[Organ cultures of mammalian testes. I. Adult mouse testis].

Organ cultures of adult normal mice testes have shown that the tubular structure is best preserved at 31 degrees C. With the simple anhormonal conditions we have used, the germ cells actively involved in meiosis disappear in ten days, following a stereotyped pattern. During the same time, Sertoli cells become enlarged, with a marked cytoplasmic vacuolisation. After 2 weeks in vitro, the seminiferous tubules are completely filled up with Sertoli cells and no more spermatocytes are recognized.

[Cellular receptors of gonadotropic hormones. Immunocytochemical evidence in rat testicle].

We have performed topical applications of HCG and LH on fixed paraffin-embedded sections of adult rat testes. Thereafter, immuno-cytochemical demonstration of the peptides was carried on and demonstrated that HCG and LH specifically bound to the cell membrane of Leydig cells. This technique is highly reproductible provided that adequate fixation (with formalin containing fixatives) as been performed.

[Fate of Leydig cells in testicular organ culture].

In organ culture experiments, the Leydig cells of mouse testes can still be recognized after 12 days at 31 degrees C. HCG binding sites are effectively demonstrated. Meanwhile, when HCG is added at the initiation of the cultures, it is selectively stored in the cytoplasm of the Leydig cells. These findings point to a fairly good survival of Leydig cells in our tissue culture system.

[Effect of pGlu-Tyr-Arg-Trp-NH2 on the serum level of gonadotropins in the male rat].

The tetrapeptide synthesized by Folkers et al. was injected intraperitonealy in normal adult male rats. The release of FSH is significant 90 and 120 minutes after injections of 150, 100, 250 microgram of this substance. An increase of LH was only observed at 120 minutes after injection of 100 microgram of the tetrapeptide. Thus, in rats dissociation between FSH and LH release can be observed.

[Effect of an ovine pineal gland extract on prolactin release in vitro].

Defatted ovine pineal powder is submitted to acetic acid extraction followed by chromatographic separation. The extracted material and one of the two separated fractions inhibit the secretion of PRL by pituitary cells in monolayer culture. Both basal and TRH stimulated secretions are reduced.

Clinical use of LH-RH test as a diagnostic tool.

The response to LH-RH is never characteristic of a given disorder of the hypothalamo-hypophysial-gonadal axis except in the cases of severe gonadal disturbances, but reflects the functional states which can be observed under various circumstances. The same functional state may be found in different diseases and, in contrast, one disease can evolve through different functional states with time.
