Influence of previous photoperiodic exposure on the reproductive response to a specific photoperiod signal in ewes.

Two experiments were carried out to determine whether the reproductive response of ewes to a specific photoperiodic signal depends on the time of year that the signal is given, and, if so, whether this dependence can be attributed to the photoperiodic history of the ewes. The aim of experiment 1 was to expand upon previous findings that the reproductive response to a specific photoperiodic challenge in ewes previously maintained on natural photoperiod varies with time of year. Ewes were transferred at one of three times of year from natural photoperiod to photochambers and were immediately exposed to 35 long days (18L:6D) followed by continuous exposure to short days (8.5L: 15.5D); this treatment is referred to as LD-->SD. The three times of year when long days started corresponded to the beginning of the breeding season, the mid-breeding season, and early anestrus (September 21, December 21, March 21, respectively). In ewes exposed to LD-->SD beginning in September, the breeding season and subsequent anestrous season was not altered. In ewes exposed to LD-->SD beginning in December, the transition to anestrus was advanced (p < 0.05) relative to that in controls maintained in simulated natural photoperiod. Subsequently, half of these ewes resumed reproductive activity within 180 days; this occurred 131 +/- 8 days after transfer to short days. In contrast, all ewes exposed to LD-->SD beginning in March resumed reproductive activity; this began 100 +/- 3 days after transfer to short days (p < 0.05 versus December group). The purpose of experiment 2 was to assess the extent to which the difference in response to a photoperiodic challenge can be attributed to photoperiodic history. Ewes were maintained on short days from the winter solstice interrupted with 35 long days from March 21, June 21, September 21, or December 21. The majority of ewes exhibited an onset of reproductive activity after exposure to LD-->SD at the different times of year, and there was no group difference in latency to onset of reproductive activity. The duration of reproductive activity, however, was longer (p < 0.05) in ewes exposed to LD-->SD beginning in June than in the other groups. Thus we conclude that the seasonal difference in the ability of the photoperiodic challenge of long followed by short days to induce reproductive activity in ewes previously maintained outdoors can be attributed, in large measure, to photoperiodic history. Other factors, such as phase of the endogenous rhythm, however, may influence the duration of reproductive activity resulting from this photoperiodic challenge.

Variation in the ability of a long day followed by a short day photoperiod signal to initiate reproductive activity in ewes at different times of the year.

Three experiments examined the importance of the time and duration of exposure to a long day followed by a short day photoperiod signal in initiating reproductive activity in ewes. In Expt 1, ewes were maintained on short days (8.5 h light:15.5 h dark) from 21 December interrupted with either 105 long days (18 h light:6 h dark; LD) from 9 February or 35 LD from 9 February, 16 March or 20 April. Exposure to long days followed by short days advanced the onset of reproductive activity in comparison to control ewes maintained on simulated natural photoperiod. Exposure to long days for 105 days delayed the onset of reproductive activity (August 2 +/- 3 days; P < 0.05) compared with 35 days beginning on the same date (July 13 +/- 5 days). The interval from the end of the long day signal to the onset of reproductive activity was shorter (P < 0.001) however, after 105 LD than after 35 LD. In Expt 2, control ewes were moved from natural photoperiod to simulated natural photoperiod on 1 November and subsequently exposed to short days from 21 December. Four other groups were also exposed to this basic photoperiodic signal sequence but it was interrupted with either 70 LD from 16 November, or 35 LD from 16 November, 21 December or 20 April. More ewes (P < 0.05) initiated reproductive activity after exposure to 70 LD from 16 November and 35 LD from 21 December or 20 April compared with control ewes maintained on short days or ewes given 35 LD from 16 November. The interval from the end of long days to the onset of reproductive activity was less (P < 0.01) in ewes given 70 LD than in ewes given 35 LD. In Expt 3, ewes on natural photoperiod were given either 90 LD from 21 September, 35 LD from 21 September, 26 October, 30 November, 4 January or 8 February followed by short days. The majority of ewes that received long followed by short days after the winter solstice resumed reproductive activity. However, all photoperiod signals given between the autumn equinox and the winter solstice failed to initiate reproductive activity in ewes during the experiment. Thus we conclude that, in ewes, the reproductive neuroendocrine axis is insensitive to long days followed by short days between the autumn equinox and the winter solstice. The reproductive axis of ewes regains sensitivity to the inductive effects of long days followed by short days at a time close to the winter solstice. Between the winter and summer solstices, long days followed by short days maintain the anoestrous state and provide the cue for initiation of reproductive activity.

Effect of the presence of male and female flockmates on reproductive activity in ewes.

