JANA: a new iterative polyexponential curve stripping program.

A new iterative polyexponential curve stripping technique for the provision of initial pharmacokinetic parameter estimates has been developed. Such estimates are required for nonlinear least-squares curve fitting. In contrast to conventional curve stripping, this new technique does not make any assumption about the relative magnitudes of the exponential rate constants. Hence, the parameter estimates which it provides are free of the bias which may arise in conventional curve stripping. A BASIC program called JANA has been developed to implement the new curve stripping procedure.

The design of pharmacokinetic experiments for model discrimination.

The need for objective design of pharmacokinetic experiments aimed at model discrimination is argued. A sequential design strategy based on information theory is outlined. The characteristics of this strategy and its applicability to pharmacokinetic experiments is examined by means of computer simulated experiments. The limitation of the technique are discussed and alternative approaches outlined.

High-resolution real-time scanning such as described in this article remains limited to the very superficial areas of the body. This article has been designed to give the reader an up-to-date glimpse of the minute world of small parts scanning and aid the clinician in the optimal use of this noninvasive modality.
