A controlled field trial of the effectiveness of cholera and cholera El Tor vaccines in the Philippines.

A controlled field trial on some 584 000 people in an endemic cholera El Tor area in the Philippines demonstrated that cholera vaccines gave moderate protection of short duration. Injection of a single dose of vaccine prepared from either Vibrio cholerae or El Tor vibrios gave over 50% protection for the first 2 months. The immunity conferred by the V. cholerae vaccine declined rapidly after 3 to 4 months. The effectiveness of the El Tor vaccine continued for 6 months. An oil-adjuvant vaccine prepared from V. cholerae conferred an equally high degree of protection for a longer period of time, but, owing to severe vaccination reactions, its use could not be recommended.

Simultaneous sequential determination of compounds of toxicological interest in mixtures by derivative spectroscopy.

An absorption spectroscopic method was developed for the sequential determination of mixture components. The proposed method is straightforward and convenient and provides optimal results that increase the potential of spectrophotometric techniques as applied to date. The method was applied to the sequential resolution of mixtures of pure components of toxicological interest yielding close UV absorption bands by using derivative spectroscopy.

Morphine determination by gas chromatography/mass spectroscopy in human vitreous humor and comparison with radioimmunoassay.

A gas chromatographic-mass spectroscopic method (GC/MS) for morphine determination in human vitreous humor was developed. Reversed-phase solid-liquid extraction gave better sample purification compared with liquid-liquid or normal-phase solid-liquid extractions. In 10 real cases of death by heroin overdose, the correlation between the results obtained by GC/MS and by radioimmunoassay (RIA) was studied.

HPLC determination of cocaine and benzoylecgonine in plasma and urine from drug abusers.

Reversed-phase high-performance liquid chromatography (HPLC) was used for the determination of cocaine and its metabolite, benzoylecgonine (BZE), in plasma and urine. Following a solid-liquid extraction with Bond-Elut Certify cartridges and using methaqualone as reference compound, chromatography was performed with a Lichrospher RP 18 (125 x 4-mm i.d.) reversed-phase column, ultraviolet (UV) detection at 235 nm, and methanol (pH 7)-phosphate buffer (70:30 v/v) as mobile phase. Linearity was obtained from 0.1 to 20 micrograms/mL with a good correlation coefficient. Recoveries ranged from 76.9 to 96.5%, and coefficients of variation were always less than 5.0%. Urine samples from live, cocaine-intoxicated patients contained concentrations of 0.21-75.55 micrograms/mL BZE and less than 4.14 micrograms/mL cocaine, whereas postmortem urine analyses gave concentrations of 5.86-198.76 micrograms/mL for BZE and 1.56-33.24 micrograms/mL for its parent compound. Finally, postmortem blood samples were all negative for cocaine and gave BZE concentrations of 0.040-0.53 micrograms/mL.
