Observation of the D1(2420)-->Dpi + pi- decays.

We report on the first observation of D0/1(2420)-->D0pi- pi+ and D+/1(2420-->D+ pi- pi+ decays (where the contribution from the dominant known D1-->D*pi decay mode is excluded) in the B- -->D0/1pi-) and (-)B0-->D+/1pi- decays, respectively. The observation is based on 15.2 x 10(7) B(-)B events collected with the Belle detector at the KEKB collider. We also set 90% confidence level upper limits for the branching fractions of the four following decays: B- -->D0/1pi-, D01-->D(*0)pi- pi+, (-)B0-->D+/1pi-, D+/1-->D(*+) pi- pi+, B- -->D(*0)2(2460)pi-, D(*0)2 -->D(*0) pi- pi+, (-)B0-->D(*+)2(2460)pi-, D(*+)2-->D(*+)pi- pi+.

Measurement of polarization and triple-product correlations in B-->phiK* decays.

We present measurements of decay amplitudes and triple-product correlations in B-->phiK* decays based on 253 fb(-1) of data recorded at the Upsilon(4S) resonance with the Belle detector at the KEKB e(+)e(-) storage ring. The decay amplitudes for the three different helicity states are determined from the angular distributions of final-state particles. The longitudinal polarization amplitudes are found to be 0.45 +/- 0.05 +/- 0.02 for B0-->phiK(*0) and 0.52 +/- 0.08 +/- 0.03 for B+ -->phiK(*+) decays. CP- and T-odd CP-violating triple-product asymmetries are measured to be consistent with zero.

Observation of B0-->pi0pi0.

We report the observation of the decay B0-->pi(0)pi(0), using a 253 fb(-1) data sample collected at the Upsilon(4S) resonance with the Belle detector at the KEKB e(+)e(-) collider. The measured branching fraction is B(B0-->pi(0)pi(0))=(2.3(+0.4+0.2)(-0.5-0.3))x10(-6), with a significance of 5.8 standard deviations including systematic uncertainties. We also make a measurement of the direct CP violating asymmetry in this mode.

Observation of a near-threshold omegaJ/psi mass enhancement in exclusive B-->KomegaJ/psi decays.

We report the observation of a near-threshold enhancement in the omegaJ/psi invariant mass distribution for exclusive B-->KomegaJ/psi decays. The results are obtained from a 253 fb(-1) data sample that contains 275 x 10(6) BB pairs that were collected near the Upsilon(4S) resonance with the Belle detector at the KEKB asymmetric energy e(+)e(-) collider. The statistical significance of the omegaJ/psi mass enhancement is estimated to be greater than 8sigma.

An open label study of the effects of rituximab in neuromyelitis optica.

Eight patients with worsening neuromyelitis optica were treated with rituximab to achieve B cell depletion. Treatment was well tolerated. Six of eight patients were relapse free and median attack rate declined from 2.6 attacks/patient/year to 0 attacks/patient/year (p = 0.0078). Seven of eight patients experienced substantial recovery of neurologic function over 1 year of average follow-up. The pretreatment median Expanded Disability Status Scale score was 7.5, and at follow-up examination was 5.5 (p = 0.013).

Observation of an isotriplet of excited charmed baryons decaying to lambda+c pi.

We report the observation of an isotriplet of excited charmed baryons, decaying into Lambda(+)(c)pi(-), Lambda(+)(c)pi(0), and Lambda(+)(c)pi(+). We measure the mass differences M(Lambda(+)(c)pi)-M(Lambda(+)(c)) and widths to be 515.4(+3.2+2.1)(-3.1-6.0) MeV/c(2), 61(+18+22)(-13-13) MeV for the neutral state; 505.4(+5.8+12.4)(-4.6-2.0) MeV/c(2), 62(+37+52)(-23-38) MeV for the charged state; and 514.5(+3.4+2.8)(-3.1-4.9) MeV/c(2), 75(+18+12)(-13-11) MeV for the doubly charged state, where the uncertainties are statistical and systematic, respectively. These results are obtained from a 281 fb(-1) data sample collected with the Belle detector near the Upsilon(4S) resonance, at the KEKB asymmetric energy e(+)e(-) collider.