Four experiments were carried out to determine the effect of the presence of ewes and rams on the reproductive state of ewes. In Expt 1, the breeding season of ewes kept with a vasectomized ram ended later (April 18 +/- 8 days; mean +/- SEM) than that of ewes isolated from rams (6 March +/- 7 days; P < 0.01). In Expt 2, the end of the breeding season was later (5 May +/- 6 days; P < 0.05) and the onset of the next breeding season earlier (29 September +/- 2 days; P < 0.001) in ewes maintained with rams, compared with ewes isolated from rams (14 April +/- 7 days and 1 November +/- 2 days, respectively). There was no difference in the timing of, or variation in, reproductive transitions between ewes maintained either as individuals or in groups. In Expt 3, all ewes exposed to artificial short days from the date of the winter solstice and interrupted with 35 long days in spring resumed cyclicity (median date, 7 September; range, 59 days). Most ewes (seven of nine) exposed to short days from the date of the winter solstice and isolated from other ewes did not resume cyclicity in the following 11 months. In contrast, all ewes resumed cyclicity (median date, 19 October; range, 144 days) when exposed to short days but housed in social contact with other ewes that became reproductively active in early September; however, the onset of cyclicity was later than in ewes exposed to long days (P < 0.01).(ABSTRACT TRUNCATED AT 250 WORDS)

Expression of plectin mutant cDNA in cultured cells indicates a role of COOH-terminal domain in intermediate filament association.

Plectin is an intermediate filament (IF) binding protein of exceptionally large size. Its molecular structure, revealed by EM and predicted by its sequence, indicates an NH2-terminal globular domain, a long rodlike central domain, and a globular COOH-terminal domain containing six highly homologous repeat regions. To examine the role of the various domains in mediating plectin's interaction with IFs, we have constructed rat cDNAs encoding truncated plectin mutants under the control of the SV-40 promoter. Mutant proteins expressed in mammalian COS and PtK2 cells could be distinguished from endogenous wild type plectin by virtue of a short carboxy-terminal antigenic peptide (P tag). As shown by conventional and confocal immunofluorescence microscopy, the transient expression of plectin mutants containing all six or the last four of the repeat regions of the COOH-terminus, or the COOH-terminus and the rod, associated with IF networks of both the vimentin and the cytokeratin type and eventually caused their collapse into perinuclear aggregates. Similar effects were observed upon expression of a protein encoded by a full length cDNA construct. Microtubules and microfilaments were unaffected. Unexpectedly, mutants containing the rod without any of the COOH-terminal repeats, accumulated almost exclusively within the nuclei of cells. When the rod was extended by the first one and a half of the COOH-terminal repeats, mutant proteins showed a partial cytoplasmic distribution, although association with intermediate filaments was not observed. Nuclear and diffuse cytoplasmic distribution was also observed upon expression of the NH2-terminal domain without rod. These results indicate that sequences located roughly within the last two thirds of the globular COOH-terminus are indispensable for association of plectin with intermediate filaments in living cells.

Treatment with desmopressin acetate to reduce blood loss after cardiac surgery. A double-blind randomized trial.

Bleeding after cardiopulmonary bypass remains a cause for concern, requiring reexploration of the chest in approximately 3 percent of patients who have had operations on the heart. We examined the possibility that this problem might be alleviated by desmopressin acetate (DDAVP), which increases the plasma level of von Willebrand factor and improves hemostasis in mild hemophilia and other conditions associated with defective platelet function. In a double-blind, prospective, randomized trial, we studied the effect of intraoperative desmopressin acetate in 70 patients undergoing various cardiac operations requiring cardiopulmonary bypass. Patients undergoing uncomplicated primary coronary-artery bypass grafting were not included. The drug significantly reduced mean operative and early postoperative blood loss (1317 +/- 486 ml in the treated group vs. 2210 +/- 1415 ml in the placebo group); of the 14 patients whose 24-hour blood loss exceeded 2000 ml, 11 had received the placebo. Plasma levels of von Willebrand factor were higher after desmopressin acetate than after placebo. Patients with the most bleeding had relatively low levels of von Willebrand factor before operation, suggesting a role for this factor in the hemorrhagic tendency induced by extracorporeal circulation. There were no untoward side effects of desmopressin acetate. We conclude that the administration of desmopressin acetate can be recommended to reduce blood loss in patients undergoing complex cardiac operations. The beneficial effect of the drug on hemostasis after cardiopulmonary bypass may be related to its effect on von Willebrand factor.

Employment status and psychological well-being: a longitudinal study of 16-year-old school leavers.

The relationship between employment status and psychological well-being is investigated in a longitudinal study of 16-year-old school leavers. Significant differences between employed, unemployed and government training scheme groups on a number of measures of well-being were found, these differences persisting after adjustment for individual differences and for other factors known to affect mental health. Implications for clinical practice and public health are considered and possible explanations for the poorer psychological health of the unemployed group are discussed.

Do government training schemes for unemployed school leavers achieve their objectives? A psychological perspective.

The increasing rates of youth unemployment have given rise to concern in academic and political circles as well as amongst the general public. So much so that successive governments have introduced employment schemes specifically for this age group. Although a variety of opinions have been expressed concerning the value of such schemes and much thought and planning has gone into the design of successive generations of schemes, there appears to be little hard evidence concerning the effects they have on the well-being of young people. In the course of a study of the wider aspects of unemployment amongst school leavers, the authors collected evidence which they believe bears directly on this question. In a prospective study, which began by assessing fifth formers before they left school, it was found that in general those who went on to YOP schemes occupied an intermediate position between those who obtained work and those who remained unemployed, on a variety of measures of psychological well-being. However, it was found that those on Schemes were less likely than those in either of the other two groups to believe that they had a measure of personal control over their lives. The findings are discussed in relation to the design of employment schemes and the longer term implications.
