmid pCBSII, containing a 3.0 kbp HindIII fragment, and component III on plasmid pCBSIIIb, containing a 1.3 kbp SalI/PstI fragment. The identities of the two components were confirmed by comparison with the authentic components from Pseudomonas sp. CBS3. Both components were expressed constitutively in E. coli. Neither component alone showed dehalogenating activity. Only in the mixture of crude extracts from both clones was 4-chlorobenzoate dehalogenase detectable. The specific activities in E. coli crude extracts were 2.9 mU (mg protein)-1 for component II and 3.5 mU (mg protein)-1 for component III. Expression analysis by minicell experiments revealed a single polypeptide chain of 29 kDa for component III and of 31 kDa for component III.

Resolution of 4-chlorobenzoate dehalogenase from Pseudomonas sp. strain CBS3 into three components.

Extracts of Pseudomonas sp. strain CBS3 grown with 4-chlorobenzoate as sole carbon source contained an enzyme that converted 4-chlorobenzoate to 4-hydroxybenzoate. This enzyme was shown to consist of three components, all necessary for the reaction. Component I, which had a molecular weight of about 3,000, was highly unstable. Components II and III were stable proteins with molecular weights of about 86,000 and 92,000.

[The metabolism of various acaloric compounds with fatlike properties in rats].

Pseudofats as total substituted long-chain esters and ethers of polyhydroxyl compounds (sucrose, triglycerol, and polyethyleneglycol) are more or less indifferent against gastrointestinal enzymes, but display specific metabolic effects on lipid metabolism especially on cholesterol synthesis in liver, plasma lipid level, and fecal cholesterol excretion. Among the different tested compounds there exist similarities as well as differences, which are to consider as an indication for a specific relationship between structure and effect. These relations are more closely investigated and critically discussed.

[Non-caloric compounds with fat-like functional properties (pseudofats)].

The review deals with the synthesis, characterization, physicochemical and physiological properties with respect to nutrition of acaloric fat-like compounds (pseudofats) with special regard to fatty acid esters of sucrose, glycerol ethers and glycerol dicarbonic acid esters. Furthermore an outlook is given of other classes of compounds that are potentially suited to substitute fat as for instance polycarbonic acid-, polyglycerol-, and polyglycol derivatives.

The inverse electro-oculogram.

An electro-oculogram ratio of less than one was found to be caused by limited ocular excursions under photopic conditions in a patient with a rod-cone dystrophy. Because this inverse electro-oculogram ratio was not caused by a decrease in standing potential under photopic conditions, it should be considered an artefact.