Observation of B+-->K1(1270)+gamma.

We report the observation of the radiative decay B+-->K1(1270)(+) gamma using a data sample of 140 fb(-1) taken at the Upsilon(4S) resonance with the Belle detector at the KEKB e+e- collider. We find the branching fraction to be B(B+-->K1(1270)(+)gamma)=(4.3+/-0.9(stat.)+/-0.9(syst.))x10(-5) with a significance of 7.3sigma. We find no significant signal for B+-->K1(1400)(+)gamma and set an upper limit B(B+-->K1(1400)(+)gamma)<1.5 x 10(-5) at the 90% confidence level. We also measure inclusive branching fractions for B+-->K+pi+pi-gamma and B0-->K0pi+pi-gamma in the mass range 1 GeV/c(2)<M(K+(0)pi+pi-)<2 GeV/c(2).

Study of the suppressed decays B- -->[K+pi-](D)K- and B- -->[K+pi-]Dpi-.

We report a study of the suppressed decays B--->[K(+)pi(-)](D)K- and B--->[K(+)pi(-)](D)pi(-), where [K(+)pi(-)](D) indicates that the K+pi(-) pair originates from a neutral D meson. These decay modes are sensitive to the unitarity triangle angle varphi(3). We use a data sample containing 275 x 10(6) BB pairs recorded at the Upsilon(4S) resonance with the Belle detector at the KEKB asymmetric e(+)e(-) storage ring. The signal for B--->[K(+)pi(-)](D)K- is not statistically significant, and we set a limit r(B)<0.27 at 90% confidence level, where r(B) is the magnitude of the ratio of amplitudes |A(B--->D 0K-)/A(B--->D0K-)|. We observe a signal with 6.4sigma statistical significance in the related mode, B--->[K(+)pi(-)](D)pi(-).

Search for D0-D0 mixing in D0 --> K+ pi(-) decays and measurement of the doubly-Cabibbo-suppressed decay rate.

We have searched for mixing in the D(0)-D (0) system by measuring the decay-time distribution of D(0) --> K(+) pi(-) decays. The analysis uses 90 fb(-1) of data collected by the Belle detector at the KEKB e(+) e(-) collider. We fit the decay-time distribution for the mixing parameters x' and y' and also for the parameter R(D), which is the ratio of the rate for the doubly-Cabibbo-suppressed decay D(0)--> K+ pi(-) to that for the Cabibbo-favored decay D(0)--> K-pi(+). We do these fits both assuming CP conservation and allowing for CP violation. We use a frequentist method to obtain a 95% C.L. region in the x'(2) - y' plane. Assuming no mixing, we measure R(D) = (0.381 +/- 0.017(+0.008)(-0.016))%.

Observation of B0-->D*sJ(2317)+K- decay.

The decays B0-->D+sJK- and B0-->D-sJpi+ are studied for the first time. A significant signal is observed in the B0-->D*sJ(2317)+K- decay channel with B(B0-->D*sJ(2317)+K-) x B(D*sJ(2317)+-->D+spi0)=(5.3(+1.5)(-1.3)+/-0.7+/-1.4) x 10(-5). No signals are observed in the B0-->D*sJ(2317)-pi+, B0-->DsJ(2460)+K-, and B 0-->DsJ(2460)-pi+ decay modes, and upper limits are obtained. The analysis is based on a data set of 140 fb(-1) collected by the Belle experiment at the asymmetric e+e- collider KEKB.

Histaminergic modulation of excitatory synaptic transmission in the rat basolateral amygdala.

The effects of histamine on excitatory synaptic transmission between the external capsule and basolateral amygdala (BLA) were examined using intracellular and field potential recordings in rat amygdala slices. Bath application of histamine (20 microM) suppressed intracellular excitatory postsynaptic potentials (EPSPs; 70.3+/-5.1% of control amplitude) in 43 of 64 BLA neurons, and potentiated EPSPs (341+/-81% of control amplitude) in 21 neurons, without changing resting membrane potential or input resistance. The histamine-induced suppression of EPSPs was accompanied by an increase in paired-pulse facilitation of the slopes of EPSPs, suggesting a presynaptic locus of the action. The suppressive effect could be blocked by the selective H3 antagonist thioperamide, and mimicked by the selective H3 agonist R-alpha-methylhistamine, indicating that the suppressive effect is mediated by the presynaptic H3 receptor. The potentiating effect of histamine on EPSPs was not accompanied by the change of paired-pulse facilitation and was not affected by the presence of H1, H2 or H3 receptor antagonists. In addition, the effective concentration of agonist to produce 50% of maximal response (EC50) of the potentiating action of histamine is 49 nM, much lower than the EC50 (470 nM) of the H3 receptor-mediated suppressive effect characterized here. These observations suggest a novel, high affinity and postsynaptically mediated effect of histamine. In extracellular recordings, histamine, at low concentration (200 nM), consistently potentiated field potentials. At high concentration (20 microM), histamine suppressed field potentials, but potentiated field potentials when H3 receptors were blocked. Taken together, these results revealed that histamine, via the presynaptic H3 receptor and a currently unknown mechanism, decreases or increases excitatory synaptic transmission in the BLA respectively. This specific histaminergic modulation of neuronal activity in the amygdala may play an important role in amygdala-mediated physiological and pathophysiological processes, such as fear, emotional learning and memory, temporal lobe epilepsy, and affective disorders.

Decomposition of carbon dioxide by metals during gamma irradiation.

We have proposed a technique to enhance the decomposition of carbon dioxide by gamma irradiation. This is possible by putting metal components into CO2 gas to promote the conversion of gamma rays to lower-energy electrons through Compton, photoelectron and cascading electron knock-on events in metals. Numerical simulations using the EGS code indicated that the number of lower-energy electrons ejected from metals into CO2 gas increases with increasing Z number and/or density of the metals; this was supported by the experimental results, i.e., the CO2-containing metals with a higher Z number exhibited a greater efficiency for production of CO. In addition, production of CO could be enhanced by carefully controlling the volume and surface area of metal components as well as the proximity to adjacent metal components. These experimental results successfully demonstrated that modification of the kinds of metal components and metal structures can control the energy and the number of electrons ejected from the metals and can lead to enhancement of production of CO from CO2.

Conformational dynamics of estrogen receptors alpha and beta as revealed by intrinsic tryptophan fluorescence and circular dichroism.

Estrogen receptors (ER alpha and ER beta) are ligand-activated nuclear receptors that mediate the action of estrogens. These receptors activate transcription by similar mechanism(s), although the overall amino acid sequence identity is only 47%. In order to compare the structural and conformational features of ER alpha and ER beta, we monitored their intrinsic tryptophan fluorescence during thermal unfolding. The 50% unfolding temperatures (T(M)) of ER alpha and ER beta were 39+/-1 and 40+/-2 degrees C, respectively. Estradiol had no significant effect on the T(M) of ER alpha or ER beta. In contrast, binding of the estrogen-response element increased the T(M) of ER alpha and ER beta by 10 degrees C. Thermal unfolding of estradiol-bound ER alpha and ligand-free ER beta showed two-step transitions, with the formation of intermediates that were stable between 36-48 and 34-42 degrees C, respectively. We confirmed the presence of intermediate states during thermal unfolding by circular dichroism spectroscopy. Atomic force microscopy showed that the ER beta intermediate consisted of discrete globular particles, whereas the ER alpha intermediate showed a speckled appearance, with sparse well-defined particles. Fluorescence-quenching studies showed the presence of two classes of tryptophan in unliganded ER alpha and ER beta. Binding of estradiol to ER beta exposed its tryptophans, whereas estradiol reduced the accessibility of the tryptophans of ER alpha. Our results illustrate the differential effects of ligands on the unfolding of ER alpha and ER beta, and identify partially unfolded intermediates. Differences in the conformational flexibility and stability of ER alpha and ER beta may represent functional differences of ligand-bound ERs in recruiting coactivator proteins and initiating transcription.

Study of time-dependent CP violation in B0-->J/psipi0 decays.

We report a measurement of CP asymmetry parameters in the decay B0(B (0))-->J/psipi(0), which is governed by the b-->cc d transition. The analysis is based on a 140 fb(-1) data sample accumulated at the Upsilon(4S) resonance by the Belle detector at the KEKB asymmetric-energy e(+)e(-) collider. One neutral B meson in the J/psipi(0) final state is fully reconstructed for events used in this analysis. The accompanying B meson flavor is identified by its decay products. From the distribution of proper-time intervals between the two B decays, we obtain the following CP-violating parameters: S(J/psipi(0))=-0.72+/-0.42(stat)+/-0.09(syst) and A(J/psipi(0))=-0.01+/-0.29(stat)+/-0.03(syst).

Search for CP violation in the decay B0-->D*+/-D-/+.

We report a search for CP-violating asymmetry in B0-->D(*+/-)D-/+ decays. The analysis employs two methods of B0 reconstruction: full and partial. In the full reconstruction method all daughter particles of the B0 are required to be detected; the partial reconstruction technique requires a fully reconstructed D- and only a slow pion from the D(*+)-->D0pi(+)(slow) decay. From a fit to the distribution of the time interval corresponding to the distance between two B meson decay points we calculate the CP-violating parameters and find the significance of nonzero CP asymmetry to be 2.7 standard deviations.

Evidence for direct CP violation in B0-->K+pi- decays.

We report evidence for direct CP violation in the decay B0-->K+pi(-) with 253 fb(-1) of data collected with the Belle detector at the KEKB e(+)e(-) collider. Using 275x10(6) BB pairs we observe a B-->K+/-pi(-/+) signal with 2140+/-53 events. The measured CP violating asymmetry is A(CP)(K+pi(-))=-0.101+/-0.025(stat)+/-0.005(syst), corresponding to a significance of 3.9sigma including systematics. We also search for CP violation in the decays B+-->K+pi(0) and B+-->pi(+)pi(0). The measured CP violating asymmetries are A(CP)(K+pi(0))=0.04+/-0.05(stat)+/-0.02(syst) and A(CP)(pi(+)pi(0))=-0.02+/-0.10(stat)+/-0.01(syst), corresponding to the intervals -0.05<A(CP)(K+pi(0))<0.13 and -0.18<A(CP)(pi(+)pi(0))<0.14 at 90% confidence level.

Convectively driven polymerase chain reaction thermal cycler.

We have fabricated a low-cost disposable polymerase chain reaction thermal chamber that uses buoyancy forces to move the sample solution between the different temperatures necessary for amplification. Three-dimensional, unsteady finite element modeling and a simpler 1-D steady-state model are used together with digital particle image velocimetry data to characterize the flow within the device. Biological samples have been amplified using this novel thermal chamber. Time for amplification is less than 30 min. More importantly, an analysis of the energy consumption shows significant improvements over current technology.

Risks of interleukin-1 genetic polymorphisms and Helicobacter pylori infection in the development of gastric cancer.

BACKGROUND: The host genetic factors that determine the clinical outcomes of Helicobacter pylori-infected individuals remain unclear. AIM: To elucidate the risks of host interleukin-1 (IL-1) genetic polymorphisms and H. pylori infection in the development of gastric cancer. METHODS: In a case-control study of 164 controls and 142 patients with gastric cancer, the IL-1B-511 biallelic polymorphisms and the IL-1RN penta-allelic variable number of tandem repeats were genotyped. RESULTS: The carriage of IL-1RN*2, male gender, old age and H. pylori infection independently increased the risk of gastric cancer, with odds ratios of 3.3 [95% confidence interval (CI), 1.4-7.7], 2.1 (95% CI, 1.2-3.8), 5.3 (95% CI, 3.1-9.0) and 2.2 (95% CI, 1.3-3.8), respectively. H. pylori-infected individuals who were carriers of IL-1RN*2 showed increased risks of both intestinal and diffuse types of gastric cancer, with odds ratios of 11.0 and 8.7, respectively. In addition, these individuals also had a higher score of intestinal metaplasia in the corpus than did uninfected non-carriers. CONCLUSIONS: This study is the first to verify IL-1RN*2 as an independent factor governing the development of gastric cancer in Asian individuals. A combination of H. pylori testing and host genotyping may target the eradication of H. pylori to high-risk individuals.
